This study presents the first year-long observations (January 2021-June 2022) of 17 non-methane hydrocarbons (NMHCs) at a high-altitude rural site in the Himalayas (Munsyari: 2200 m a.m.s.l.), a tourist destination influenced by nearby emission sources. Total NMHCs exhibited seasonal variability, with the lowest levels in winter and summer- monsoon (159.5-174.5 ppbC) and the highest levels in spring and autumn (174.5-197.8 ppbC). Aromatic hydrocarbons dominated the NMHC composition throughout the year, contributing about 85-89% of the total NMHCs. This seasonal variability suggests the combine

Analysis of volatile organic compounds (VOC) within exhaled breath is subject to numerous sources of methodological and physiological variability. Whilst breathing pattern is expected to influence the concentrations of selected exhaled VOCs, it remains challenging to investigate respiratory rate and depth accurately in awake subjects. Online breath sampling was performed in 20 mechanically ventilated patients using proton transfer reaction time-of-flight mass spectrometry (PTR-ToF-MS). The effect of variation in respiratory rate (RR) and tidal volume (TV) on the VOC release profiles was examin

The study reports an integrated environmental quality monitoring of a 100 km2 area in central Italy mostly occupied by an underground station of methane storage, working since 1982. The nitrogen oxides, ozone and isoprene concentration detached with a network monitoring of passive filters were compared with the results of lichens biomonitoring. Data from the two monitorings were in accordance: there was an inversely correlation between lichen biodiversity index (IBL) and NOx (-0.96) and ozone (-0.80), and a positive correlation between IBL and isoprene (0.67). IBL indicated that the area range

Photosynthesis for the generation of fuels and chemicals from cyanobacteria and microalgae offers the promise of a single host organism acting both as photocatalyst and processor, performing sunlight absorption and utilization, as well as CO(2) assimilation and conversion into product. However, there is a need to develop methods for generating, sequestering, and trapping such bio-products in an efficient and cost-effective manner that is suitable for industrial scale-up and exploitation. A sealed gaseous/aqueous two-phase photobioreactor was designed and applied for the photosynthetic generati

A new method is described for the collection and assay of volatile compounds in the breath. Subjects expired into a pump-assisted collecting apparatus in which the breath was drawn through a water trap and then through an adsorptive trap where the volatile compounds were captured on graphitized carbon and molecular sieve. The sample was subsequently eluted from the trap by thermal desorption, concentrated by two-stage cryofocusing, then assayed by gas chromatography with flame ionization and flame photometric detection. Several compounds were regularly observed in the breath of normal human vo

While proton-transfer-reaction time-of-flight mass spectrometry (PTR-ToF-MS) is widely used for ambient volatile organic compounds (VOCs) quantification, its accuracy is limited by isomeric speciation and ionization byproducts. Here, gas chromatography (GC) coupled with PTR-ToF-MS was deployed at a suburban site in Hong Kong to resolve isomers and quantify interferences for ambient VOCs measurement. We identified 48 compounds using GC-PTR and resolved their isomer profiles in direct-PTR data. Our analysis revealed that direct-PTR measurements substantially underestimated long-chain aldehydes (

Interactions between anthropogenic and biogenic emissions play a critical role in shaping atmospheric chemistry, yet their combined effects remain insufficiently understood. To elucidate these interactions, we selected toluene and isoprene as representative anthropogenic and biogenic volatile organic compound (AVOC and BVOC), respectively. A series of photooxidation experiments were conducted under both low and high NOx regimes to investigate how isoprene influences the photooxidation of toluene and the formation of reaction products. In toluene-only experiments, the fractions of toluene consu

The emission of isoprenoids (e.g. isoprene and monoterpenes) by plants plays an important defensive role against biotic and abiotic stresses. Little is known, however, about the functional traits linked to species-specific variability in the types and rates of isoprenoids emitted and about possible co-evolution of functional traits with isoprenoid emission type (isoprene emitter, monoterpene emitter or both). We combined data for isoprene and monoterpene emission rates per unit dry mass with key functional traits (foliar nitrogen (N) and phosphorus (P) concentrations, and leaf mass per area) a

Isobutanol is a promising next-generation biofuel with a high energy density and is considered a sustainable aviation fuel precursor. Klebsiella pneumoniae is an industrial workhorse microorganism used for the production of various chemicals. An endogenous isobutanol synthesis pathway, interconnected with the branched-chain amino acid and 2,3-butanediol biosynthesis pathways, has been identified in K. pneumoniae. However, isobutanol production by K. pneumoniae is inefficient due to the substantial formation of by-products. In this study, we enhanced isobutanol synthesis by systematically engin

The Lyme disease pathogen Borrelia burgdorferi contains a highly reduced genome lacking many primary metabolic pathways. However, B. burgdorferi retains the mevalonate pathway that synthesizes isopentenyl pyrophosphate (IPP), the precursor to the peptidoglycan carrier lipid. While the mevalonate pathway and the enzyme that catalyzes its rate-limiting step (3-hydroxy-3-methyl glutaryl coenzyme A reductase, HMGR) are well studied in vertebrates, little is known about the pathway in B. burgdorferi and many pathogenic bacteria. In this work, we reveal that HMGR is a critical metabolic enzyme in B.

Intracellular redox homeostasis in the airway epithelium is closely regulated through adaptive signaling and metabolic pathways. However, inhalational exposure to xenobiotic stressors such as secondary organic aerosols (SOA) can alter intracellular redox homeostasis. Isoprene hydroxy hydroperoxide (ISOPOOH), a ubiquitous volatile organic compound derived from the atmospheric photooxidation of biogenic isoprene, is a major contributor to SOA. We have previously demonstrated that exposure of human airway epithelial cells (HAEC) to ISOPOOH induces oxidative stress through multiple mechanisms incl

Mulberry (Morus alba) is a significant plant with numerous economic benefits; however, its growth and development are affected by nutrient levels. A high level of magnesium (Mg) or magnesium nutrient starvation are two of the significant Mg factors affecting plant growth and development. Nevertheless, M. alba's metabolic response to different Mg concentrations is unclear. In this study, different Mg concentrations, optimal (3 mmol/L), high (6 mmol/L and 9 mmol/L), or low (1 and 2 mmol/L) and deficient (0 mmol/L), were applied to M. alba for three weeks to evaluate their effects via physiologic

The primarily emitted compounds by human presence, e.g., skin and volatile organic compounds (VOCs) in breath, can react with typical indoor air oxidants, ozone (O3), and hydroxyl radicals (OH), leading to secondary organic compounds. Nevertheless, our understanding about the formation processes of the compounds through reactions of indoor air oxidants with primary emitted pollutants is still incomplete. In this study we performed real-time measurements of nitrous acid (HONO), nitrogen oxides (NOx = NO + NO2), O3, and VOCs to investigate the contribution of human presence and human activity, e

Over the last decades, post-illumination bursts (PIBs) of isoprene, acetaldehyde and green leaf volatiles (GLVs) following rapid light-to-dark transitions have been reported for a variety of different plant species. However, the mechanisms triggering their release still remain unclear. Here we measured PIBs of isoprene-emitting (IE) and isoprene non-emitting (NE) grey poplar plants grown under different climate scenarios (ambient control and three scenarios with elevated CO2 concentrations: elevated control, periodic heat and temperature stress, chronic heat and temperature stress, followed by

Dimethylallyltryptophan synthase is an aromatic prenyltransferase that catalyzes an electrophilic aromatic substitution reaction between dimethylallyl diphosphate (DMAPP) and L-tryptophan. The synthase is found in a variety of fungi, where it catalyzes the first committed step in the biosynthesis of the ergot alkaloids. The enzymatic reaction could follow either a dissociative mechanism involving a discrete dimethylallyl cation intermediate or an associative mechanism in which the indole ring directly displaces diphosphate in a single step. In this work, positional isotope exchange (PIX) exper

BACKGROUND: While assumed to protect against coronavirus transmission, face masks may have effects on respiratory-haemodynamic parameters. Within this pilot study, we investigated immediate and progressive effects of FFP2 and surgical masks on exhaled breath constituents and physiological attributes in 30 adults at rest.
METHODS: We continuously monitored exhaled breath profiles within mask space in older (age 60-80 years) and young to middle-aged (age 20-59 years) adults over the period of 15 and 30 min by high-resolution real-time mass-spectrometry. Peripheral oxygen saturation (S pO2 ) and

The gut microbiota produces numerous metabolites that influence the epithelial barrier function. Bacterial catabolism of amino acids produces a wide variety of metabolites whose effects on the intestinal epithelium remain to be identified. In this study, we investigated the effects of amino acid-derived metabolites (isovalerate, isobutyrate, 2-methylbutyrate, 5-aminovalerate, cadaverine, putrescine, and tryptamine) in cell monolayers derived from porcine ileum organoids. Our results show that the branched-chain fatty acid (BCFA) isovalerate improved the epithelial barrier function, as assessed

The apicoplast, a peculiar organelle of red algal origin surrounded by four membranes, is important for several metabolic processes in the malaria parasite, including isopentenyl pyrophosphate (IPP) and coenzyme A (CoA) biosynthesis. Transporters are required to provide substrates and export products for these metabolic pathways and are therefore excellent novel drug targets. On the basis of known apicoplast pathways, we discuss which functions are expected to be fulfilled by the Plasmodium apicoplast transportome, which comprises 11 confirmed and 17 putative apicoplast transporters identified

This study explores the role of ammonia (NH3) in the formation of nitrogen-containing secondary organic aerosol (SOA) using smog chamber experiments. Reactive organic gases, including toluene, m-xylene, ethylbenzene, α-pinene, and isoprene, were analyzed under NH3-present and NH3-absent conditions. High resolution mass spectrometry revealed that toluene, m-xylene, ethylbenzene and isoprene exhibited higher signal-to-noise (S/N) ratios and an increased abundance of nitrogen-containing oxygenated compounds in the presence of NH3. Conversely, α-PIN demonstrated high S/N ratios in both conditions,
