This unit presents methods for assaying sialic acids, reducing sugars, and hexosamines. The BCA assay detects free reducing terminii in sugars released from glycoconjugates by appropriate treatments. Assays employing Ehrlich reagent (DMAB) detect hexosamines and N-acetylhexosamines, including a method for hydrolyzing the glycosidic linkages of the hexosamines and a method for re-N-acetylation. The TBA and DMB assays can be used to quantitate and fractionate free forms of many types of sialic acids. Techniques for liberating the sialic acids from the parent glycoconjugates are also provided.

Brain glycogen has long been regarded uniquely as a source of energetic support in situations of emergency or heightened activity. Recently, brain glycogen was found to contain a significant amount of glucosamine, which is used to sustain protein glycosylation. In this update, we highlight that glucosamine synthesis through the hexosamine pathway would subtract glutamine, which is indispensable for glutamate and GABA recycling. Brain glycogen seems then to serve an additional role. By providing glucosamine and, through it, inhibiting the hexosamine pathway, glycogen avoids glutamine depletion.

The molar ratios of N-acetyl neuraminic acid, hexose, hexosamine, and sphingosine have been determined for the abnormal ganglioside in Tay-Sachs disease that was previously detected as a fast-moving band in thin-layer chromatography, and in two abnormal fast-moving bands of gangliosides from the cortex and white matter of the brain in cases of gargoylism and Niemann-Pick's disease. The fastest-moving ganglioside band in these two conditions contains neither hexosamine nor glucose.

Damage of the gastric mucosal barrier by Helicobacter pylori (Hp) has been investigated with estimation of antral mucosal hexosamine and phospholipid, and the degree of pathologic changes to be also evoluted in 100 cases of chronic superficial gastritis. There are 42% of 50 cases of Hp positive patients with active gastric lesions, and significantly high than 50 cases of Hp negative patients (8%). A heteropolysaccharide formed by hexoses, hexosamines, fucose, sialic acid, uronic acids and sulphates was isolated from submandibular glands of rabbits during postnatal development. The presence of sulphates in the submandibular glands of growing animals and their absence in the submandibular glands of adult animals is rather difficult to explain. This biochemical investigation and preliminary results obtained by light and electron microscopy on the sublingual gland seem to suggest a substitutive role of the submandibular gland during the neonatal period.

An increased level of glycoproteins (seromucoid, protein-bound hexoses, protein-bound hexosamines, protein-bound sialic acids) was found in the blood serum from patients with rheumatoid arthritis (RA) as compared with healthy individuals. Higher glycoprotein content was observed in the seropositive RA patients than in the seronegative ones. Higher levels of glycoproteins was also observed in the synovial fluid from seropositive RA patients than in the seronegative ones.
