The Knowns Unknowns: Exploring the Homologous Recombination Repair Pathway in Toxoplasma gondii.

Toxoplasma gondii is an apicomplexan parasite of medical and veterinary importance which causes toxoplasmosis in humans. Great effort is currently being devoted toward the identification of novel drugs capable of targeting such illness. In this context, we believe that the thorough understanding of the life cycle of this model parasite will facilitate the identification of new druggable targets in T. gondii. It is important to exploit the available knowledge of pathways which could modulate the sensitivity of the parasite to DNA damaging agents. The homologous recombination repair (HRR) pathwa

Germline Chromothripsis Driven by L1-Mediated Retrotransposition and Alu/Alu Homologous Recombination.

Chromothripsis (CTH) is a phenomenon where multiple localized double-stranded DNA breaks result in complex genomic rearrangements. Although the DNA-repair mechanisms involved in CTH have been described, the mechanisms driving the localized "shattering" process remain unclear. High-throughput sequence analysis of a familial germline CTH revealed an inserted SVAE retrotransposon associated with a 110-kb deletion displaying hallmarks of L1-mediated retrotransposition. Our analysis suggests that the SVAE insertion did not occur prior to or after, but concurrent with the CTH event. We also observed

Homologous recombination mediates functional recovery of dysferlin deficiency following AAV5 gene transfer.

The dysferlinopathies comprise a group of untreatable muscle disorders including limb girdle muscular dystrophy type 2B, Miyoshi myopathy, distal anterior compartment syndrome, and rigid spine syndrome. As with other forms of muscular dystrophy, adeno-associated virus (AAV) gene transfer is a particularly auspicious treatment strategy, however the size of the DYSF cDNA (6.5 kb) negates packaging into traditional AAV serotypes known to express well in muscle (i.e. rAAV1, 2, 6, 8, 9). Potential advantages of a full cDNA versus a mini-gene include: maintaining structural-functional protein domain

The Fanconi anemia pathway promotes homologous recombination repair in DT40 cell line.

Fanconi anemia (FA) is a rare hereditary disorder characterized by bone marrow failure, compromised genome stability, and increased incidence of cancer. FA is caused by abnormalities that occur in components of the FA core complex, a key factor FancD2, breast cancer susceptibility protein BRCA2/FancD1, or BRIP1/FancJ. These proteins are proposed to function in a common biochemical process (FA pathway), however, its precise role is still unclear. In this chapter, we will summarize our genetic analysis on the FA pathway using DT40 cells line. Our data revealed that (1) FA pathway promotes DNA re

m7G modification in mRNA cooperates with QKI to promote homologous recombination repair.

DNA double strand breaks (DSBs) are repaired through distinct pathways, with homologous recombination (HR) favored when DNA ends undergo extensive resection. Although cytoplasmic N7-methylguanosine (m7G)-modified RNA increases after UV-induced single-strand breaks, its role in DSB repair (including HR) remains unclear. Here, we show that m7G-modified RNA accumulates at DSB sites, where it recruits the m7G reader protein Quaking (QKI)5, to selectively promote HR. Mechanistically, QKI5 directly interacts with MRE11 and enhances its DNA binding affinity, thereby facilitating DNA end resection. De

RAD51 succinylation regulates homologous recombination and contributes to the chemosensitivity in cancer.

Genomic instability and metabolic reprogramming are core hallmarks of cancer, yet how they are mechanistically interconnected remains unclear. Here, we demonstrate that succinyl-coenzyme A (CoA), a tricarboxylic acid (TCA) cycle metabolite and protein succinylation donor, modulates homologous recombination (HR) by regulating RAD51 succinylation. OXCT1 succinylates RAD51 at K285, whereas HDAC11 removes this modification. RAD51 succinylation disrupts BRCA2 interaction, impairs RAD51 foci formation, and suppresses HR. Upon DNA damage, ATM-dependent phosphorylation of HDAC11 enhances the interacti

MND1 reduces breast cancer chemosensitivity by promoting RAD51-mediated homologous recombination repair.

Breast cancer remains a major global health challenge for women. Chemotherapy resistance is a key driver of breast cancer recurrence and metastasis. Although meiotic nuclear division 1 (MND1) has been characterized as an oncogenic factor involved in mitotic progression and homologous recombination (HR), the precise molecular mechanisms underlying these MND1-mediated processes remain unclear. The expression of MND1 in breast cancer cell lines was evaluated by RT-qPCR and western blot. The functional effects of MND1 were assessed through a series of in vitro and in vivo assays, including cell pr

ATM promotes PFKP nuclear translocation to balance glycolysis and homologous recombination repair.

Emerging evidence suggests a critical interplay between glycolysis and DNA damage repair (DDR) during tumor progression. However, the precise molecular mechanisms coordinating these processes remain unclear. This study reveals a previously unrecognized role of the platelet isoform of phosphofructokinase-1 (PFKP) in homologous recombination (HR) repair. Our investigations demonstrated that ataxia-telangiectasia mutated (ATM) kinase phosphorylates PFKP at residue T278, promoting its transition from tetramers to dimers in response to ionizing radiation (IR) and high glucose (HG) stimulation. HELZ-BRCA2 complex resolves R-loops to drive transcription-coupled homologous recombination.

R-loops are transcription-induced, three-stranded nucleic acid structures that, if not properly resolved, can disrupt DNA repair and compromise genome stability. BRCA2, a tumor suppressor vital for homologous recombination (HR), also contributes to R-loop regulation, though the underlying mechanisms remain poorly understood. Here, we identify HELZ as a direct BRCA2 interactor and characterize it as an ssRNA-specific R-loop resolvase. BRCA2 enhances HELZ helicase activity and promotes its recruitment to R-loops. Importantly, HELZ resolves R-loops at DNA double-strand breaks, enabling efficient

dUTPase modulates mycobacterial homologous recombination and interacts with the AdnAB helicase-nuclease.

This study identifies a previously unrecognized interaction between Mycobacterium tuberculosis dUTPase (Dut) and the AdnAB homologous recombination complex. Using a combination of yeast two-hybrid screening, mycobacterial protein fragment complementation, and biochemical analyses with purified proteins, we show that dUTPase physically interacts with the N-terminal region of AdnA and modulates the activity of the AdnAB helicase-nuclease complex. Biochemical assays demonstrate that Dut enhances AdnAB activity on DNA substrates and alters the AdnAB-DNA interaction. Mutational perturbation of Dut,

Homologous recombination deficiency in endometrial cancer: shedding light on recent clinical findings.

Endometrial cancer is the most common gynecologic malignancy in the United States, with rising incidence and high recurrence rates. Immune checkpoint inhibitors (ICIs) benefit patients with mismatch repair-deficient (dMMR) tumors, but options remain limited for those with mismatch repair-proficient (pMMR) disease. Homologous recombination deficiency (HRD), a genomic instability phenotype, has emerged as a therapeutic target. Poly(adenosine diphosphate-ribose) polymerase inhibitors (PARPis) are being investigated in endometrial cancer, with studies exploring whether HRD predicts response, parti

WDFY2 promotes MRN complex formation required for homologous recombination-mediated DNA repair.

The MRE11-RAD50-NBS1 (MRN) complex is fundamental for detecting and repairing DNA double-strand breaks (DSBs), thereby safeguarding genome integrity. However, the precise mechanism governing MRN complex recruitment to DSBs remains largely unexplored. Here, we identify WD40- and FYVE domain-containing protein 2 (WDFY2) as an important regulator of MRN complex formation at DNA damage sites, facilitating homologous recombination (HR) repair. Mechanistically, WDFY2 is phosphorylated at serine 84 by the ATM-CHK2 axis, priming it for recruitment to DSBs. Through direct interactions with MRE11 and NB

Dynamic immunoediting by macrophages in homologous recombination deficiency-stratified pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease, notably resistant to existing therapies. Current research indicates that PDAC patients deficient in homologous recombination (HR) benefit from platinum-based treatments and poly-ADP-ribose polymerase inhibitors (PARPi). However, the effectiveness of PARPi in HR-deficient (HRD) PDAC is suboptimal, and significant challenges remain in fully understanding the distinct characteristics and implications of HRD-associated PDAC. We analyzed 16 PDAC patient-derived tissues, categorized by their homologous recombination deficiency (HRD) scores

[TRIP13 Enhances Radioresistance of Lung Adenocarcinoma Cells
through the Homologous Recombination Pathway].

BACKGROUND: Radiation therapy is one of the most common treatments for non-small cell lung cancer (NSCLC). However, the insensitivity of some tumor cells to radiation is one of the major reasons for the poor efficacy of radiotherapy and the poor prognosis of patients, and exploring the underlying mechanisms behind radioresistance is the key to solving this clinical challenge. This study aimed to identify the molecules associated with radioresistance in lung adenocarcinoma (LUAD), identified thyroid hormone receptor interactor 13 (TRIP13) as the main target initially, and explored whether TRIP1

The BRCA1/BARD1 complex recognizes pre-ribosomal RNA to facilitate homologous recombination.

The BRCA1/BARD1 complex plays a key role in the repair of DNA double-strand breaks (DSBs) in both somatic cells and germ cells. However, the underlying molecular mechanism by which this complex mediates DSB repair is not fully understood. Here, we examined the XY body of male germ cells, where DSBs are accumulated. We show that the recruitment of the BRCA1/BARD1 complex to the unsynapsed axis of the XY body is mediated by pre-ribosomal RNA (pre-rRNA). Similarly, the BRCA1/BARD1 complex associates with pre-rRNA in somatic cells, which not only forms nuclear foci in response to DSBs, but also ta

Neuroendocrine Neoplasms Associated with Germline Pathogenic Variants in the Homologous Recombination Pathway.

Neuroendocrine neoplasms (NENs) have been primarily associated with germline pathogenic variants in genes involved in chromatin remodeling (MEN1), cell cycle control (CDKN1B), PI3K/mTOR signaling (TSC1/2, PTEN) as well as pseudohypoxia (VHL, SDHx). Recent work has implicated various genes involved in DNA repair pathways in the pathophysiology of a subset of pancreatic neuroendocrine neoplasms, including BRCA2, via the homologous recombination pathway (HRD). To date, germline variants in other HRD pathway genes have not been described to contribute to NEN. PALB2, RAD51C, and BARD1 are additiona

Dynamics and impact of homologous recombination on the evolution of Legionella pneumophila.

Legionella pneumophila is an environmental bacterium and the causative agent of Legionnaires' disease. Previous genomic studies have shown that recombination accounts for a high proportion (>96%) of diversity within several major disease-associated sequence types (STs) of L. pneumophila. This suggests that recombination represents a potentially important force shaping adaptation and virulence. Despite this, little is known about the biological effects of recombination in L. pneumophila, particularly with regards to homologous recombination (whereby genes are replaced with alternative allelic v

Roles for APRIN (PDS5B) in homologous recombination and in ovarian cancer prediction.

APRIN (PDS5 cohesin associated factor B) interacts with both the cohesin complex and the BRCA2 tumor suppressor. How APRIN influences cohesion and DNA repair processes is not well understood. Here, we show that APRIN is recruited to DNA damage sites. We find that APRIN interacts directly with RAD51, PALB2 and BRCA2. APRIN stimulates RAD51-mediated DNA strand invasion. APRIN also binds DNA with an affinity for D-loop structures and single-strand (ss) DNA. APRIN is a new homologous recombination (HR) mediator as it counteracts the RPA inhibitory effect on RAD51 loading to ssDNA. We show that APR

Interactions between BRCA2 and RAD51 for promoting homologous recombination in Leishmania infantum.

In most organisms, the primary function of homologous recombination (HR) is to allow genome protection by the faithful repair of DNA double-strand breaks. The vital step of HR is the search for sequence homology, mediated by the RAD51 recombinase, which is stimulated further by proteins mediators such as the tumor suppressor BRCA2. The biochemical interplay between RAD51 and BRCA2 is unknown in Leishmania or Trypanosoma. Here we show that the Leishmania infantum BRCA2 protein possesses several critical features important for the regulation of DNA recombination at the genetic and biochemical le
