Hypochlorous acid triggered fluorescent probes for in situ imaging of a psoriasis model.

Psoriasis is a common skin disease with complex pathogenesis that lacks diagnostic methods. Typically, psoriasis is an inflammation-related disease accompanied by high expression of reactive oxygen species (ROS) in the infected part. However, due to the lack of suitable tools, it is difficult to identify the ROS, especially certain types of ROS (e.g., HOCl) in the psoriasis model. Here, two HOCl-specific fluorescent probes, G1 and G2, were designed and synthesized based on oxazine 1. Both probes could react with HOCl with high selectivity among other ROS under physiological conditions. The sel

Hypochlorous acid is antipruritic and anti-inflammatory in a mouse model of atopic dermatitis.

BACKGROUND: It has been reported that topical hypochlorous acid (HOCl) formulations lead to relief of itch in human patients with atopic dermatitis; however, the specific antipruritic mechanism of action remains unclear. OBJECTIVE: To confirm itch relief and reduction of lesions in a mouse model of atopic dermatitis and to elucidate possible HOCl's mode of action. METHODS: In this study, the effects of topical administration of HOCl hydrogel (0.05%) on atopic dermatitis-like lesions in NC/Nga mice model as well as in vitro effects of HOCl on dorsal root ganglia neurons and mouse bone marrow-…

What are the plasma targets of the oxidant hypochlorous acid? A kinetic modeling approach.

Myeloperoxidase (MPO) is a heme enzyme, released by activated leukocytes at sites of inflammation, which catalyzes the formation of the potent oxidant, hypochlorous acid (HOCl), from H2O2. HOCl is a key component of the inflammatory response and is bactericidal but has been linked with several human pathologies as a result of damage to host tissue. Elevated plasma MPO levels are a strong independent risk factor, and predictor of outcomes, for cardiovascular disease. Rate constants for reaction of HOCl with individual biological targets and the products of these reactions have been determined, …

Reactions of hypochlorous acid with biological substrates are activated catalytically by tertiary amines.

The activation of reactions of HOCl with a variety of model substrates by tertiary amines was investigated spectroscopically by tandem-mix and stopped-flow techniques. HOCl-induced chlorination of salicylate can be sped up by several orders of magnitude by catalytic amounts of trimethylamine (TMN). The effect is obviously due to the fast generation of reactive quarternary chloramonium ions, TMN+ Cl, which act as chain carrier in a catalytic reaction cycle. Of various catalysts tested, quinine shows the highest activity; this is attributable to the quinuclidine (QN) substituent, a bicyclic tert

HYPOChLorous Acid TEsting Studies in percutaneous DRAINAGE (HYPOCLATES:DRAINAGE): protocol for a single-centre, controlled, ambispective cohort study comparing hypochlorous acid versus saline irrigation in percutaneous drainage of intra-abdominal abscesses.

INTRODUCTION: Percutaneous drainage is the standard of care for intra-abdominal abscesses. Additional saline irrigation provides mechanical cleansing but lacks antimicrobial activity. Hypochlorous acid (HOCl), an endogenous antiseptic with broad-spectrum microbicidal activity and high tissue compatibility, is a promising alternative for intracavitary infection control. This study evaluates whether HOCl irrigation shortens the indwelling time of percutaneous irrigation drains and systemic antibiotic duration compared with saline irrigation in patients with intra-abdominal abscesses. METHODS AN…

Visualization of endogenous hypochlorous acid during ferroptosis based on Rhodamine B fluorescent probe.

Hypochlorous acid (HClO) plays critical roles in ferroptosis process and the specific molecular mechanism related to it is still obscure. To reveal HClO fluctuation in the ferroptosis cells, a fluorescence turn-on probe RDM-577 that can detect intracellular HClO was skillfully engineered. The probe exhibited a substantial fluorescence enhancement in response to HClO (F/F0 = 35), enabling sensitive detection of HClO in both in vitro and in vivo environments. Additionally, it demonstrated remarkable performance characteristics, such as an exceptionally low detection limit (5.8 nM). Consequently,…

Detecting mitochondrial hypochlorous acid and viscosity in atherosclerosis models via NIR fluorescent probes.

The assessment of early atherosclerosis (AS) via fluorescence imaging is crucial for advancing early diagnosis research. Abnormal inflammation biomarkers, including hypochlorous acid (HClO) and viscosity within mitochondria, have been closely linked to the pathogenesis of AS. However, current fluorescent probes predominantly rely on unimodal imaging that targets a single biomarker and lacks mitochondrial specificity, which can result in potential false signal readouts due to the complex intracellular environment. Herein, we report the development of three fluorescence probes (M-1, M-2, and M-3…

Neutrophil-Derived Myeloperoxidase and Hypochlorous Acid Critically Contribute to 20-Hydroxyeicosatetraenoic Acid Increases that Drive Postischemic Angiogenesis.

Compensatory angiogenesis is an important adaptation for recovery from critical ischemia. We recently identified 20-hydroxyeicosatetraenoic acid (20-HETE) as a novel contributor of ischemia-induced angiogenesis. However, the precise mechanisms by which ischemia promotes 20-HETE increases that drive angiogenesis are unknown. This study aims to address the hypothesis that inflammatory neutrophil-derived myeloperoxidase (MPO) and hypochlorous acid (HOCl) critically contribute to 20-HETE increases leading to ischemic angiogenesis. Using Liquid Chromatography-Mass Spectrometry/Mass Spectrometry, La…

Fluorescence-based detection of hypochlorous acid using catalytic hairpin assembly and G-quadruplex.

Hypochlorous acid (HClO), a key reactive oxygen species (ROS), plays a crucial role in maintaining innate immunity and physiological homeostasis. Abnormal HClO levels can disrupt biological equilibrium, thereby contributing to the onset and progression of diseases such as ulcerative colitis and atherosclerosis. Herein, we developed a highly sensitive and specific quantitative analytical method for the accurate detection of HClO. This approach enhances the fluorescence of the system through G-quadruplex-ThT binding and further amplifies the signals by inducing catalytic hairpin assembly. Under …

Mycobacterium smegmatis Resists the Bactericidal Activity of Hypochlorous Acid Produced in Neutrophil Phagosomes.

Neutrophils are often the major leukocyte at sites of mycobacterial infection, yet little is known about their ability to kill mycobacteria. In this study we have investigated whether the potent antibacterial oxidant hypochlorous acid (HOCl) contributes to killing of Mycobacterium smegmatis when this bacterium is phagocytosed by human neutrophils. We found that M. smegmatis were ingested by neutrophils into intracellular phagosomes but were killed slowly. We measured a t 1/2 of 30 min for the survival of M. smegmatis inside neutrophils, which is 5 times longer than that reported for Staphyloco…

Selective and absolute quantification of endogenous hypochlorous acid with quantum-dot conjugated microbeads.

Endogenous hypochlorous acid (HOCl) secreted by leukocytes plays a critical role in both the immune defense of mammalians and the pathogenesis of various diseases intimately related to inflammation. We report the first selective and absolute quantification of endogenous HOCl produced by leukocytes in vitro and in vivo with a novel quantum dot-based sensor. An activated human neutrophil secreted 6.5 ± 0.9 × 10(8) HOCl molecules into its phagosome, and kinetic measurement for the secretions showed that the extracellular generation of HOCl was temporally retarded, but the quantity eventually atta…

Hypochlorous acid alters bronchial epithelial cell membrane properties and prevention by extracellular glutathione.

In chronic inflammatory diseases of the airways, such as cystic fibrosis, hypochlorous acid (HOCl) generated by neutrophils is involved in airway injury. We examined the effects of HOCl on 16HBE14o- bronchial epithelial cells by bolus addition or by generation with glucose oxidase plus myeloperoxidase. HOCl produced both carbonyl formation of a discreet number of proteins and modification of surface targets that were recognized by an antibody raised against HOCl-modified protein. Bolus or enzymatically generated HOCl decreased transepithelial resistance, but surprisingly bolus HOCl also increa…

HYPOChLorous Acid TEsting Studies in elective groin VASCULAR surgery (HYPOCLATES:VASCULAR): protocol for an ambispective cohort study evaluating a standard change to hypochlorous acid lavage in patients undergoing elective groin vascular surgery.

BACKGROUND: Surgical site infections remain a frequent and clinically relevant complication following elective open vascular groin surgery, particularly in procedures involving implantation of alloplastic material. Despite routine use in many surgical disciplines, the role of standardized intraoperative wound irrigation in vascular groin surgery is insufficiently defined. Hypochlorous acid is a physiologically occurring antimicrobial agent with broad-spectrum activity and favourable tissue compatibility, making it a promising option for intraoperative wound lavage. However, clinical evidence f…

Development of a near-infrared fluorescent probe for visualizing of hypochlorous acid in vitro and in vivo.

Hypochlorous acid (HClO) played momentous role in various physiological processes and the aberrant expression of HClO was related to kinds of diseases including inflammation and cancer. Therefore, optical tracking of HClO was of significance for exploring its physiological and pathological effects. In this study, we designed and synthesized a new near-infrared (NIR) fluorescent probe (TMNDN) for the detection of HClO. Attributed to the robust electron-withdrawing performance of recognition moiety 2,4-dinitroaniline, the ICT process of fluorophore was interrupted and the fluorescence was switch…

The Fibrosis and Immunological Features of Hypochlorous Acid Induced Mouse Model of Systemic Sclerosis.

Fibrotic animal models are critical for the pathogenesis investigations and drug explorations in systemic sclerosis (SSc). The bleomycin (BLM)-induced mouse model is the classical and most widely used fibrosis model. However, traditional subcutaneous injection of BLM rarely induced diffuse skin and lung lesions. Hypochlorous acid (HOCl)-induced mice are a more representative model that have diffuse cutaneous lesions, lung fibrosis and renal involvement. However, the fibrotic and immunological features of this model are not fully elucidated. Here, we injected BALB/c mice subcutaneously with HOC…
