Magnesium deficiency modulates hepatic lipogenesis and apolipoprotein gene expression in the rat.

The present study was performed to determine the effects of magnesium (Mg) deficiency upon plasma lipoproteins and hepatic apolipoprotein ge … RNA editing. These changes in apolipoprotein gene expression were associated with increased hepatic lipogenesis, despite the observation that net triacylglycerol secretion rates were not different between Mg-deficient and control animals. Taken together, the data demonstrate a complex pattern of alterations in hepatic lipid metabolism and apolipoprotein gene …

Targeting host lipogenesis with a diarylamide inhibitor disrupts SARS-CoV-2 replication.

The vulnerability of direct-acting antivirals to resistance has motivated interest in host-directed strategies that target conserved cellular pathways essential for viral replication. Here, we report a diarylamide molecule, compound 10 (compd. 10), which inhibits SARS-CoV-2 more potently than the parent clofoctol. Mechanistic studies revealed that it is associated with suppression of host lipogenesis, evidenced by downregulation of fatty acid synthase (FASN) and stearoyl-CoA desaturase 1 (SCD1), and partial reversal of antiviral activity upon fatty acid supplementation. Compd. 10 was found to …

Upregulated FASN-mediated lipogenesis in senescent macrophages contributes to liver fibrosis progression.

BACKGROUND: Liver fibrosis represents a common advanced pathological stage of various chronic liver diseases. Macrophages serve as key regulators of innate immunity and play important roles in the development of liver fibrosis. Cellular senescence is an irreversible cell cycle arrest state. However, the mechanisms underlying the role of senescent macrophages in liver fibrosis remain incompletely understood. METHODS: Liver fibrosis was induced in young (8-week) and middle-aged (12-month) mice by intraperitoneal injection of 25% carbon tetrachloride (CCl4). Hepatic function and the immune micro

Blue Light Deactivation of Catalase Suppresses Candida Hyphae Development Through Lipogenesis Inhibition.

Hyphae formation is a key step for fungal penetration into epithelial cells and escaping from macrophages or neutrophils. We found that 405 nm light-induced catalase deactivation results in the inhibition of hyphae growth in Candida albicans. The treatment is capable of inhibiting hyphae growth across multiple hyphae-producing Candida species. Metabolic studies on light-treated C. albicans reveal that light treatment results in a strong reduction in both lipid and protein metabolism. A significant decrease in unsaturated and saturated fatty acids was detected through mass spectroscopy, indicat

Acute intermittent hypoxia drives hepatic de novo lipogenesis in humans and rodents.

BACKGROUND AND AIMS: Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver condition. It is tightly associated with an adverse metabolic phenotype (including obesity and type 2 diabetes) as well as with obstructive sleep apnoea (OSA) of which intermittent hypoxia is a critical component. Hepatic de novo lipogenesis (DNL) is a significant contributor to hepatic lipid content and the pathogenesis of NAFLD and has been proposed as a key pathway to target in the development of pharmacotherapies to treat NAFLD. Our aim is to use experimental models to investigate the impact of …

TTC39B destabilizes retinoblastoma protein promoting hepatic lipogenesis in a sex-specific fashion.

BACKGROUND & AIMS: Molecular mechanisms underlying the different susceptibility of men and women to non-alcoholic fatty liver disease (NAFLD) are poorly understood. The TTC39B locus encodes a scaffolding protein, associates with gynecological disorders and its deletion protects mice from diet-induced steatohepatitis. This study aimed to elucidate the molecular mechanisms linking TTC39B (T39) to the expression of lipogenic genes and to explore sex-specific effects. METHODS: Co-expression in HEK293A cells validated the novel T39/pRb interaction predicted by a protein-protein interaction algorit

Monosaccharide-induced lipogenesis regulates the human hepatic sex hormone-binding globulin gene.

The liver produces plasma sex hormone-binding globulin (SHBG), which transports sex steroids and regulates their access to tissues. In overw … lar palmitate levels and could be mimicked by treatment with palmitoyl-CoA. Moreover, inhibition of lipogenesis prevented monosaccharide-induced downregulation of HNF-4alpha and reduced SHBG expression in HepG2 cells. Thus, monosaccharide-induced lipogenesis reduced hepatic HNF-4alpha levels, which in turn attenuated SHBG expression. This provides a biologica …

Targeting de novo lipogenesis as a novel approach in anti-cancer therapy.

BACKGROUND: Although altered membrane physiology has been discussed within the context of cancer, targeting membrane characteristics by drugs being an attractive therapeutic strategy has received little attention so far. METHODS: Various acetyl-CoA carboxylase 1 (ACC1), and fatty acid synthase (FASN) inhibitors (like Soraphen A and Cerulenin) as well as genetic knockdown approaches were employed to study the effects of disturbed phospholipid composition on membrane properties and its functional impact on cancer progression. By using state-of-the-art methodologies such as LC-MS/MS, optical twe

The effects of vitamin D3 on lipogenesis in the liver and adipose tissue of pregnant rats.

Obesity is a worldwide individual and public health issue, and contributes to the development of numerous chronic diseases. In particular, m … reduces fat in pregnant animals, as well as body weight. Our results suggest that the alteration of lipogenesis through the administration of VD3 may help to reduce excessive weight gain during pregnancy and prevent obesity‑related pregnancy complications such as pre-eclampsia, gestational diabetes, hypertension and issues with labor.

Corticotropin-releasing hormone: an autocrine hormone that promotes lipogenesis in human sebocytes.

Sebaceous glands may be involved in a pathway conceptually similar to that of the hypothalamic-pituitary-adrenal (HPA) axis. Such a pathway has been described and may occur in human skin and lately in the sebaceous glands because they express neuropeptide receptors. Corticotropin-releasing hormone (CRH) is the most proximal element of the HPA axis, and it acts as central coordinator for neuroendocrine and behavioral responses to stress. To further examine the probability of an HPA equivalent pathway, we investigated the expression of CRH, CRH-binding protein (CRH-BP), and CRH receptors (CRH-R)

ZRSR1 protects against obesity-associated hepatic lipogenesis independent of minor intron splicing.

Minor intron retention resulting from dysregulation of minor intron splicing factors is an emerging risk factor for the metabolic dysfunctio … nism. Notably, ZRSR1 cell-autonomously suppressed liver X receptor (LXR) activation-induced de novo lipogenesis in both hepatocyte models and mouse models. Mechanistically, we demonstrate that ZRSR1 directly interacted with LXRα, and this physical interaction suppressed the binding activity of the LXRα/retinoid X receptor α (RXRα) complex toward the promoter …

Sirtuin 6 activator UBCS039 ameliorates hepatic lipogenesis through liver X receptor deacetylation.

Histone deacetylase sirtuin 6 (SIRT6) regulates hepatic lipogenesis by modulating key transcription factors, such as liver X receptor (LXR) and sterol regulatory element binding transcription factor 1 (SREBF1). Despite its potential role in the regulation of hepatic lipogenesis, SIRT6 activators that inhibit hepatic lipogenesis and ameliorate steatotic liver diseases have not been developed yet. We investigated the effects of UBCS039, a selective SIRT6 activator, on lipogenic gene expression and inflammation in hepatocytes. Analysis of publicly available RNA sequencing datasets revealed signif…

Targeting p300 and CBP abolishes HOXB13-loss-induced lipogenesis and tumor metastasis.

HOXB13 is a prostate-specific transcription factor best known for its role as an androgen receptor (AR) cofactor. Recent evidence suggests t … ed cell motility. Importantly, pharmacological inhibition of p300 and CBP blocks HOXB13-loss-driven lipogenesis, reduces MMP expression, and decreases cell migration in vitro and tumor metastasis in vivo. Analysis of clinical samples revealed that HOXB13 expression is reduced in metastatic hormone-sensitive PCa compared with matched primary tumors, further su …

Glycerol-3-phosphate activates ChREBP, FGF21 transcription and lipogenesis in citrin deficiency.

Citrin deficiency (CD) is caused by the inactivation of SLC25A13, a mitochondrial membrane protein required to move electrons from cytosolic NADH to the mitochondrial matrix in hepatocytes. People with CD do not like sweets. Here we show that SLC25A13 loss causes the accumulation of glycerol-3-phosphate (G3P), which activates the carbohydrate response element-binding protein (ChREBP) to transcribe FGF21, which acts in the brain to restrain intake of sweets and alcohol and to transcribe key genes driving lipogenesis. Mouse and human data suggest that G3P-ChREBP is a mechanistic component of the…

There was also increased gene expression in the GRβ-Ad mice for lipogenesis, eicosanoid synthesis, and inflammatory pathways. These indicate that GRβ-induced glucocorticoid resistance may drive hepatic fat accumulation, providing new therapeutic advantages.

CD36 promotes de novo lipogenesis in hepatocytes through INSIG2-dependent SREBP1 processing.

OBJECTIVE: Enhanced de novo lipogenesis (DNL) in hepatocytes is a major contributor to nonalcoholic fatty liver disease (NAFLD). Fatty acid translocase (FAT/CD36) is involved in the pathogenesis of NAFLD through facilitating free fatty acids uptake. Here, we explored the effects of CD36 on DNL and elucidated the underlying mechanisms. METHODS: We generated hepatocyte-specific CD36 knockout (CD36LKO) mice to study in vivo effects of CD36 on DNL under high-fat diet (HFD). Lipid deposition and DNL were analyzed in primary hepatocytes isolated from CD36LKO mice or HepG2 cells with CD36 overexpres…
