An orthotopic mouse model of hepatocellular carcinoma with underlying liver cirrhosis.

Subcutaneous xenografts have been used for decades to study hepatocellular carcinoma (HCC). These models do not reproduce the specific pathophysiological features of HCCs, which occur in cirrhotic livers that show pronounced necroinflammation, abnormal angiogenesis and extensive fibrosis. As these features are crucial for studying the role of the pathologic host microenvironment in tumor initiation, progression and treatment response, alternative HCC models are desirable. Here we describe a syngeneic orthotopic HCC model in immunocompetent mice with liver cirrhosis induced by carbon tetrachlor

Factor from rat liver with antiinvasive potential on rat ascites hepatoma cells.

An acid/ethanol extract of normal rat liver, both in vitro and in vivo, inhibited the invasion by a highly invasive subpopulation of rat ascites hepatoma cells, AH 130 (LC-AH cells). The addition of 10-80 micrograms/ml extract inhibited the formation of penetrated colonies of LC-AH cells underneath the cultured mesothelial cell (M-cell) monolayer. The tumor cells pretreated with the extract showed the diminished colony formation. Preincubation of the extract with plasma membranes prepared from LC-AH cells abolished the effect of the extract, suggesting a binding of the inhibitory entity [tenta

Arsenic and cancer: the still unanswered question.

For some 2,500 years, arsenic was considered an important element of pharmacopeias and was praised for its medicinal qualities. However, an incidental link with cancer was suggested during the nineteenth century, and since that time a controversy has ensued with regard to the compound's tumorigenicity. Several epidemiological surveys, for example, have suggested that arsenic induces lung, liver, or skin cancer in humans. On the other hand, all experimental attempts to reproduce such neoplasms in laboratory animals have consistently failed, thereby denying support to the human data. Recently ar

The Use of Induced Pluripotent Stem Cells for the Study and Treatment of Liver Diseases.

Liver disease is a major global health concern. Liver cirrhosis is one of the leading causes of death in the world and currently the only therapeutic option for end-stage liver disease (e.g., acute liver failure, cirrhosis, chronic hepatitis, cholestatic diseases, metabolic diseases, and malignant neoplasms) is orthotropic liver transplantation. Transplantation of hepatocytes has been proposed and used as an alternative to whole organ transplant to stabilize and prolong the lives of patients in some clinical cases. Although these experimental therapies have demonstrated promising and beneficia

Toxic and carcinogenic effects of dimethylnitrosamine (DMNA) in the blue fox (Alopex lagopus).

Single doses of DMNA from 8 to 15 mg/kg body weight (B.W.) were given in the feed, by stomach tube or by subcutaneous application to 37 foxes. The course and intensity of the disease was not influenced by the application route, but was directly related to the amount of DMNA given per kg body weight, and caused hemorrhagic centrolobular liver necrosis and acute vessel changes especially in the hepatic vein system. The possibility of liver regeneration after a single DMNA exposure depends on the degree of damage in the hepatic vein system. Some animals can recover from the acute disease caused b

Safranal improves the anticancer efficacy of sorafenib via transcriptomic reprogramming and metabolomic changes in a rat model of diethylnitrosamine-induced cirrhosis-hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is a leading cause of cancer-related mortality. Sorafenib (SB) remains an option for advanced HCC in patients who cannot receive, or do not respond to, immunotherapy. Safranal (SF), a bioactive saffron compound, possesses anti-inflammatory and anticancer properties. This study evaluated SF with SB in a diethylnitrosamine-induced cirrhosis-HCC rat model. Rats were assigned to control, HCC, HCC + SB, HCC + SF, and HCC + SF + SB groups and treated for three weeks. Liver morphology, function, histology, apoptosis, cell proliferation, transcriptomics, and metabolomics

Insulin-Like Growth Factor (IGF) System in Liver Diseases.

Hepatocyte differentiation, proliferation, and apoptosis are affected by growth factors produced in liver. Insulin-like growth factor 1 and 2 (IGF1 and IGF2) act in response to growth hormone (GH). Other IGF family components include at least six binding proteins (IGFBP1 to 6), manifested by both IGFs develop due to interaction through the type 1 receptor (IGF1R). The data based on animal models and/or in vitro studies suggest the role of IGF system components in cellular aspects of hepatocarcinogenesis (cell cycle progression, uncontrolled proliferation, cell survival, migration, inhibition o

Neoplastic and nonneoplastic lesions in aging (C57BL/6N x C3H/HeN)F1 (B6C3F1) mice.

Neoplastic and nonneoplastic lesions in untreated (C57BL/6N x C3H/HeN)F1 (B6C3F1) mice used as controls in carcinogenesis tests were tabulated and evaluated. The most common neoplasms in 2,543 male mice were hepatocellular adenomas and carcinomas. In 2,522 female mice, common tumors were lymphomas, leukemias, pulmonary adenomas and carcinomas, hepatocellular adenomas and carcinomas, and pituitary adenomas. The risk of developing most neoplasms increased with the age of the mouse. Hepatocellular carcinomas metastasized in 12% of the animals with these tumors. Other than lymphomas and leukemias,

Tumor regrowth after irradiation; an experimental approach.

Structural changes in irradiated tumors and their regrowth were studied in a rat hepatoma, AH109A, using histological and transparent-chamber techniques. The development of the tumour was examined by means of vascular morphometry as observed in the chamber. Schematically, the tumour tissue was divided into four isocentric layers according to vascular morphology and measurements of vessel volume, surface area, and length per mm3 of tissue. The vascularity was greatest in the outermost region, decreased towards the inner parts and reached an absence of vascularity at the central necrosis. The tu

Diethyl-nitrosamine hepatocarcinogenesis in cirrhotic rats.

The aim of this study was to clarify how the CCl4-induced cirrhosis modifies the process of diethylin-nitrosamine hepatocarcinogenesis. Two strains of rats (CFY and F-344) were used. CFY rats proved to be resistant toward both the cirrhogenic effect of CCl4 and carcinogenic effect of DEN. In the F-344 rats, on the other hand, a large number of foci, neoplastic nodules and hepatocellular carcinomas occurred following DEN treatment only and fullblown liver cirrhosis did develop in the CCl4-treated group. In F-344 rats with cirrhotic liver, foci and neoplastic nodules appeared in the usual number

Procarbazine carcinogenicity in methotrexate-treated or lipotrope-deficient male rats.

Procarbazine hydrochloride (PCZ), a chemotherapeutic agent used extensively to treat Hodgkins disease and other tumors, induces leukemia, lymphoma, mammary gland and other solid tumors in rodents and non-human primates and is strongly implicated as a leukemogen in humans. Lipotrope (choline and methionine) deficiency is a powerful potentiator of chemical carcinogenesis in liver and, under some conditions, in other tissues in rodents. Methotrexate (MTX), another commonly used chemotherapeutic agent, interferes with one-carbon metabolism and limits availability of lipotropes. Studies of PCZ carc

The xanthine oxidase inhibitor oxypurinol reduces cancer cachexia-induced cardiomyopathy.

BACKGROUND: Cachexia is a common complication of cancer and may be responsible for 22% of all cancer-related deaths. The exact cause of death in cancer cachexia patients is unknown. Recently, atrophy of the heart has been described in cancer cachexia animal models, which resulted in impaired cardiac function and is likely to contribute to mortality. In cancer patients hyperuricaemia independent of tumour lysis syndrome is often associated with a worse prognosis. Xanthine oxidase (XO) metabolizes purines to uric acid and its inhibition has been shown to improve clinical outcome in patients with

Carcinogenicity of heptachlor and heptachlor epoxide.

Heptachlor and its metabolite heptachlor epoxide are unequivocally carcinogenic in rats and mice. The chemicals induced carcinomas of the liver, which were highly significant. There were neoplasms at other sites in rats. Neoplasms at all sites, as well as malignant tumors, were increased in heptachlor-treated male rats. There were similar increases in benign and malignant neoplasms of endocrine organs, particularly in female rats. Neoplasms of the thyroid and pituitary were increased in male rats and neoplasms of the reproductive system, including the ovary and uterus, in female rats given hep

Cross-immunity among allogeneic tumors in rats immunized with gamma-irradiated ascites tumors.

Non-inbred rats of the Gifu strain were intraperitoneally challenged with Hirosaki sarcoma (tetraploid type, 10(5) cells) after repeated immunization with gamma-irradiated (13,000 rads 60Co) allogeneic non-viral tumors of ascites type (tetraploid or diploid type of Hirosaki sarcoma, Usubuchi sarcoma or AH130). In rats immunized not only with the same tumor as the immunizing tumor but also with a different tumor, the growth of the challenge tumor was markedly inhibited as compared with the control in non-immunized rats. It is considered that these tumors retained common antigen(s) by the resist

Potential Therapeutic Effects of Chrysin and Apigenin in a Diethylnitrosamine-Induced Hepatocellular Carcinoma Rat Model.

Hepatocellular carcinoma (HCC) is an aggressive cancer with poor prognosis and high mortality rates. The increasing mortality associated with HCC highlights the urgent need for alternative therapeutic approaches. Natural products and their structural analogs have shown promise as novel treatment options. This study aimed to evaluate the potential therapeutic effects of the natural flavonoids apigenin and chrysin, administered individually and in combination, in an experimental HCC model. Forty-eight rats were divided into five groups: Control, DEN, DEN + Apigenin, DEN + Chrysin, and DEN + Comb

The Apoptotic Effect of Trichinella spiralis Infection Against Experimentally Induced Hepatocellular Carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is the sixth most common type of cancer. Prognosis of HCC remains unsatisfactory. Therefore, developing new therapeutic modalities is still mandatory. Tumor biotherapy is a novel concept developed as a therapeutic strategy for cancer treatment. There is a similarity between the regulatory mechanism of Trichinella spiralis nurse cell formation and tumor cell apoptosis signal regulation. OBJECTIVES: Induction of apoptosis by T. spiralis can represent a new strategy for tumor treatment. METHODS: Experimental animals were divided in four groups; negativ
