Cationic hydrogel with sustained lubrication and anti-calcification for osteoarthritis therapy.

BACKGROUND: Osteoarthritis is characterized by cartilage degradation and abnormal subchondral bone remodeling, primarily due to disrupted lubrication-calcification coupling. Current treatments mainly relieve symptoms but lack effective strategies against calcification, and lubrication supplements offer only temporary relief. Here, we present a novel dual-functional cationic hydrogel designed to simultaneously enhance lubrication and inhibit pathological calcification, targeting the dysregulated lubrication-calcification interaction in OA. This approach offers a promising disease-modifying stra…

Confined Liquids Studied by Resonance Shear Measurement: Molecular Mechanism of Lubrication.

Confined liquids present intriguing phenomena not only for foundational research but also for various engineering applications, particularly in tribology. When liquids are confined in a nanospace between solid substrates, they exhibit unique properties different from those of the bulk state due to altered molecular packing and motion restrictions and/or molecular interaction with the substrate surfaces. It has profound implications in the study of lubrication, especially in boundary lubrication where energy efficient low-viscosity lubricants typically lead to high friction and wear. Recent fin…

The Last Dance of Lubrication: Capturing Ultralow Friction before Full Evaporation.

Unsteady friction phenomena driven by lubricant evaporation are pervasive across a wide range of applications, from precision machinery to e … 0 s, while liquid water abruptly raised the COF above 1.7. Dynamic modulation between dry and vapor lubrication enabled rapid COF switching (2-3 to 0.02) within seconds. These findings redefine friction dynamics and provide insights into advanced friction control and system optimization.

Insights into the Multiscale Lubrication Mechanism of Edible Phase Change Materials.

Investigation of a lubrication behavior of phase change materials (PCM) can be challenging in applications involving relative motion, e.g., sport (ice skating), food (chocolates), energy (thermal storage), apparel (textiles with PCM), etc. In oral tribology, a phase change often occurs in a sequence of dynamic interactions between the ingested PCM and oral surfaces from a licking stage to a saliva-mixed stage at contact scales spanning micro- (cellular), meso- (papillae), and macroscales. Often the lubrication performance and correlations across length scales and different stages remain poorly…

Bioinspired Hyaluronic Acid/Phosphorylcholine Polymer with Enhanced Lubrication and Anti-Inflammation.

Under pathological conditions, the joint is not well lubricated, which inevitably leads to osteoarthritis. Currently, in clinics injection of hyaluronic acid (HA) as an intra-articular viscosupplement is one of the main methods for alleviation of osteoarthritis. However, the viscosity of HA reduces dramatically under high shear rate due to the shear-thinning effect. Therefore, it is crucial to enhance the lubrication property of HA in order to treat osteoarthritis effectively. In this study, we successfully grafted 2-methacryloyloxyethyl phosphorylcholine (MPC), which is a zwitterionic biomate…

Aqueous Lubrication, Structure and Rheological Properties of Whey Protein Microgel Particles.

Aqueous lubrication has emerged as an active research area in recent years due to its prevalence in nature in biotribological contacts and its enormous technological soft-matter applications. In this study, we designed aqueous dispersions of biocompatible whey-protein microgel particles (WPM) (10-80 vol %) cross-linked via disulfide bonding and focused on understanding their rheological, structural and biotribological properties (smooth polydimethylsiloxane (PDMS) contacts, Ra < 50 nm, ball-on-disk set up). The WPM particles (Dh = 380 nm) displayed shear-thinning behavior and good lubricating …

An analysis of the squeeze-film lubrication mechanism for articular cartilage.

An asymptotic analysis of a lubrication problem is presented for a model of articular cartilage and synovial fluid under the squeeze-film condition. This model is based upon the following constitutive assumptions: (1) articular cartilage is a linear porous-permeable biphasic material filled with a linearly viscous fluid (i.e. Newtonian fluid); (2) synovial fluid is also a linearly viscous fluid. The geometry of the problem is defined by assuming that (1) cartilage is a uniform layer of thickness H; (2) synovial fluid is a very thin layer compared to H; (3) the radius R of the load-supporting a…

Drop Friction on Textured Lubricant-Coated Surfaces.

Understanding drop friction on textured surfaces has implications in microfluidics and lab-on-a-chip devices. In this work, we investigated the drop friction on lubricant-coated pillars by systematically varying pillar height and density. First, we measured the friction force on a moving drop using a cantilever force sensor that has ±0.1 μN sensitivity. This measurement shows that drop friction on tall dense pillars is comparable to drop friction on short pillars, a significant result that suggests the presence of a Landau-Levich-Derjaguin (LLD) film underneath the moving drop. Second, we vali

The Osmolality of Sex-the Science of Vaginal Moisturizers and Lubricants.

More than half of women report using vaginal lubricants during their lifetime. Despite such ubiquitous use, most healthcare providers do not ask about their use, and they may not feel comfortable counseling a patient about how to choose the best product. The purpose of this review is to explain the difference between various vaginal products, address the factors that affect the efficacy of vaginal products, and provide evidence on the use of these products for masturbation and the use of sex toys, penile-vaginal intercourse, anal intercourse, and for couples seeking to become pregnant.

Allergenic characterization of best-selling personal lubricants.

Personal lubricants vary appreciably in ingredient composition. Although some products are labeled hypoallergenic, these claims are seldom evidence-based. The objective was to determine the potential allergenicity of popular lubricants and elucidate the validity of hypoallergenic product claims. This cross-sectional study involved publicly available data on Amazon's 50 best-selling lubricants as of July 2023. Product ingredient lists were cross-referenced with the North American Contact Dermatitis Group and American Contact Dermatitis Society allergen series. Associations were explored between

Lubricating gel influence on vaginal microbiome sampling.

Gel lubrication is routinely used during gynecological examination to prevent or reduce pain, yet its impact on microbial composition during sampling remains unclear. This study aimed to investigate whether lubricating gel affects the microbial composition of vaginal samples. We included 31 pregnant women presenting during their third trimester to clinics or emergency room and collected 143 unique vaginal samples for 16S amplicon microbial analysis. Vaginal samples were obtained using sterile swabs under various conditions: without gel-immediately frozen (n = 30), with gel-immediately frozen, …

Biomimetic zwitterionic copolymerized chitosan as an articular lubricant.

Restoration of the lubrication functions of articular cartilage is an effective treatment to alleviate the progression of osteoarthritis (OA). Herein, we fabricated chitosan-block-poly(sulfobetaine methacrylate) (CS-b-pSBMA) copolymer via a free radical polymerization of sulfobetaine methacrylate onto activated chitosan segment, structurally mimicking the lubricating biomolecules on cartilage. The successful copolymerization of CS-b-pSBMA was verified by Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, and 1H nuclear magnetic resonance. Friction test confirmed that th…

Nourish and nurture: development of a nutrient ocular lubricant.

PURPOSE: The authors aimed to produce a new tear substitute capable of providing both lubrication and nutrition, based on a novel nutrient-containing therapeutic ocular surface medium (TOSM). METHODS: Viscous substances, including hypromellose (HPMC), carbopol, and sodium hyaluronate (SH) were added to the TOSM at various concentrations. Three commercial preservative-free artificial tear substitutes, Hypromellose (Pharmacy of Moorfields Eye Hospital, London, UK), Thilo-Tears (a carbomer; Alcon Pharma GmbH, Freiburg, Germany), and Vislube (a hyaluronate; Chemedica AG, Munich, Germany) were use…

Anaphylaxis to an urethral lubricant: chlorhexidine as the "hidden" allergen.

We report on a severe allergic reaction during urethral catheterisation, initially erroneously attributed to natural rubber latex (NRL). However, the negative investigations for latex, the uneventful further applications of NRL, the positive skin test and basophil activation test for chlorhexidine strongly support diagnosis of chlorhexidine anaphylaxis. This case illustrates that it is not widely appreciated that "innocent" products such as urethral lubricants may elicit serious life-threatening reactions. Careful inspection of the medical records is once again re-emphasised to be mandatory in

Pain during Female Urethral Catheterization: Intraurethral Lubricant Injection versus Catheter Tip Lubrication--A Prospective Randomized Trial.

PURPOSE: Urethral lubrication during catheterization can be performed by instilling the gel directly in the urethra or by pouring the gel on the catheter tip. In this study we compared the pain level associated with each technique during female urethral catheterization in the setup of a multichannel urodynamic study. MATERIALS AND METHODS: A total of 94 women with a mean ± SD age of 55 ± 14 years who were referred for a multichannel urodynamic study were prospectively randomized into 2 groups according to lubrication technique, including 1) instillation of 5 ml 2% lidocaine gel in the urethra…

Astringency and Lubrication of Polyphenol Systems under Dynamic Salivary Replenishment.

Polyphenol-saliva interactions disrupt mucin-mediated lubrication and induce astringency, yet most models neglect dynamic salivary replenishment. Here, a stepwise artificial saliva (AS) addition model was developed to compare tannic acid (TA) and (-)-epigallocatechin gallate (EGCG), with or without xanthan gum (XG). Four systems (TA-AS, EGCG-AS, TA-XG-AS, EGCG-XG-AS) underwent sequential AS replenishment and were characterized using colloidal, spectroscopic, tribological, and electronic tongue analyses. TA-AS progressively formed larger aggregates and exhibited the highest friction, whereas EG…

Exogenous Collagen Crosslinking is Highly Detrimental to Articular Cartilage Lubrication.

Healthy articular cartilage is a remarkable bearing material optimized for near-frictionless joint articulation. Because its limited self-repair capacity renders it susceptible to osteoarthritis (OA), approaches to reinforce or rebuild degenerative cartilage are of significant interest. While exogenous collagen crosslinking (CXL) treatments improve cartilage's mechanical properties and increase its resistance to enzymatic degradation, their effects on cartilage lubrication remain less clear. Here, we examined how the collagen crosslinking agents genipin (GP) and glutaraldehyde (GTA) impact car…
