Mannose-binding lectin: structure, function, genetics and disease associations.

Mannose-binding lectin (MBL), a serum protein characterised by both collagenous regions and lectin domains, plays an important role in innate immune defence. It binds to the repeating sugar arrays on many microbial surfaces through multiple lectin domains and, following binding, is able to activate the complement system via an associated serum protease, MASP-2. Serum levels of MBL are influenced by three mutations clustered in exon 1 of the gene and are further modulated by various promoter region polymorphisms. The exon 1 mutations lead to secondary structural abnormalities of the collagenous…

Pneumocystis carinii enhances soluble mannose receptor production by macrophages.

Phagocytosis of extracellular organisms in the alveolar spaces of the lungs represents the first-line of host defense against pulmonary pathogens. Disruption of this process is likely to interfere with the generation of appropriate specific immune responses, and lead to a delayed or inefficient clearance of the pathogen. Pneumocystis carinii, an opportunistic pathogen in immunodeficient individuals, is cleared from the lung by alveolar macrophages. In the absence of specific anti-Pneumocystis antibodies, phagocytosis is dependent on the non-opsonic macrophage mannose receptor (MR). Recent stud…

Transfer of mannose from mannosyl retinyl phosphate to protein.

Upon incubation of [14C]mannose-labeled mannosyl retinyl phosphate with a membrane fraction from rat liver, mannose was transferred to an endogenous acceptor precipitable withchloroform/methanol to the extent of about 7%. The reaction proceeded linearly with time for 120 min at a pH optimum of about 7.0. The acceptor thus labeled with mannose could be solubilized by sodium dodecyl sulfate/mercaptoethanol. More than half of this acceptor appeared in the void volume of a Sephadex G-100 column. When it was digested with Pronase, a substantial proportion of it appeared between the void and bed vol…

Purification of a D-mannose isomerase from Mycobacterium smegmatis.

An enzyme, d-mannose ketol isomerase, catalyzing the isomerization of d-mannose and d-fructose was purified approximately 60-fold from cells of Mycobacterium smegmatis grown on mannose as the sole carbon source. This enzyme was shown to catalyze the conversion of d-mannose and d-lyxose to ketoses. The ketose produced from mannose was identified as fructose by chemical and chromatographic methods. The reaction was shown to be reversible, the equilibrium ratio of fructose to mannose being approximately 65 to 35. The pH optimum was about 7.5, and the K(m) for mannose was estimated to be 7 x 10(-3…

D-Glucose and D-mannose-based metabolic probes. Part 3: Synthesis of specifically deuterated D-glucose, D-mannose, and 2-deoxy-D-glucose.

Altered carbohydrate metabolism in cancer cells was first noted by Otto Warburg more than 80 years ago. Upregulation of genes controlling the glycolytic pathway under normoxia, known as the Warburg effect, clearly differentiates malignant from non-malignant cells. The resurgence of interest in cancer metabolism aims at a better understanding of the metabolic differences between malignant and non-malignant cells and the creation of novel therapeutic and diagnostic agents exploiting these differences. Modified d-glucose and d-mannose analogs were shown to interfere with the metabolism of their r…

Measurement of erythrocyte membrane mannoses to assess splenic function.

Red blood cells (RBCs) lose plasma membrane in the spleen as they age, but the cells and molecules involved are yet to be identified. Sickle cell disease and infection by Plasmodium falciparum cause oxidative stress that induces aggregates of cross-linked proteins with N-linked high-mannose glycans (HMGs). These glycans can be recognised by mannose-binding lectins, including the mannose receptor (CD206), expressed on macrophages and specialised phagocytic endothelial cells in the spleen to mediate the extravascular haemolysis characteristic of these diseases. We postulated this system might al…

Might there be a link between mannose binding lectin and vitiligo?

Mannose binding lectin (MBL) is a calcium dependent lectin that causes predisposition to infections and autoimmune diseases. This study aimed to examine the presence of any association between MBL2 gene variants and vitiligo. Codon 54 (allele B) and codon 57(allele C) polymorphisms in the exon 1 of the MBL2 gene were investigated in samples belonged to 50 healthy controls and 40 patients diagnosed as vitiligo. The PCR-RFLP method was used to investigate the polymorphisms in the MBL2 gene. Codon 57 polymorphism was not detected in any of the subjects from either group. The frequencies of low le…

Impact of mannose-binding lectin on susceptibility to infectious diseases.

When the adaptive immune response is either immature or compromised, the innate immune system constitutes the principle defense against infection. Mannose-binding lectin (MBL) is a C-type serum lectin that plays a central role in the innate immune response. MBL binds microbial surface carbohydrates and mediates opsonophagocytosis directly and by activation of the lectin complement pathway. A wide variety of clinical isolates of bacteria, fungi, viruses, and parasites are bound by MBL. Three polymorphisms in the structural gene MBL2) and 2 promoter gene polymorphisms are commonly found that res…

The structure and function of vertebrate mannose lectin-like proteins.

Sugar-specific recognition is now well established as an important determinant of cell-cell interactions and host defence mechanisms. Macrophages, in particular, are known to express a variety of lectin-like proteins that are specific for oligosaccharides terminating in mannose, fucose, galactose and sialic acid. This review focuses on mannose recognition systems. Terminal mannose is rarely found on mammalian cell surfaces whereas it is ubiquitous on the surfaces of lower organisms. Mammals have evolved at least two mechanisms to recognize terminal mannose residues. These are, first, a mannose…

Pharmacodynamics of D-mannose in the prevention of recurrent urinary infections.

Recurring urinary tract infections (rUTIs) are frequently caused by Escherichia coli, which invades urothelial cells and forms quiescent bacterial reservoirs. D-mannose, an inert monosaccharide, represents a notable agent for rUTI prevention; however, there is no agreement on its dosage. To provide pharmacological basis for an effective dose, we evaluated its ability to inhibit adhesion of E. coli to urothelial cells. E. coli strains isolated from the urine of a woman with recurrent urinary tract infections were selected according to adhesion capacity. Anti-adhesive efficacy and invasion were …

Mannose Receptor and the Mystery of Nonhealing Leishmania major Infection.

Scientists have long puzzled over the ability of Leishmania major Seidman (LmSd) to form nonhealing cutaneous lesions in the face of a strong Th1 response. A recent study identified a population of dermal macrophages that are preferentially infected by LmSd in a mannose receptor 1-, C-type 1 (MRC1/CD206)-dependent manner.

Cloning and characterization of a novel mannose-binding protein of Acanthamoeba.

Acanthamoebae produce a painful, blinding infection of the cornea. The mannose-binding protein (MBP) of Acanthamoeba is thought to play a key role in the pathogenesis of the infection by mediating the adhesion of parasites to the host cells. We describe here the isolation and molecular cloning of Acanthamoeba MBP. The MBP was isolated by chromatography on the mannose affinity gel. Gel filtration experiments revealed that the Acanthamoeba lectin is a approximately 400-kDa protein that is constituted of multiple 130-kDa subunits. Cloning and sequencing experiments indicated that the Acanthamoeba…

A potent mannose-modified pillararene-BODIPY system for photodynamic therapy.

Photodynamic therapy (PDT) offers a promising, non-invasive approach to cancer treatment. However, its efficacy is often limited by the poor water solubility, low cellular uptake, and high dose requirements of traditional photosensitizers, which can lead to side effects like skin photosensitivity. This study presents a novel supramolecular photosensitizer, PS3⊂WP5, comprising a mannosylated pillar[5]arene (WP5) host and a near-infrared BODIPY photosensitizer (PS3). This host-guest complex exhibits a strong binding affinity (Ka = 5.10 × 106 M-1) and self-assembles into nanoparticles in water. P

Agaricus bisporus Mannose-Binding Protein Stimulates the Innate Immune Cells.

PURPOSE: A lectin-like protein from the mushroom Agaricus bisporus has been shown to slightly increase the proliferation of RAW 264.7 cells. Following its identification as a mannose-binding lectin, henceforth called A. bisporus mannose-binding protein (Abmb), the protein is hypothesized to stimulate the innate immune cells response. The present work was aimed to substantiate that hypothesis. Furthermore, this study complements Abmb exploration as a potential agent for anti-breast cancer, which its treatment is hampered with compromised immunity of patient receiving chemotherapy. METHODS: Abm…

Mannose-binding lectin (MBL) levels in children with Hashimoto's thyroiditis.

OBJECTIVE: Hashimoto's thyroiditis (HT) was described many years ago, but the etiopathogenesis remains unclear. Mannose-binding lectin (MBL) initiates complement activation in the lectin pathway. We determined MBL levels in children with HT and the associations thereof with thyroid hormone and thyroid autoantibody levels. METHODS: Thirty-nine patients with HT and 41 controls were enrolled from the pediatric outpatient clinics. Subjects were grouped according to their thyroid functions: Euthyroid, marked hypothyroidism and clinical/subclinical hyperthyroidism. MBL levels were compared among th…

Introducing carbohydrate patterning in mannose-decorated supramolecular assemblies and hydrogels.

Benzene-1,3,5-tricarboxamide (BTA) glyco-monomers containing one, two or three mannose units are synthesized and formulated into differently patterned supramolecular glycopolymers through homo-assembly or co-assembly with non-functionalized BTAs. Unfortunately, no cellular activity could be detected. Excitingly, these glyco-BTA monomers could be formulated into hydrogels, paving the way for (immune) cell culture.

Mannose-Binding Lectins as Potent Antivirals against SARS-CoV-2.

The SARS-CoV-2 entry into host cells is mainly mediated by the interactions between the viral spike protein (S) and the ACE-2 cell receptor, which are highly glycosylated. Therefore, carbohydrate binding agents may represent potential candidates to abrogate virus infection. Here, we evaluated the in vitro anti-SARS-CoV-2 activity of two mannose-binding lectins isolated from the Brazilian plants Canavalia brasiliensis and Dioclea violacea (ConBR and DVL). These lectins inhibited SARS-CoV-2 Wuhan-Hu-1 strain and variants Gamma and Omicron infections, with selectivity indexes (SI) of 7, 1.7, and …

Effect of D-mannose on Philadelphia chromosome-positive leukemia cells.

BACKGROUND: Although Abelson (ABL) tyrosine kinase inhibitors (TKIs) have demonstrated potency against chronic myeloid leukemia (CML), resistance to ABL TKIs can develop in CML patients after discontinuation of therapy. OBJECTIVE: Glucose metabolism may be altered in CML cells because glucose is a key metabolite used by tumor cells. We investigated whether D-mannose treatment induced metabolic changes in CML cells and reduced CML growth in the presence of ABL TKIs. METHODS: We investigated whether D-mannose treatment induced metabolic changes in CML cells and reduced CML growth in the presen…
