The status and consequences of Mansonella perstans infection in Calabar, Nigeria.

Two aetiological studies on filariasis were conducted from January to June, 1993. In the first study, 690 blood samples clinically diagnosed of filariasis were examined. In the other, 150 blood samples and 150 skin snips collected from the same group of eye clinic Patients clinically diagnosed of filariasis were investigated. Of the 690 subjects, 108 (15.64%) had filariasis with M. perstans and Loa loa accounting for 98.15 and 1.85% of positive cases respectively. Among the eye clinic patients, 16% had M. perstans while O. volvulus was seen in 4% of patients. There was no concurrent infection

Comparison of different anthelminthic drug regimens against Mansonella perstans filariasis.

Mansonella perstans filariasis is widely present in Africa and equatorial America and its pathogenicity has recently been reconsidered. Effective treatment is lacking and there is no consensus on the optimal therapeutic approach. The aim of this study was to compare the effects of different drug regimens on M. perstans infection. Six different anthelminthic therapeutic protocols were undertaken on 165 subjects with M. perstans infection and their effects on microfilariae burden were evaluated. Diethylcarbamazine (DEC) was able to reduce microfilariae density in the majority of cases, but it se…

Infection with Mansonella perstans Nematodes in Buruli Ulcer Patients, Ghana.

During August 2010-December 2012, we conducted a study of patients in Ghana who had Buruli ulcer, caused by Mycobacterium ulcerans, and found that 23% were co-infected with Mansonella perstans nematodes; 13% of controls also had M. perstans infection. M. perstans co-infection should be considered in the diagnosis and treatment of Buruli ulcer.

Molecular identification of Wolbachia from the filarial nematode Mansonella perstans.

Wolbachiae are bacterial endosymbionts of insects and many filarial nematodes whose products trigger inflammation in filarial infections. The dependence of the parasites on their endosymbionts has also led to the use of antibiotics directed against the Wolbachiae, therapy that has been demonstrated to have a profound salutary effect on filarial infections. The identification of Wolbachiae in Mansonella species has been conclusively shown for Mansonella ozzardi (Mo), but not for Mansonella perstans (Mp). Using primers known to amplify the 16S ribosomal DNA of other filarial Wolbachiae, an ident…

Emergence of Mansonella sp. in free-ranging primates in southern Brazil.

Mansonellosis is a neglected and emerging tropical disease. Among all zoonotic filarial diseases, it is probably the most prevalent and least studied, with approximately 114 million people infected. The parasites of Mansonella spp. are among the most common blood parasitemias and are widely found in Africa and Latin America. Through molecular analysis of blood samples from free-ranging primates Sapajus nigritus (n 33) and Alouatta guariba clamitans (n 5) in the southern states of Brazil (Santa Catarina and Rio Grande do Sul), we identified samples positive for Mansonella perstans in two specim…

Mansonella ozzardi corneal lesions in the Amazon: a cross-sectional study.

OBJECTIVES: To characterise and confirm the presence of Mansonella ozzardi microfilariae in the cornea by biomicroscopy and corneal confocal microscopy. DESIGN: Cross-sectional study. SETTINGS: Clinical practice study in patients from rural communities in Coari city on the Solimões river, Amazonas state, Brazil. PARTICIPANTS: The eyes of 212 consecutive volunteer patients were examined using a flash light and their blood checked for the presence of microfilariae by an expert microscopist. Patients with suspicious corneal lesions (characterised as nummular keratitis) were submitted to biomic…

The morphological discrimination of microfilariae of Onchocerca volvulus from Mansonella ozzardi.

There is no published account which allows the morphological discrimination of microfilariae of Onchocerca volvulus and M. ozzardi from each other. However, they occur together in parts of Brazil and Venezuela, and presumably there is always the possibility that migration could establish new sympatric populations in the future. The objective of this study was to evaluate simple morphological characters that might be used for species-diagnosis of microfilariae. The conclusions were that the location of microfilariae in the blood or skin, the body size and the nucleation of the nerve ring are ex

Hypogonadism and ecdysteroid production in Loa loa and Mansonella perstans filariasis.

Possible endocrinological repercussions of infection with Loa loa and Mansonella perstans filariae were studied in Gabonese subjects. Microfilaremic males were compared with amicrofilaremic controls. In the infected group 13/105 subjects (12%) presented only abnormally low serum levels of testosterone (less than 4 ng/ml), 25/105 (24%) only abnormally high serum levels of gonadotrophins, FSH (greater than 15 mIU/ml) and LH (greater than 20 mIU/ml), and 22/105 (21%) presented anomalies in both testosterone and gonadotrophin levels. One out of 68 control subjects had 3.6 ng/ml seric testosterone …

Susceptibility of laboratory primates to infection with Mansonella ozzardi from man.

The study was undertaken to determine primates were susceptible to infection with Caribbean and/or Amazon strains of Mansonella ozzardi of man. Twenty-three animals including three chimpanzees, four squirrel monkeys, one capuchin, five rhesus and 10 patas monkeys each received, by subcutaneous or intraperitoneal injection, 25-170 infective larvae of M. ozzardi harvested either from infected Culicoides furens collected in Haiti or Simulium sp. (sanguineum complex) collected in the Colombian Amazon and transported to our laboratories in Louisiana. Patent infections were obtained in seven of 10 p…

The prevalence and distribution of Mansonella ozzardi in coastal north Trinidad, W.I.

A day blood smear survey for Mansonella ozzardi in the north coast communities of Trinidad recorded a prevalence of 4.8% in 4,488 persons examined. Prevalence rates were highest in the four western-most communities, where the vector, Culicoides phlebotomus, is a severe nuisance problem. Prevalence rates increased with age and were higher in males than in females. Mean microfilaria densities were low in both sexes up to 50 years of age but in older males the density increased with age. Wuchereria bancrofti infections were detected in five of the ten communities surveyed.

Further evidence for the transmission of Mansonella ozzardi by Simulium amazonicum in Brazil.

The transmission of Mansonella ozzardi was studied in two rubber collecting villages on the River Purus, state of Amazonas, Brazil. Haematophagous insects were collected from human and bovine baits during the day and night: 687 Mansonia amazonensis and 154 Culicoides sp. indet. were free from infection with M. ozzardi. The former species is probably not a vector, but the low numbers of culicoides dissected preclude any determination of its vector status. Thirty-five (0-99%) of 3530 Simulium amazonicum dissected were found naturally infected with larvae of M. ozzardi. Two hundred and ninety-nin…

High Prevalence of Mansonella ozzardi Infection in the Amazon Region, Ecuador.

We reviewed Giemsa-stained thick blood smears, obtained through the national malaria surveillance program in the Amazon region of Ecuador, by light microscopy for Mansonella spp. microfilariae. Of 2,756 slides examined, 566 (20.5%) were positive. Nested PCR confirmed that the microfilariae were those of M. ozzardi nematodes, indicating that this parasite is endemic to this region.

[Loa Loa and Mansonella perstans coinfection in a patient from Guinea].

We present a case of mixed infection by two filariae (Loa loa and Mansonella perstans). It seems that the infection was suspected due to a pronounced eosinophilia in a routine analysis. Sheathed and unsheathed microfilariae were observed in the spread of peripheral blood, which enabled a diagnosis to be established of mixed infection by filariae. The definitive diagnosis of the species was carried out at the National Centre of Microbiolgy of Majadahonda. A treatment was initiated with dietilcarbamazina and mebendazole which resulted in the patient's being cured and in the elimination of the mi…

Onchocercosis mansonella co-infection presenting as a subcutaneous nodule in a child.

A 11-year-old girl presented with a subcutaneous lesion on the forehead, thought to be an implantation dermoid cyst. Microscopic examination revealed an onchocercoma. Microfilariae were found in the blood indicating a co-infection with Mansonella perstans. This case demonstrates the diagnostic difficulties of a parasitic co-infection uncommonly encountered in Europe.

Mansonella ozzardi: the course of patency in experimentally-infected patas monkeys.

Twenty-five of 30 patas monkeys (Erythrocebus patas) inoculated with varying numbers (35 to 135) of third-stage larvae of Mansonella ozzardi developed patent infections in an average of 163 days. There was no correlation between the size of the inoculum and the length of the prepatent period. Ten of the monkeys were monitored thereafter by regular blood examination for extended periods of time in order to characterize the onset, course and duration of patency. Typically, with the onset of patency, microfilaremias increased steadily, peaking at about 20 weeks and then decreased slowly stabilizi…
