Mercuric chloride enhances immunoglobulin E-dependent mediator release from human basophils.

Mercuric chloride (HgCl2) is an industrial agent known to cause autoimmune disorders and induce IgE synthesis, which plays a crucial role in the manifestation of allergic diseases. In rodents, the immunomodulatory effects of HgCl2 have been shown to involve the enhancement of mast cell-derived IL-4 secretion, which facilitates both Th2-lymphocyte development and IgE production. In humans, rapid allergen-dependent release of IL-4 and the related cytokine IL-13 from histamine-containing cells occurs primarily in basophils, along with other proinflammatory mediators such as histamine and LTC4. Effectiveness of N-acetylcysteine in protecting against mercuric chloride-induced nephrotoxicity.

Mercuric chloride (HgCl2)-induced nephrotoxicity, as measured by functional and biochemical parameters was evaluated in rats at different kidney non-protein sulfhydryls (NPS) levels. Diethylmaleate (DEM) induced a 75% of NPS diminution 1 h after the administration. Renal function (clearance) and biochemical measurements (gamma-glutamyltranspeptidase activity in urine, and lipoperoxides in kidney tissue) were impaired when the animals were HgCl2-treated. Values were highly impaired when the kidneys were NPS-depleted and were improved when NPS pools were previously increased although they were n

Plasma mercury during prolonged acute renal failure after mercuric chloride ingestion.

Serial measurements of plasma mercury were made in a patient with severe and prolonged acute renal failure due to poisoning with mercuric chloride. An initial mercury concentration in whole blood of 1200 micrograms/l (6 mumol/l) was recorded, and recovery of renal function coincided with a fall in plasma mercury concentration to below 100 micrograms/l (0.5 mumol/l). The case demonstrates that survival and recovery of renal function is possible despite very high concentrations of mercury in the blood and oliguric renal failure of nearly six weeks' duration.

Complement studies in BN rats with mercuric chloride-induced immune glomerulonephritis.

Whole complement haemolytic activity (CH50) changes were examined in 71 rats, developing and HgCl2-induced biphasic immune glomerulonephritis. In some rats, serum C4 level was also studied. Whole CH50 level was compared with proteinuria and C3 glomerular deposits. In control rats or in rats injected with low doses of HgCl2, there was no complement decrease. In rats injected with a high dose, after an initial increase (also observed in controls), CH50 level decreased between day 12 and day 20, sometimes dramatically. This decrease, observed during the first phase, was transient, CH50 levels ret

Lactating and nonlactating rats differ to renal toxicity induced by mercuric chloride: the preventive effect of zinc chloride.

This study evaluated the effects of HgCl2 on renal parameters in nonlactating and lactating rats and their pups, as well as the preventive role of ZnCl2 . Rats received 27 mg kg(-1) ZnCl2 for five consecutive days and 5 mg kg(-1) HgCl2 for five subsequent days (s.c.). A decrease in δ-aminolevulinic acid dehydratase (δ-ALA-D) activity in the blood and an increase in urine protein content in renal weight as well as in blood and urine Hg levels were observed in lactating and nonlactating rats from Sal-Hg and Zn-Hg groups. ZnCl2 prevented partially the δ-ALA-D inhibition and the proteinuria in non

Localization of mercury in CNS of the rat. II. Intraperitoneal injection of methylmercuric chloride (CH3HgCl) and mercuric chloride (HgCl2).

The autometallographic method has been used to determine the precise localization of mercury in the brain and spinal cord of adult Wistar rats which had been treated with repeated ip injections of methylmercuric chloride (CH3HgCl; 0.2 to 10.0 mg) or mercuric chloride (HgCl2; 0.2 to 10.0 mg). The distribution of mercury was uneven following administration of HgCl2, while it was fairly homogeneous following CH3HgCl. With both compounds, however, heavy deposits of mercury were present in the motor nuclei of rhombencephalon. In contrast, cerebellar Purkinje cells, Golgi cells, and Golgi epithelial

Protective effect of diallylsulphide against mercuric chloride-induced hepatic injury in rats.

The present study was undertaken to evaluate the effect of diallylsulphide (DAS) against mercuric chloride (HgCl2)-induced oxidative stress in rat livers. Rats were randomly divided into four groups of six rats each and exposed to HgCl2 (50 mg/kg/body weight (b.w.)) intraperitoneally and/or DAS (200 mg/kg/b.w.) by gavage. HgCl2 administration enhanced alanine aminotransferase (AST) and aspartate aminotransferase (ALT) levels (p < 0.05) with reduction in the levels of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px). However, treatment with DAS markedly attenuated

Effect of thimerosal, methylmercury, and mercuric chloride in Jurkat T Cell Line.

Mercury is a ubiquitous environmental toxicant that causes a wide range of adverse health effects in humans. Three forms of mercury exist: elemental, inorganic and organic. Each of them has its own profile of toxicity. The aim of the present study was to determine the effect of thimerosal, a topical antiseptic and preservative in vaccines routinely given to children, methyl mercury, and mercuric chloride on cellular viability measured by MTT in Jurkat T cells, a human T leukemia cell line. The treatment of Jurkat T cells with thimerosal caused a significant decrease in cellular viability at 1

Use of methyl prednisolone and antioxidants in mercuric chloride-induced experimental vasculitis.

The systemic vasculitides are characterized by necrotizing inflammation of blood vessels. Neutrophils are implicated in tissue damage by their presence at the site of injury. They can mediate injury by release of cellular contents including proteinases, cytokines and reactive oxygen species. Antioxidants such as vitamin E and N-acetyl cysteine (NAC) may therefore be predicted to ameliorate oxidative damage in vivo and could be a cheap and non-toxic form of therapy. We examined this hypothesis in an experimental model of vasculitis which has some similarities to human disease, and in which depl

Mercuric chloride induces autoantibodies against U3 small nuclear ribonucleoprotein in susceptible mice.

Autoantibodies to nucleolar components are a common serological feature of patients suffering from scleroderma, a collagen vascular autoimmune disease. While animal models, which spontaneously develop abundant anti-nucleolar antibodies, have not yet been described, high titers of such antibodies may be induced by treating susceptible strains of mice with mercuric chloride. We have identified the nucleolar autoantigen against which the HgCl2-induced IgG autoantibodies from mice of strain B10.S are directed. It is a protein with an apparent molecular mass of 36 kDa and a pI value of approximatel

Effects of decomplementation on mercuric chloride-induced glomerulonephritis in Brown-Norway rats.

The course of mercuric chloride-induced immune glomerulonephritis is characterized by complement activation, intensive proteinuria, linear and then granular IgG and C3 deposits in the glomeruli. To assess the role of complement activation in the occurrence of the disease, decomplementation was achieved by intravenous injections of cobra venom factor in rats injected with mercuric chloride. In these animals, proteinuria still appeared while rats were decomplemented by cobra venom factor through the alternative pathway. These rats exhibited linear IgG deposits without detectable C3 deposits. In

Alterations of the renal papilla during mercuric chloride-induced acute tubular necrosis.

Marked morphologic changes were seen in all structural elements of the renal papilla during mercuric chloride-induced acute tubular necrosis. Expect for the descriptions of increased numbers of leukocytes within the medullary vasa recta, papillary changes have not been described previously. The techniques used in this study included light microscopy, scanning electron microscopy, and transmission electron microscopy. The renal interstitial cells developed an extremely irregular contour and were penetrated by large irregular cavelike structures. A marked decrease occurred in the size and number

N-acetylcysteine pretreatment ameliorates mercuric chloride-induced oxidative renal damage in rats.

The effectiveness of the antioxidant thiol, N-acetylcysteine (NAC), in enhancing methylmercury (CH3HgCl) excretion and its utility as a possible antidote in CH3HgCl poisoning has been reported. NAC, however, has been reported to be ineffective in accelerating excretion of divalent toxic metals, including inorganic mercury, Hg2+. In this study, we evaluated the possible protective effect of short-term pretreatment with NAC against mercuric chloride (HgCl2) toxicity in rat model. This is aimed at determining its chemopreventive or prophylactic benefit in situations of high risk exposure (occupat

Metal accumulation and nephron heterogeneity in mercuric chloride-induced acute renal failure.

The present study was designed to assess the effects of mercury on glomerular integrity during the early phase of acute renal failure. The silver amplification method showed distribution of mercury in midcortical and juxtamedullary glomeruli and on the brush border of the S2 segment of the proximal tubule 15 min after treatment. At 30 min, there was a decrease in glomerular staining and increased mercury in the proximal tubule. After 3 hr, mercury was no longer detectable in glomeruli but was widespread in the lumen of the proximal tubule. By 24 hr, mercury was prominent in all proximal tubula

Histochemical studies of oxidoreductases in rat kidney regenerating after mercuric chloride injury.

The activities of succinic dehydrogenase, glucose-6-phosphate dehydrogenase, lactate dehydrogenase and cytochrome oxidase, representatives of the tricarboxylic acid cycle, pentose phosphate pathway, glycolysis and terminal oxidation, respectively, were investigated after necrosis induced with mercuric chloride. 24 hours after the administration of mercuric chloride, the activities of all four enzymes were decreased in the necrotic proximal tubular segments. By the third day of regeneration the activities had decreased or disappeared in all three portions of the necrotic proximal tubules as com
