Detection of organomercurials with sensor bacteria.

Mercury and its organic compounds, especially methylmercury, are hazardous compounds that concentrate in biota via biomagnification and cause severe neurological disorders in animals. In this paper, a recombinant whole-cell bacterial sensor for the detection of the organic compounds of mercury was constructed. The sensor carries firefly luciferase gene as a reporter under the control of the mercury-inducible regulatory part of broad spectrum mer operon from pDU1358. In addition, a gene-encoding organomercurial lyase (an enzyme necessary for cleavage of the mercury-carbon bond) was coexpressed

[Bibliographical study of the toxicity of organic mercury compounds].

OBJECTIVES: The aim of this study is to correct the misunderstanding that the toxicity of organic mercury compounds is unknown at the time of the outbreak of Minamata disease (May 1, 1956). RESULTS AND DISCUSSION: Two case reports of organic mercury (methylmercury) intoxication were published already in 1865 and 1866. The conversion of inorganic mercury added in acetoaldehyde synthesis was already pointed out in 1921. In 1930 several cases of organic mercury poisoning among workers engaged in acetoaldehyde production were reported. Many reports on not only in occupational exposure but a oral

The early effects of methylmercury on the developing rat brain.

The effects of organic mercury compounds on the development of the brain are well known since the exposure of people at a large scale to methylmercury in the Minamata Bay area and in Iraq. The neuropathological examination of the brains of children prenatally exposed revealed dysplasia of the cerebral and cerebellar cortex, neuronal ectopia and several other developmental disturbances. In this experimental study we examined developmental mechanisms involved in methylmercury-induced cerebral anomalies. By examining the fetuses soon after treatment we concentrated in the initial effects of the t

Inactivation by Hg2+ and methylmercury of the glutamine/amino acid transporter (ASCT2) reconstituted in liposomes: Prediction of the involvement of a CXXC motif by homology modelling.

The effect of HgCl(2), methylmercury and mersalyl on the glutamine/amino acid (ASCT2) transporter reconstituted in liposomes has been studied. Mercuric compounds externally added to the proteoliposomes, inhibited the glutamine/glutamine antiport catalyzed by the reconstituted transporter. Similar effects were observed by pre-treating the proteoliposomes with the mercurials and then removing unreacted compounds before the transport assay. The inhibition was reversed by DTE, cysteine and N-acetyl-cysteine but not by S-carboxymethyl-cysteine. The data demonstrated that the inhibition was due to c

Molecular Fates of Organometallic Mercury in Human Brain.

Mercury is ubiquitous in the environment, with rising levels due to pollution and climate change being a current global concern. Many mercury compounds are notorious for their toxicity, with the potential of organometallic mercury compounds for devastating effects on the structures and functions of the central nervous system being of particular concern. Chronic exposure of human populations to low levels of methylmercury compounds occurs through consumption of fish and other seafood, although the health consequences, if any, from this exposure remain controversial. We have used high energy res…

Genotoxicity of Mercury and Its Derivatives Demonstrated In Vitro and In Vivo in Human Populations Studies. Systematic Review.

Beside partial coverage in three reviews so far (1994, 2009, 2019), there is no review on genotoxic studies dealing with mercury (Hg) and human exposure using the most usual genotoxic assays: sister chromatid exchanges (SCE), chromosomal aberrations (CA), cytochalasin B blocked micronucleus assay (CBMN), and single-cell gel electrophoresis (SCGE or alkaline comet assay). Fifty years from the first Hg genotoxicity study and with the Minamata Convention in force, the genotoxic potential of Hg and its derivatives is still controversial. Considering these antecedents, we present this first systema

[Methylmercury human exposure in riverside villages of Tapajos basin, Pará State, Brazil].

Evaluation of total human exposure to mercury and methylmercury was effected in riverine villages along the Tapajos river and in the metropolitan area of Belem city, state of Para, Brazil, by using total mercury and methylmercury concentrations in hair samples in 1994 and 1995. It was observed that average concentrations of total mercury are in a range from 2 +/- 1 microg/g-1 to 20.5 +/- 12. 1 microg/g-1. While methylmercury average concentration varies from 1. 4 +/- 0.7 microg/g-1 to 18.5 +/- 11 microg/g-1. These results confirm mercury contamination in the Tapajos river and possible appearan

Synergistic Effects of a Chalkophore, Methanobactin, on Microbial Methylation of Mercury.

Microbial production of the neurotoxin methylmercury (MeHg) is a significant health and environmental concern, as it can bioaccumulate and biomagnify in the food web. A chalkophore or a copper-binding compound, termed methanobactin (MB), has been shown to form strong complexes with mercury [as Hg(II)] and also enables some methanotrophs to degrade MeHg. It is unknown, however, if Hg(II) binding with MB can also impede Hg(II) methylation by other microbes. Contrary to expectations, MB produced by the methanotroph Methylosinus trichosporium OB3b (OB3b-MB) enhanced the rate and efficiency of Hg(I

[Investigation of the effect of polluted water in Song-hua River on the chromosome aberrations and SCE of inhabitants].

This investigation indicated that the chromosome aberration of lymphocytes in inhabitants influenced by mercury- or methylmercury-polluted water was higher than that in control persons although the difference was not significant; but the SCE of them was significantly higher than the control. Both endpoints had a tendency of decreasing with the distance away from the polluted river water. Smoking and age did not have any effect. It was suggested that the polluted river water by mercury or methylmercury compounds could have some cytogenetic effects, although other factors cannot be excluded.

Chemical forms of mercury and selenium in fish following digestion with simulated gastric fluid.

Fish is a major dietary source of potentially neurotoxic methylmercury compounds for humans. It is also a rich source of essential selenium. We have used in situ mercury L(III)-edge and selenium K-edge X-ray absorption spectroscopy to chemically characterize the methylmercury and selenium in both fresh fish and fish digested with simulated gastric fluid. For the mercury, we confirm our earlier finding [ Harris et al. ( 2003 ) Science 301 , 1203 ] that the methylmercury is coordinated by a single thiolate donor, which resembles cysteine, and for the selenium, we find a mixture of organic forms …

Mechanisms of mercury disposition in the body.

Today the most widespread human exposures to mercury are to mercury vapor emitted from amalgam tooth fillings, to ethylmercury as a preservative in vaccines, and to methylmercury in edible tissues of fish. This review will focus on the mechanisms of transport of these three species of mercury. All three species are freely moveable throughout the body. Inhaled vapor in view of its physical properties as an uncharged atomic gas is believed to be transported by passive diffusion. Methylmercury and ethylmercury also move freely in the body. Methylmercury, and presumably its closely related chemica

Genotoxicity of mercury compounds. A review.

This article reviews literature data concerning the genotoxicity of 29 mercury-containing agents, including laboratory compounds as well as ingredients of preparations used as fungicides, dyes, disinfectants and drugs. A variety of genetic end-points were investigated in bacteria, yeasts, moulds, plants, insects, cultured cells from fishes, rodents or humans, aquatic organisms, amphibians, mammalia and exposed humans. The overall evaluation is quite complex. Mercury compounds failed to induce point mutations in bacteria but often exerted clastogenic effects in eukaryotes, especially by binding

Occupational and environmental toxicology of mercury and its compounds.

Mercury exists in various chemical forms. The important forms from a toxicological viewpoint are the metallic form, also called the elemental form, the divalent inorganic forms and methylmercury compounds. Elemental (Hg0) mercury has a high vapor pressure and the vapor causes a number of cases of poisoning via inhalation. Classical mercury poisoning is characterized by a triad of signs, namely tremors, erethism and gingivitis. Mercurial erethism, which is characterized by behavioral and personality changes such as extreme shyness, excitability, loss of memory, and insomnia are also observed. R…

Secondary sex ratio in regions severely exposed to methylmercury "Minamata disease".

PURPOSE: Secondary sex ratio (i.e., male proportion at birth) is considered to function as a sentinel health indicator. Thus, examining this ratio spatially and temporally in regions with severe environmental exposure to compounds such as methylmercury may provide insight into the evolution of exposure. METHODS: We evaluated spatial and temporal distributions of the secondary sex ratio in Minamata, Japan, and neighboring areas, where severe methylmercury poisoning occurred in the 1950s and 1960s. We selected four areas exposed to methylmercury: Minamata, Ashikita, Goshonoura, and Izumi. After

Mercury compounds disrupt neuronal glutamate transport in cultured mouse cerebellar granule cells.

Cerebellar granule cells are targeted selectively by mercury compounds in vivo. Despite the affinity of mercury for thiol groups present in all cells, the molecular determinant(s) of selective cerebellar degeneration remain to be elucidated fully. We studied the effect of mercury compounds on neuronal glutamate transport in primary cultures of mouse cerebellar granule cells. Immunoblots probed with an antibody against the excitatory amino acid transporter (EAAT) neuronal glutamate transporter, EAAT3, revealed the presence of a specific band in control and mercury-treated cultures. Organomercurials. Their formation and pathways in the environment.

The most important mercury species in the environment is monomethylmercury (MMHg), the topic of this chapter. This organic mercury compound is normally not released into the environment but formed by natural processes. Mercuric mercury (Hg²(+)) is methylated by bacteria and to a lesser extent through abiotic pathways. Highest rates of formation are found in anoxic aquatic environments. Terrestrial systems are mostly irrelevant for MMHg production and not a concern. Most productive environments are sediments, wetlands, and coastal marshes, but also the anoxic hypolimnion of lakes and anaerobic

Kinetics of methylmercury and inorganic mercury in lactating and nonlactating mice.

The elimination of mercury was followed for 9 days (Days 10-19 of lactation) in milk and/or 21 days in blood and plasma of lactating and nonlactating mice administered a single iv injection of either 203Hg-labeled methylmercuric chloride or 203Hg-labeled mercuric chloride (0.5 mg Hg/kg body wt). Demethylation of methylmercury to inorganic mercury was taken into consideration by analyzing the data with a combined pharmacokinetic model based on the assumption of constant blood plasma ratios for methylmercury and inorganic mercury. A three-compartment model fitted the blood and plasma concentrati

Environmental impacts of the Tennessee Valley Authority Kingston coal ash spill. 2. Effect of coal ash on methylmercury in historically contaminated river sediments.

The Tennessee Valley Authority Kingston coal ash spill in December 2008 deposited approximately 4.1 million m(3) of fly ash and bottom ash into the Emory and Clinch River system (Harriman, Tennessee, U.S.A.). The objective of this study was to investigate the impact of the ash on surface water and sediment quality over an eighteen month period after the spill, with a specific focus on mercury and methylmercury in sediments. Our results indicated that surface water quality was not impaired with respect to total mercury concentrations. However, in the sediments of the Emory River near the coal a

Genotoxic evaluation of different doses of methylmercury (CH₃Hg⁺) in Hoplias malabaricus.

The survey of the effects of toxic metals and its organic compounds are of critical importance because these compounds tend to accumulate in aquatic environments. In the present work, the genotoxic potential of methylmercury, an organomercurial compound with high toxicity and present in large amounts in fish of the Amazon due to the mining process, was evaluated using the piscine micronucleus test and comet assay in fish. Specimens of Hoplias malabaricus (popularly known as traira), a neotropical specie, was exposed to methylmercury via food web, over 70 days, in two different concentrations:
