Multiplex Real-Time PCR Assay Targeting Eight Parasites Customized to the Korean Population: Potential Use for Detection in Diarrheal Stool Samples from Gastroenteritis Patients.

Intestinal parasitic diseases occur worldwide and can cause diarrhea or gastroenteritis; however, their diagnosis is quite difficult, especially in low-endemism countries. We developed a multiplex real-time PCR assay for detection of eight intestinal parasites and prospectively evaluated it for patients with gastroenteritis. The assay targeted Cryptosporidium parvum, Giardia lamblia, Entamoeba histolytica, Blastocystis hominis, Dientamoeba fragilis, Clonorchis sinensis, Metagonimus yokogawai, and Gymnophalloides seoi. Performance characteristics were evaluated based on recovery after DNA extra

Locomotion of sponges and its physical mechanism.

Active locomotion by individual marine and freshwater sponges across glass, plastic and rubber substrata has been studied in relation to the behavior of the sponges' component cells. Sequential tracing of sponge outlines on aquarium walls shows that sponges can crawl up to 160 microns/hr (4 mm/day). Time-lapse cinemicrography and scanning electron microscopy reveal that moving sponges possess distinctive leading edges composed of motile cells. Sponge locomotion was found to be mechanically similar to the spreading of cell sheets in tissue culture both with respect to exertion of traction (whic

Medicinal plant extracts interfere in gastric cancer stem cells fluorescence-based assays.

Fluorescence is used in various biological assays due to its high sensitivity, versatility, and precision. In recent years, studies using medicinal plant extracts have increased. However, fluorescence-based assays could be biased by plant metabolites autofluorescence. To address this issue, this study investigated the interference caused by methanolic extracts and chloroform fractions of three medicinal plants in three fluorescence-based assays on gastric cancer stem cells(CSC): resazurin reduction, confocal microscopy, and flow cytometry. CSC were isolated based on CD44 surface marker, incuba

EFFECT OF ADENYLATE KINASE 1 ON THE GROWTH AND DEVELOPMENT OF SCHISTOSOMA JAPONICUM SCHISTOSOMULUM.

We investigated the effect of Schistosoma japonicum adenylate kinase 1 (Sjak1) on the growth and development of schistosomula. Quantitative real-time PCR showed that Sjak1 mRNA was expressed in 3-, 10-, 14-, 18-, and 21-day-old schistosomula, and its levels increased gradually with the development of S. japonicum. Using immunohistochemical techniques, ak1 protein was found to be mainly distributed in the tegument and some parenchymal tissues of the schistosomula. Double-stranded RNA-mediated knockdowns of ak1 decreased ak1 mRNA transcripts by more than 90%, and western blot results showed that

Role of adenylate kinase 1 in the integument development of Schistosoma japonicum schistosomula.

Schistosomula antigens play an important role in the growth and development of Schistosoma japonicum. We investigated the role of S. japonicum adenylate kinase 1 (SjAK1) in the growth and development of schistosomula. Quantitative real-time PCR showed that SjAK1 mRNA was expressed in all schistosomula stages, but increased gradually with the development of S. japonicum schistosomula. Using immunohistochemical techniques, the AK1 protein was found to be mainly distributed in the tegument and in some parenchymal tissues of the schistosomula. Back-illumination interference tomography for imaging weak scattering in thick tissues.

Biological tissues are composed of discrete compartments of biochemical media that often exhibit subtle differences in refractive index. Light propagating through these compartments partially diffracts in a forward direction with a π / 2 phase shift. We introduce a microscopy technique to image this scattering signal in thick tissues, called back-illumination interference tomography (BIT). An incoherent source is demagnified and imaged past the focal plane of a high-numerical aperture objective lens, producing a small, semicoherent source of backscattered light. This backscattered light underg

Anti-proteasome autoantibodies contribute to anti-nuclear antibody patterns on human larynx carcinoma cells.

BACKGROUND: Autoantibodies to the 20S proteasome represent an unspecific but common serological phenomenon in patients with systemic autoimmune diseases. Interestingly, a high prevalence of these antibodies have been observed in patients with connective tissue diseases, where anti-nuclear antibodies (ANAs) serve as an important diagnostic screening test. OBJECTIVE: To disclose interference of anti-proteasome antibodies with known ANA patterns. METHODS: Anti-proteasome antibodies were isolated for comprehensive immunofluorescence analyses. The immunofluorescence pattern of human anti-proteaso

Trifluoperazine inhibition of fibrinogen receptor redistribution in surface activated platelets: correlative video-enhanced differential interference contrast light microscopic, high voltage electron microscopic and scanning electron microscopic studies.

Video-enhanced differential interference contrast (VDIC) light microscopy in conjunction with fibrinogen labelled colloidal gold was employed as a probe to follow the mobility of the fibrinogen receptor on platelets. Correlative studies by both high voltage and scanning electron microscopy confirms localization of labels relative to platelet ultrastructural and surface characteristics, respectively. Treatment of platelets with trifluoperazine prior to and after incubation with fibrinogen-gold labels results in a concentration dependent inhibition of receptor movement. The results obtained from

Expansion of calcium microdomains regulates fast exocytosis at a ribbon synapse.

We investigated the Ca2+ signal regulating fast exocytosis at the ribbon synapse of retinal bipolar cells by using total internal reflection fluorescence microscopy to image fluorescent Ca2+ indicators and interference reflection microscopy to monitor exocytosis. Depolarization generated Ca2+ "microdomains" that expanded over the time scale during which the rapidly releasable pool (RRP) of vesicles was released (<40 ms). Replacing mobile Ca2+ buffers in the terminal with 10 mM EGTA prevented expansion of microdomains and decreased the number of rapidly releasable vesicles by a factor of 2. Con

Structural basis for the inhibition of IAPP fibril formation by the co-chaperonin prefoldin.

Chaperones, as modulators of protein conformational states, are key cellular actors to prevent the accumulation of fibrillar aggregates. Here, we integrated kinetic investigations with structural studies to elucidate how the ubiquitous co-chaperonin prefoldin inhibits diabetes associated islet amyloid polypeptide (IAPP) fibril formation. We demonstrated that both human and archaeal prefoldin interfere similarly with the IAPP fibril elongation and secondary nucleation pathways. Using archaeal prefoldin model, we combined nuclear magnetic resonance spectroscopy with electron microscopy to establ

TIM-4 interference in Kupffer cells against CCL4-induced liver fibrosis by mediating Akt1/Mitophagy signalling pathway.

OBJECTIVES: T-cell immunoglobulin domain and mucin domain-4 (TIM-4) is selectively expressed on antigen-presenting cells (APCs) and modulates various immune responses. However, the role of TIM-4 expressed by Kupffer cells (KCs) in liver fibrosis remains unclear. The present study aimed to explore whether and how TIM-4 expressed by KCs is involved in liver fibrosis. MATERIALS AND METHODS: Mice chronic liver fibrosis models were established and divided into the olive-induced control group, CCL4-induced control group, olive-induced TIM-4 interference group and CCL4-induced TIM-4 interference gro

A New Isospora (Apicomplexa: Eimeriidae) from the Summer Tanager, Piranga rubra (Aves: Passeriformes: Cardinalidae), in Oklahoma.

The summer tanager, Piranga rubra (L., 1758) is a medium-sized songbird formerly belonging to the tanager family Thraupidae but now has been placed within the family Cardinalidae. Nothing is known about the coccidian parasites of this stunningly colorful bird. Feces from 2 P. rubra found dead in McCurtain County, Oklahoma were collected and examined for coccidia; 1 was found to be passing a new species of Isospora. Oocysts of Isospora mccurtainensis n. sp. are subspheroidal to ovoidal with a smooth bilayered wall, measure (length × width [L × W]) 21.7 × 19.5 μm, and have a L/W ratio of 1.1; a

The presence of androgen receptors in the epididymis and prostate of the stallion and cryptorchid horse--a preliminary study.

Distribution of androgen receptors (ARs) in the epididymal duct and prostate of three entire stallions and one bilaterally cryptorchid horse was studied immunohistochemically using a polyclonal rabbit antiserum against the ARs. In both the healthy stallions and the cryptorchid, the epithelial cells of the epididymides showed nuclear staining for ARs. The intensity of AR-staining in the principal cells of the epididymis was stronger than that of the basal cells. In the prostate, the glandular secretory cells were moderately stained whereas the basal cells expressed weak AR-staining. Immunostain

Theoretical study of multispectral structured illumination for depth resolved imaging of non-stationary objects: focus on retinal imaging.

Current implementations of structured illumination microscopy for depth-resolved (three-dimensional) imaging have limitations that restrict its use; specifically, they are not applicable to non-stationary objects imaged with relatively poor condenser optics and in non-fluorescent mode. This includes in-vivo retinal imaging. A novel implementation of structured illumination microscopy is presented that overcomes these issues. A three-wavelength illumination technique is used to obtain the three sub-images required for structured illumination simultaneously rather than sequentially, enabling use

Detection of Trimethylamine Based on a Manganese Tetraphenylporphyrin Optical Waveguide Sensing Element.

The sensitive detection of trimethylamine has been accomplished by using a homogeneous optical waveguide sensor system. Also the sensor can be easily fabricated by using tetraphenylporphyrin manganese (MnTPP) as sensitive materials to detect different volatile organic compounds (VOC). NMR (1H-NMR), field-emission scanning electron microscopy (FESEM), and X-ray photoelectron spectroscopy (XPS), infrared (IR), and ultraviolet-visible (UV-vis) instrumental means were used to characterize its structure. Gas-sensing measurements indicated that the sensing element has shown good selectivity, high se

Human Metapneumovirus Is Capable of Entering Cells by Fusion with Endosomal Membranes.

Human metapneumovirus (HMPV), a member of the Paramyxoviridae family, is a leading cause of lower respiratory illness. Although receptor binding is thought to initiate fusion at the plasma membrane for paramyxoviruses, the entry mechanism for HMPV is largely uncharacterized. Here we sought to determine whether HMPV initiates fusion at the plasma membrane or following internalization. To study the HMPV entry process in human bronchial epithelial (BEAS-2B) cells, we used fluorescence microscopy, an R18-dequenching fusion assay, and developed a quantitative, fluorescence microscopy assay to follo

Artemia is capable of spreading oocysts of Cryptosporidium and the cysts of Giardia.

The capability for ingesting and spreading the fixed oocysts of Cryptosporidium and fixed cysts of Giardia, two waterborne protozoan parasites, by Artemia franciscana, a microcrustacean widely used as live diet in fish and shellfish larviculture, was demonstrated using differential interference contrast and immunofluorescence microscopy. Our findings suggest the possibility that this microcrustacean could serve as a disseminating vehicle of both parasites in aquatic environments.

Splenic function in children with hemoglobin SC disease and sickle beta-thalassemia.

Splenic function in children with hemoglobin SC disease and sickle β-thalassemia was evaluated using direct interference contrast microscopy. Children with both disorders demonstrated significant degrees of hyposplenism as evidenced by increased numbers of erythrocytes containing surface "pits." Patients with these variants of sickle cell disease frequently exhibit splenic dysfunction and may be at increased risk to develop the life-threatening septicemias associated with hyposplenism.
