The antileishmanial activity of the antarctic brown alga Ascoseira mirabilis Skottsberg.

Leishmaniasis is a group of diseases that have limited and high toxic therapeutic options. Herein, we evaluated the antileishmanial potential and cytotoxicity of hexanic extract obtained from the Antarctic brown alga Ascoseira mirabilis using bioguided fractionation against Leishmania amazonensis and murine macrophages, which was fractionated by SPE, yielding seven fractions (F1-F7). The fraction F6 showed good anti-amastigote activity (IC50 = 73.4 ± 0.4 μg mL-1) and low cytotoxicity (CC50 > 100 μg mL-1). Thus, in order to identify the bioactive constituent(s) of F6, the fraction was separated…

Intracranial Abscess and Proteus mirabilis: A Case Report and Literature Review.

An intracranial abscess caused by Proteus mirabilis is rarely reported in adults. A 17-year-old girl presented with generalized tonic-clonic seizure, high-grade fever, headache, and vomiting with a history of slowly progressing apathy, clumsiness, and disorientation. She had meningeal signs and altered sensorium with a Glasgow Coma Scale of 10. The laboratory analysis revealed leukocytosis and elevated erythrocyte sedimentation rate. Brain computed tomography (CT) revealed a cystic lesion in the left temporal lobe with perilesional edema and a slight midline shift. She was commenced on empiric…

Elucidating the genetic basis of crystalline biofilm formation in Proteus mirabilis.

Proteus mirabilis forms extensive crystalline biofilms on urethral catheters that occlude urine flow and frequently complicate the management of long-term-catheterized patients. Here, using random transposon mutagenesis in conjunction with in vitro models of the catheterized urinary tract, we elucidate the mechanisms underpinning the formation of crystalline biofilms by P. mirabilis. Mutants identified as defective in blockage of urethral catheters had disruptions in genes involved in nitrogen metabolism and efflux systems but were unaffected in general growth, survival in bladder model system…

Alternate pathway activation of complement in a Proteus mirabilis ulceration of the cornea.

A 63-year-old patient had an extensive infiltration, ulceration, and eventual perforation of the cornea caused by Proteus mirabilis. Histopathological examination of corneal tissue that was obtained at keratoplasty disclosed that the stroma was ulcerated with a diffuse infiltration of polymorphonuclear leukocytes. Immunopathological examination of the cornea revealed prominent and diffuse staining for properdin and C3 complement. There was an absence of staining for IgG, IgA, IgM, IgE, and C4 complement. These findings suggest that the alternate pathway of complement was activated in this ulce…

The effect of fluoroquinolones and antioxidans on biofilm formation by Proteus mirabilis strains.

BACKGROUND: Fluoroquinolones are a group of antibiotics used in urinary tract infections. Unfortunately, resistance to this group of drugs is currently growing. The combined action of fluoroquinolones and other antibacterial and anti-biofilm substances may extend the use of this therapeutic option by clinicians. The aim of the study was to determine the effect of selected fluoroquinolones and therapeutic concentrations of ascorbic acid and rutoside on biofilm formation by Proteus mirabilis. MATERIALS AND METHODS: The study included 15 strains of P. mirabilis isolated from urinary tract infect…

Molecular epidemiology of Proteus mirabilis infections of the catheterized urinary tract.

Proteus mirabilis compromises the care of many patients undergoing long-term indwelling bladder catheterization. It forms crystalline bacterial biofilms in catheters which block the flow of urine, causing either incontinence due to leakage or painful distention of the bladder due to urinary retention. If it is not dealt with, catheter blockage can lead to pyelonephritis and septicemia. We have examined the epidemiology of catheter-associated P. mirabilis infections by use of pulsed-field gel electrophoresis (PFGE) of NotI restriction enzyme digests of bacterial DNA. This technique was shown to…

A lytic phage targeting virulent Proteus mirabilis for effective biofilm eradication.

Proteus mirabilis is an opportunistic pathogen frequently associated with catheter-associated urinary tract infections (CAUTIs), where robust crystalline biofilms contribute to chronicity, antimicrobial tolerance, and recurrence. The rising prevalence of multidrug-resistant (MDR) P. mirabilis has prompted interest in bacteriophage therapy as an alternative to conventional antibiotics. In this study, we isolated a lytic bacteriophage, vB_PmiM_ZX7, from sewage using a highly virulent clinical strain, P. mirabilis YV2, as the host. vB_PmiM_ZX7 displayed a broad host range among MDR P. mirabilis i…

Selective sugar transport supports Proteus mirabilis fitness in the urinary tract.

Proteus mirabilis is a leading cause of complicated urinary tract infections (UTIs). Prior work showed P. mirabilis metabolizes sugars during experimental UTI, yet the role of sugar import systems in pathogenesis remains poorly defined. To investigate this, we generated a panel of 47 targeted mutants in predicted sugar transporter genes and assessed their growth in vitro and fitness in vivo. Growth screening in nutrient-rich and minimal media revealed carbon source-dependent defects in several phosphotransferase system (PTS) mutants, including ptsH and ptsI. Pooled insertion sequencing (In-seq…

Lautropia mirabilis: An Exceedingly Rare Cause of Peritoneal Dialysis-Associated Peritonitis.

Lautropia mirabilis is a gram-negative coccoid bacterium isolated from oral and upper respiratory sites with unclear pathogenic potential. We present an exceedingly rare case of peritoneal dialysis-associated peritonitis due to L. mirabilis in a patient with a recent history of periodontal infection, successfully treated with intraperitoneal antibiotics. We propose that clinicians consider this organism as a potential cause of illness.

A novel functional class 2 integron in clinical Proteus mirabilis isolates.

OBJECTIVES: To describe a novel functional class 2 integron that was found in clinical Proteus mirabilis isolates. METHODS: Class 1 and 2 integrons were screened by PCR in 153 clinical Proteus isolates. The variable regions of class 1 and 2 integrons were determined by restriction analysis and sequencing. The mutations of internal stop codons in class 2 integrons and their common promoters were also determined by sequencing. Enterobacterial repetitive intergenic consensus (ERIC)-PCR was used to analyse the phylogenetic relations of class 2 integron-positive P. mirabilis isolates. RESULTS: Cl…

Tamm-horsfall protein protects against urinary tract infection by proteus mirabilis.

PURPOSE: Proteus mirabilis is a common cause of urinary tract infection. We determined the role of Tamm-Horsfall protein as a host defense factor against the cystitis and pyelonephritis caused by P. mirabilis. MATERIALS AND METHODS: We generated Tamm-Horsfall protein gene knockout mice using homologous recombination. We introduced P. mirabilis transurethrally into the bladder of Tamm-Horsfall protein deficient (THP(-/-)) and genetically similar WT (THP(+/+)) mice. We cultured urine to quantitate the degree of bacteriuria. We examined bladders and kidneys grossly and histomorphometrically to d…

The Podophage PM16 Enhances the Humoral Immune Response Against Proteus mirabilis.

Considering the therapeutic potential of the Proteus mirabilis PM16 podophage, the interaction between PM16, its host strain, and the mouse immune system was investigated. We evaluated how pre-existing humoral immunity to PM16 influences the immune response against P. mirabilis and the neutralization of the phage itself. Balb/c mice were divided into three groups and immunized two times with (1) 0.9% NaCl, (2) adjuvants, or (3) a mixture of PM16 and an adjuvant. Then, each group was subdivided into three subgroups: mock infection, infection with P. mirabilis, and infection with P. mirabilis fo…

Detection of blaOXA-23-Positive Proteus mirabilis Isolate, United States, 2024.

We report detection of a blaOXA-23-positive Proteus mirabilis clinical isolate through Antimicrobial Resistance Laboratory Network testing that is closely related to OXA-23 producers identified in Europe. Combined with similar reports internationally, this finding suggests that Enterobacterales might serve as a silent reservoir of carbapenemase genes commonly associated with Acinetobacter species.

[Proteus mirabilis isolated from urine, resistance to antibiotics and biofilm formation].

BACKGROUND: In the rather rare urinary pathogen Proteus mirabilis resistance to antibiotics and ability to form biofilm were studied. MATERIAL AND METHODS: The strains Proteus mirabilis were isolated from urine samples from ambulatory and hospitalized patients diagnosed with urinary tract infection (UTI) between April 2008 and April 2010. Resistance to antibiotics was investigated using a disk diffusion antimicrobial susceptibility test. Biofilm formation was demonstrated by modified Christensen method. RESULTS: Two hundred and thirteen of P. mirabilis strains were tested. Eighty-two (38.5 %…

Hemagglutinin, urease, and hemolysin production by Proteus mirabilis from clinical sources.

Proteus mirabilis, a common cause of urinary tract infection, can lead to serious complications including pyelonephritis. Adherence factors, urease, and hemolysin may be virulence determinants. These factors were compared for bacteria cultured from 16 patients with acute pyelonephritis and 35 with catheter-associated bacteriuria and for 20 fecal isolates. Pyelonephritis isolates were more likely (P less than .05) to express the mannose-resistant/Proteus-like (MR/P) hemagglutinin in the absence of mannose-resistant/Klebsiella-like (MR/K) hemagglutinin than were catheter-associated or fecal isol…

A case of infective endocarditis following urinary tract infection by Proteus mirabilis.

Infective endocarditis (IE) is most commonly caused by gram-positive cocci, whereas that caused by gram-negative rods, particularly Enterobacteriaceae, is uncommon. In particular, only a few dozen cases of IE caused by Proteus mirabilis have been reported globally. This report describes a 93-year-old woman who developed calculous pyelonephritis, renal impairment, and P. mirabilis bacteremia. She presented with multiple cerebral infarcts on cranial imaging after her consciousness was impaired. Transthoracic echocardiography revealed mobile vegetation on the mitral valve, leading to a diagnosis …

Structure and Assembly of the Proteus mirabilis Flagellar Motor by Cryo-Electron Tomography.

Proteus mirabilis is a Gram-negative Gammaproteobacterium and a major causative agent of urinary tract infections in humans. It is characterized by its ability to switch between swimming motility in liquid media and swarming on solid surfaces. Here, we used cryo-electron tomography and subtomogram averaging to reveal the structure of the flagellar motor of P. mirabilis at nanometer resolution in intact cells. We found that P. mirabilis has a motor that is structurally similar to those of Escherichia coli and Salmonella enterica, lacking the periplasmic elaborations that characterize other more…

Clinical relevance of Proteus mirabilis in hospital patients: a two year survey.

A retrospective study was performed on 1072 non-duplicate isolates of Proteus mirabilis, taken in the period April 1996 to March 1998, and on 100 patient charts randomly selected during the same period. P. mirabilis isolates accounted for 7.7% of Enterobacteriaceae. The isolates were predominantly from urine (70.2%); of the total, 38.0% were penicillinase-producing isolates, 6.9% were extended-spectrum beta-lactamase (ESBL)-producing isolates and 3.6% produced inhibitor-resistant beta-lactamase (IRB). ESBL-producing isolates were observed in long-stay and intensive care and IRB-producing isola…
