Contractile proteins in retinal endothelium and other non-muscle tissues of the eye.

The present study is the first investigation to demonstrate, by employing the combined approach of immunologically and electron microscope methods, the presence of actin-like contractile proteins in the mammalian retina, the corneal epithelium and endothelium, the iris, and the ciliary body, and to confirm their presence in lens epithelium. This is also the first report to demonstrate by these methods the presence of microfilaments and intermediate filaments in retinal vascular endothelium. Since we have shown that actin filaments are especially abundant in immature retinal endothelial cells,

Regulation of myonuclear positioning and muscle function by the skeletal muscle-specific CIP protein.

The appropriate arrangement of myonuclei within skeletal muscle myofibers is of critical importance for normal muscle function, and improper myonuclear localization has been linked to a variety of skeletal muscle diseases, such as centronuclear myopathy and muscular dystrophies. However, the molecules that govern myonuclear positioning remain elusive. Here, we report that skeletal muscle-specific CIP (sk-CIP) is a regulator of nuclear positioning. Genetic deletion of sk-CIP in mice results in misalignment of myonuclei along the myofibers and at specialized structures such as neuromuscular junc

Protein interactome of muscle invasive bladder cancer.

Muscle invasive bladder carcinoma is a complex, multifactorial disease caused by disruptions and alterations of several molecular pathways that result in heterogeneous phenotypes and variable disease outcome. Combining this disparate knowledge may offer insights for deciphering relevant molecular processes regarding targeted therapeutic approaches guided by molecular signatures allowing improved phenotype profiling. The aim of the study is to characterize muscle invasive bladder carcinoma on a molecular level by incorporating scientific literature screening and signatures from omics profiling.

Androgens and the control of skeletal muscle protein synthesis.

Athletes have long supported the concept that anabolic steroids increase skeletal muscle mass. However, it was only recently that both testosterone and its synthetic analogue, oxandrolone, were proven capable of inducing myotrophic effects in postabsorptive human skeletal muscle. These findings have provided the physiological evidence that anabolic steroids deserve attention in the clinical arena as a pharmacological intervention against losses in lean body mass associated with age, disease, trauma and burn injury. However, we are lacking in vivo molecular evidence that would directly or indir

Effects of temperature on protein phosphorylation in postmortem muscle.

BACKGROUND: Phosphorylation is one of the most important post-translational modifications. Currently, many postmortem protein phosphorylation studies in muscle have been related to meat quality such as tenderness and color stability. However, the effects of various storage temperatures (25, 15, 4 and -1.5 °C) on the phosphorylation level of protein are poorly understood. Changes in the protein phosphorylation levels in postmortem ovine muscle at various storage temperatures were determined in this study. RESULTS: The obtained data showed that pH decline rate was significantly inhibited at -1.

N-Carbamylglutamate Improves Production Performance and Muscle Growth by Regulating Protein Digestive Function and Muscle Protein Synthesis in Broiler Chickens.

N-carbamylglutamate (NCG) is a structural analog of N-acetylglutamate and has multifunctional roles in animal production. However, few studies have been conducted to evaluate the effects of NCG on protein digestive function and muscle growth in broiler chickens. This study investigated whether NCG could improve muscle growth via protein digestive ability and amino acid metabolism in broiler chickens. A total of 144 one-day-old male broiler chickens were randomly allocated to four treatments with six replicates of 6 broiler chickens each. The treatments were a basal diet and a basal diet with N

Muscle protein turnover and low-protein diets in patients with chronic kidney disease.

Adaptation to a low-protein diet (LPD) involves a reduction in the rate of amino acid (AA) flux and oxidation, leading to more efficient use of dietary AA and reduced ureagenesis. Of note, the concept of 'adaptation' to low-protein intakes has been separated from the concept of 'accommodation', the latter term implying a decrease in protein synthesis, with development of wasting, when dietary protein intake becomes inadequate, i.e. beyond the limits of the adaptive mechanisms. Acidosis, insulin resistance and inflammation are recognized mechanisms that can increase protein degradation and can

Mutant Muscle LIM Protein C58G causes cardiomyopathy through protein depletion.

Cysteine and glycine rich protein 3 (CSRP3) encodes Muscle LIM Protein (MLP), a well-established disease gene for Hypertrophic Cardiomyopathy (HCM). MLP, in contrast to the proteins encoded by the other recognised HCM disease genes, is non-sarcomeric, and has important signalling functions in cardiomyocytes. To gain insight into the disease mechanisms involved, we generated a knock-in mouse (KI) model, carrying the well documented HCM-causing CSRP3 mutation C58G. In vivo phenotyping of homozygous KI/KI mice revealed a robust cardiomyopathy phenotype with diastolic and systolic left ventricular

Cytoplasmic mRNA-protein complexes of chicken muscle cells and their role in protein synthesis.

Irradiation of chicken muscle cells with ultraviolet light (254 nm) to cross-link RNA and protein moieties was used to examine the polypeptide complements of cytoplasmic mRNA-protein complexes (mRNP). The polypeptides of translationally active mRNP complexes released from polysomes were compared to the repressed nonpolysomal cytoplasmic (free) mRNP complexes. In general, all of the polypeptides present in free mRNPs were also found in the polysomal mRNPs. In contrast to polysomal mRNPS, polypeptides of Mr 28 000, 32 000, 46 000, 65 000 and 150 000 were either absent or present in relatively sm

A 63 kDa skeletal muscle protein associated with eye muscle inflammation in Graves' disease is identified as the calcium binding protein calsequestrin.

It is generally accepted that thyroid-associated ophthalmopathy (TAO) is an autoimmune disease of the eye muscle (EM) and the surrounding orbital connective tissue in which circulating antibodies play an important role. Antibodies against EM membrane proteins of 63-67kDa mol. wt. seem to be the best markers of ophthalmopathy in patients with autoimmune thyroid disease. We purified a 63 kDa EM protein using SDS-polyacrylamide gel electrophoresis technology and TAO patients' sera as probes, digested the protein with cyanogen bromide and sequenced immunoreactive peptides. We also screened a human

Proximity Ligation Assay to Determine Protein-Protein Interactions in Muscle Stem Cells.

Satellite cells are present only in skeletal muscle and are crucial for regeneration. In recent years, satellite cell heterogeneity has identified different subpopulations, such as bona fide muscle stem cells. These populations are rare and in small numbers, making studying them very difficult. Moreover, satellite cells are sensitive to the environment and niche. To avoid niche disruption and changes to the cell state, our lab developed a mix of different techniques to study protein-protein interactions ex vivo on cultured myofibers. This protocol is based on the Proximity Ligation Assay (PLA)

Gene and protein expression associated with protein synthesis and breakdown in paraplegic skeletal muscle.

Spinal cord injury reduces the rate of skeletal muscle protein synthesis and increases protein breakdown, resulting in rapid muscle loss. The purpose of this study was to determine whether long-term paraplegia would eventually result in a downregulation of muscle mRNA and protein expression associated with both protein synthesis and breakdown. After 10 weeks of spinal cord transection, soleus muscle from 12 rats (6 sham-control, 6 paraplegic) was studied for mRNAs and proteins associated with protein synthesis and breakdown using real-time polymerase chain reaction and immunoblotting technique

[Protein metabolism in vitamin A deficient rats. II. Protein synthesis in striated muscle].

Parameters of skeletal muscle protein synthesis was studied in vivo and in vitro. The experimental design was previously described. Muscle humidity, protein and DNA content are not modified by vitamin A deficiency. RNA and free amino acids are modified. Glutamine, serine and threonine level decrease while phenylalanine and leucine increase in vitamin A deficients. After a single intra-peritoneal injection of (14C) leucine, the incorporation of radioactivity into total ribosomes and postribosomal supernatant protein was studied. The rats were killed 20, 40, 60 minutes and 2,6,10 days after inje

Expression of apoptosis related proteins in normal and diseased muscle: a possible role for Bcl-2 in protection of striated muscle.

The unique absence of major histocompatibility complex class I antigen (MHC-I) expression in normal muscle is one possible mechanism protecting striated muscle. In order to define their possible involvement in protection of normal muscle, we investigated the expression of molecules involved in muscle fibre death and survival mechanisms (Bcl-2, Fas, Fas-ligand and TRAIL), focusing on disorders with possible involvement of cytotoxic T cells. We studied muscle biopsies from 20 healthy volunteers, from 10 patients affected by polymyositis and 10 by Duchenne muscular dystrophy. By using immunohisto

Degradative activity of granzyme A on skeletal muscle proteins in vitro: a possible molecular mechanism for muscle fiber damage in polymyositis.

The importance of cytotoxic T lymphocytes (CTLs) in the autoimmune inflammatory myopathies, especially polymyositis (PM), has been emphasized. We have studied the degradative activity of granzyme A, a cytotoxic molecule with trypsin-like specificity in CTL granules, on several muscle proteins in vitro. Our study reveals that granzyme A hydrolyzes dystrophin, myosin, and nebulin, but not laminin, alpha-actinin, vinculin, and connectin in vitro. Among these proteins, nebulin is more susceptible to proteolysis, followed by dystrophin, myosin heavy chain, and myosin light chains, in that order. Th…

Ror2 signaling regulated by differential Wnt proteins determines pathological fate of muscle mesenchymal progenitors.

Skeletal muscle mesenchymal progenitors (MPs) play a critical role in supporting muscle regeneration. However, under pathological conditions, they contribute to intramuscular adipose tissue accumulation, involved in muscle diseases, including muscular dystrophy and sarcopenia, age-related muscular atrophy. How MP fate is determined in these different contexts remains unelucidated. Here, we report that Ror2, a non-canonical Wnt signaling receptor, is selectively expressed in MPs and regulates their pathological features in a differential ligand-dependent manner. We identified Wnt11 and Wnt5b as

Skeletal and Cardiac Muscle Disorders Caused by Mutations in Genes Encoding Intermediate Filament Proteins.

Intermediate filaments are major components of the cytoskeleton. Desmin and synemin, cytoplasmic intermediate filament proteins and A-type lamins, nuclear intermediate filament proteins, play key roles in skeletal and cardiac muscle. Desmin, encoded by the DES gene (OMIM *125660) and A-type lamins by the LMNA gene (OMIM *150330), have been involved in striated muscle disorders. Diseases include desmin-related myopathy and cardiomyopathy (desminopathy), which can be manifested with dilated, restrictive, hypertrophic, arrhythmogenic, or even left ventricular non-compaction cardiomyopathy, Emery-
