C9orf72 in myeloid cells suppresses STING-induced inflammation.

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are neurodegenerative disorders that overlap in their clinical present … n expression in brain and peripheral blood cells4-6. Here we show in mice that loss of C9orf72 from myeloid cells alone is sufficient to recapitulate the age-dependent lymphoid hypertrophy and autoinflammation seen in animals with a complete knockout of C9orf72. Dendritic cells isolated from C9orf72-/- mice show marked early activation of the type I interferon …

Myeloid-Derived Suppressor Cells in Some Oncohematological Diseases

Myeloid-derived suppressor cells are immature myeloid cells with immunosuppressive properties. The review presents characteristics of myeloid-derived suppressor cells. It includes phenotype variants, mechanisms of the suppressive effect on the immune system, and tumor recruitment mechanisms of myeloid suppressors. It provides a brief description of works which studied myeloid suppressor in oncohematological diseases including multiple myeloma, lymphomas, and leukemias.

CpG oligodeoxynucleotide induces bone marrow precursor cells into myeloid-derived suppressor cells.

Dendritic cells (DCs) and myeloid-derived suppressor cells (MDSCs) perform a number of functions in different immunological settings. In standard in vitro experiments, DCs are produced from mouse bone marrow (BM) cells in the presence of granulocyte-macrophage colony-stimulating factor (GM-CSF) and IL-4. Our previous study demonstrated that BM precursor cells could differentiate into MDSCs when co-cultured with poly (I:C). In the present study, BM precursor cells cultured in GM-CSF and IL-4 were treated with CpG oligodeoxynucleotide (CpG ODN). We observed that Gr1+CD11b+ cells exhibiting MDSC

Advancing our understanding of the influence of myeloid-derived suppressor cells in chronic myeloid leukemia.

Chronic myeloid leukemia (CML) is a malignant clonal proliferative disease originating from hematopoietic stem cells. While treatment with tyrosine kinase inhibitors (TKIs) effectively eliminates the majority of leukemia cells in patients, residual leukemia cells can still be detected in those achieving deep molecular remission, ultimately leading to drug resistance or relapse. But the exact mechanism is unclear. In recent years, the immune microenvironment has been a hot research topic in hematologic malignancies. CML patients exhibit abnormalities in antitumor immunity. Myeloid-derived suppr

Loss of phosphatase and tensin homolog (PTEN) in myeloid cells controls inflammatory bone destruction by regulating the osteoclastogenic potential of myeloid cells.

OBJECTIVE: Local bone destruction in rheumatic diseases, which often leads to disability and severely reduced quality of life, is almost exc … USIONS: These data demonstrate that loss of PTEN and, therefore, sustained PI3-Kinase signalling in myeloid cells especially, elevates the osteoclastogenic potential of myeloid cells, leading to enhanced inflammatory local bone destruction. Therefore, although our study allows no direct translational conclusion since we used a conditional knockout approach, the …

CD7 CAR-T cells eliminate chronic myeloid leukemia stem cells specifically and effectively.

OBJECTIVES: Leukemia stem cells (LSCs) in chronic myeloid leukemia (CML) are associated with disease recurrence and progression, cannot be eliminated by tyrosine kinase inhibitors (TKIs), and affect TKI discontinuation in CML patients. Chimeric antigen receptor T cells (CAR-T) therapy can eliminate tumor cells expressing specific antigens. CD7 is highly expressed on LSCs and can distinguish LSCs from normal hematopoietic stem cells (HSCs). This study explored the biological characteristics of CD7, and evaluated the effects of CD7 CAR-T cells on CD7+ CML tumor cells and CD7+ LSCs in vitro. MET

Crosstalk between efferocytic myeloid cells and T-cells and its relevance to atherosclerosis.

The interplay between myeloid cells and T-lymphocytes is critical to the regulation of host defense and inflammation resolution. Dysregulation of this interaction can contribute to the development of chronic inflammatory diseases. Important among these diseases is atherosclerosis, which refers to focal lesions in the arterial intima driven by elevated apolipoprotein B-containing lipoproteins, notably low-density lipoprotein (LDL), and characterized by the formation of a plaque composed of inflammatory immune cells, a collection of dead cells and lipids called the necrotic core, and a fibrous c…

[Calcium ionophore induces the differentiation of chronic myeloid leukemia cells into dendritic cells].

The aim of this study was to investigate the ability of calcium ionophore (CI ) to induce the differentiation of CML cells into dendritic cells (DC), to analyze the P210 expression in DCs and to evaluate the stimulatory effect of CML-DC on production of cytotoxic activity against CML cells via activating the autologous T cells. The mononuclear cells were isolated from bone marrow of CML patients whose WBC counts were more than 30x10(9)/L when samples were collected, then the lymphocytes and monocytes were discarded by pouring out supernatant twice at different culture time point. Myeloid-Derived Suppressor Cells Induce Exhaustion-Like CD8+ T Cells during JEV Infection.

Japanese encephalitis (JE), caused by Japanese encephalitis virus (JEV), is a mosquito-borne zoonotic disease and a leading cause of viral encephalitis worldwide. While JEV has the ability to traverse the blood-brain barrier (BBB), the precise mechanisms by which it inhibits the immune response prior to penetrating the BBB remain unclear, presenting obstacles in the development of efficacious therapeutic interventions. This study investigated the impact of JEV on CD8+ T cell responses, with a particular focus on the dysfunction of CD8+ T cells during JEV infection. Our results demonstrated tha

Clinical significance of plasmacytoid dendritic cells and myeloid-derived suppressor cells in melanoma.

BACKGROUND: Immune markers in the peripheral blood of melanoma patients could provide prognostic information. However, there is currently no consensus on which circulating cell types have more clinical impact. We therefore evaluated myeloid-derived suppressor cells (MDSC), dendritic cells (DC), cytotoxic T-cells and regulatory T-cells (Treg) in a series of blood samples of melanoma patients in different stages of disease. METHODS: Flow cytometry was performed on peripheral blood mononuclear cells of 69 stage I to IV melanoma patients with a median follow-up of 39 months after diagnosis to mea

Recombinant Myeloid Hematopoietic Growth Factors and Clinical Stimulation of Acute Myeloid Leukemia Cells: A Narrative Review.

BACKGROUND: The advent of molecularly cloned hematopoietic growth factors (rhuG-CSF and rhuGM-CSF) enhanced the safety in treating acute myeloid leukemia (AML) by reducing the duration of neutropenia and thus decreasing the risks for infection. However, early in vitro studies demonstrated leukemia cell proliferation in response to these agents, raising reasonable concerns related to whether these factors can cause or contribute to relapse or progression of AML. SUMMARY: Clinical studies using recombinant myeloid hematopoietic growth factors have supported their safety, as there is little or n

Myeloid-derived suppressor cells mitigate inflammation in periodontal disease.

Myeloid-derived suppressor cells (MDSCs) are immature, immunosuppressive myeloid cells whose role in periodontal disease (PD) is not well defined. Here, we show that gingiva from PD patients exhibit increased infiltration of MDSCs, including granulocytic (G-MDSC) and monocytic (M-MDSC) subsets, alongside elevated CD4+ T cells and CD19+ B cells. Functionally, these MDSC subsets suppress autologous CD4+ T cell proliferation in co-culture, confirming their immunoregulatory capacity. In a murine ligature-induced periodontitis (LIP) model, both gingival MDSC subsets increased over time and correlat…

The Roles of Myeloid Cells in Breast Cancer Progression.

This chapter reviews tumor-associated myeloid cells, including macrophages, neutrophils, and other innate immune cells, and their multifaceted roles in supporting breast cancer progression and metastasis. In primary tumors, myeloid cells play key roles in promoting tumor epithelial-mesenchymal transition (EMT) and invasion. They can facilitate intravasation (entry into the bloodstream) and colonization, disrupting the endothelial cell layer and reshaping the extracellular matrix. They can also stimulate angiogenesis, suppress immune cell responses, and enhance cancer cell adaptability. In the …

TREM2-Mediated Myeloid Cells Protect Against Pathological Choroidal Neovascularization.

Choroidal neovascularization (CNV) is a hallmark of neovascular age-related macular degeneration, a leading cause of irreversible vision loss in the elderly. While immune dysregulation and myeloid cell activation have been implicated in CNV pathogenesis, the molecular mechanisms by which myeloid subsets influence NV remain incompletely understood. Triggering receptor expressed on myeloid cells 2 (TREM2) is an immunomodulatory receptor enriched in microglia and tissue macrophages, known to play protective roles in retinal and neurodegenerative diseases. However, its function in CNV has not been…

Myeloid cells as IFNα producers in systemic lupus erythematosus.

Type I interferons (IFNs) play crucial roles in the pathogenesis of systemic lupus erythematosus (SLE). Plasmacytoid dendritic cells (pDCs) stimulated by Toll-like receptor (TLR) pathways have been thought to be the major producers of IFNα in patients with SLE. However, the responsiveness of pDCs from SLE patients to stimuli that produce IFNα differs depending on the type of TLR pathway involved. In addition to pDCs, monocytes from SLE patients were found to produce IFNα when responding to the cGAS-STING pathway. Here, we outline the major pathways that induce IFNα production by myeloid cells …

Gilteritinib Reduces FLT3 Expression in Acute Myeloid Leukemia Cells.

Acute myeloid leukemia (AML) is a genetically diverse and challenging malignancy, with mutations in the FLT3 gene being particularly common and deleterious. Gilteritinib, a potent FLT3 inhibitor, has been approved by the U.S. Food and Drug Administration (FDA) for the treatment of relapsed/refractory AML with FLT3 mutations. Although gilteritinib was developed based on its inhibitory activity against FLT3 kinase, it is important to understand the precise mechanisms of its antileukemic activity in managing drug resistance and discovering biomarkers. This study was designed to elucidate the effe
