We present an individual with three distinct malformation complexes, DiGeorge syndrome, CHARGE association and Dandy-Walker malformation. An extensive literature review has shown that DiGeorge syndrome and CHARGE association rarely occur simultaneously. The presence of both these malformation complexes with Dandy-Walker malformation has not been previously reported. These three malformation complexes may all be related by neural crest maldevelopment.

A 5-month-old infant had an enlarged left eye removed for a possible intraocular malignancy. The eye had a thin cornea, a superior staphyloma, a completely cupped optic nerve head and no iris or lens. Histopathologic study showed little corneal or uveal stroma, small rudiments of ciliary body superiorly with no smooth muscle, pigment epithelium and nonpigmented neuroepithelium that lined the entire interior of the globe, focal superior retinal dysplasia and a focal superior scleral thickening containing a melanotic malformation. Except for the absence of the lens and the presence of neuroepith

During vertebrate embryonic development, a population of dorsal neural tube-derived stem cells, termed the neural crest (NC), undergo a series of morphogenetic changes and extensive migration to become a diverse array of cell types. Around the developing eye, this multipotent ocular NC cell population, called the periocular mesenchyme (POM), comprises migratory mesenchymal cells that eventually give rise to many of the elements in the anterior of the eye, such as the cornea, sclera, trabecular meshwork, and iris. Molecular cell biology and genetic analyses of congenital eye diseases have provi

For well over half of the 150 years since the discovery of the neural crest, the special ability of these cells to function as a source of species-specific pattern has been clearly recognized. Initially, this observation arose in association with chimeric transplant experiments among differentially pigmented amphibians, where the neural crest origin for melanocytes had been duly noted. Shortly thereafter, the role of cranial neural crest cells in transmitting species-specific information on size and shape to the pharyngeal arch skeleton as well as in regulating the timing of its differentiatio

Aebp2 is a potential targeting protein for the mammalian Polycomb Repression Complex 2 (PRC2). We generated a mutant mouse line disrupting the transcription of Aebp2 to investigate its in vivo roles. Aebp2-mutant homozygotes were embryonic lethal while heterozygotes survived to adulthood with fertility. In developing mouse embryos, Aebp2 is expressed mainly within cells of neural crest origin. In addition, many heterozygotes display a set of phenotypes, enlarged colon and hypopigmentation, similar to those observed in human patients with Hirschsprung's disease and Waardenburg syndrome. These p

Endothelin receptor B (Ednrb) plays a critical role in the development of melanocytes and neurons and glia of the enteric nervous system. These distinct neural crest-derived cell types express Ednrb and share the property of intercalating into tissues, such as the intestine whose muscle precursor cells also express Ednrb. Such widespread Ednrb expression has been a significant obstacle in establishing precise roles for Ednrb in development. We describe here the production of an Ednrb allele floxed at exon 3 and its use in excising the receptor from mouse neural crest cells by use of Cre-recomb

Three children are described who show a spectrum of clinical abnormalities affecting their eyes and non ocular tissues. These entities are best explained as disorders of neural crest migration and once recognised should lead to a search for other systemic developmental disorders. The mechanism of combined facial skeleton and eye malformation is discussed in the light of our current understanding of ocular embryology, and the justification for considering these and other entities as examples of Neurocristopathies is explored.

Malformations of the anterior segment of the eye can be associated with different systemic findings. The regular combination of aplasia of the permanent central and lateral incisors, typical ocular changes, and umbilical dysmorphology is known as the Rieger syndrome. Aplasia of the permanent incisors can serve as a diagnostic symptom. We report on three siblings who had characteristical symptoms of Rieger syndrome. The tissue involved derives from the neural crest cells. Mesenchymal dysgenesis is discussed as the etiology of this disorder.

By means of the method for formation of quail-hen transplantation chimeras , contribution of the neural crest cells in the eye development has been studied in early avian embryos. In 6-9 day-old chimeras the neural crest cells participate in formation of the eye sclera and ciliary body, and they also form corneal endothelium and stroma.

The thymus is formed by contributions from different sources which must interact in a precisely timed sequence for proper development. Pharyngeal pouch epithelium interacts with embryonic connective tissue in preparation for migration of blood-borne lymphoid stem cells into the thymic primordium. The embryonic connective tissue in this region is derived from neural crest. Ablation of small portions of neural crest prevents or alters the development of the thymus. Other pharyngeal pouch derivatives such as thyroids and parathyroids are affected. Furthermore, defects in the heart and great vesse

Elimination of limited areas of the cephalic neural crest in stage 9 or 10 chick embryos markedly reduced the size of the thymus gland or resulted in its absence. Small thymic lobes contained both thymocytes and epithelial cells but showed delayed development. Parathyroid and thyroid glands sometimes were reduced in size or missing from the normal location on one or both sides. Heart defects were consistently present. Thymus development may depend on direct interaction of mesenchymal derivatives of the neural crest with pharyngeal epithelium. Multiple defects, such as the Di George syndrome, m

The neural crest is a highly migratory and multipotent cell population that contributes to many defining features of vertebrates. As a uniquely vertebrate cell type, the neural crest is an excellent model for studying cell lineage and diversification during embryonic development because of its multipotency, motility, and capacity to form a plethora of derivatives. Neural crest cells migrate extensively throughout the body and contribute to many of the defining features of vertebrate embryos, including the craniofacial skeleton, most of the peripheral nervous system and pigmentation of the skin

In cardiac myxomas, the malignant transformation process, selecting incidental gene mutations and leading to loss of proliferation control, has not a so drastic effects in terms of growth rate of tumor mass, but frequently the particular location of lesion engrosses the high risk for health. For accurate cancer cell profiling, it is important to establish the embryologic origin of malignant cells and their initial commitments, above all, in the sight of therapeutic strategies and solutions. Here, we advance, for cardiac myxoma, the hypothesis of an origin from cardiac neural crest cells and we

Understanding the molecular mechanisms of stem cell maintenance is crucial for the ultimate goal of manipulating stem cells for the treatment of disease. Foxd3 is required early in mouse embryogenesis; Foxd3(-/-) embryos fail around the time of implantation, cells of the inner cell mass cannot be maintained in vitro, and blastocyst-derived stem cell lines cannot be established. Here, we report that Foxd3 is required for maintenance of the multipotent mammalian neural crest. Using tissue-specific deletion of Foxd3 in the neural crest, we show that Foxd3(flox/-); Wnt1-Cre mice die perinatally wi

The head-trunk interface lies at the occipito-cervical boundary, which corresponds to the somite 5/6 level. Previous studies have demonstrated that neural crest cells also behave differently either side of this boundary and that this may be due to intrinsic differences between cranial and trunk crest. However, it is also possible that some of the observed differences between cranial and trunk crest are assigned by environmental cues. We have therefore scrutinised the behaviour of the neural crest cells generated either side of the occipito-cervical boundary in chick and, interestingly, find th

The neural crest is a transient embryonic structure whose cells migrate extensively before giving rise to a variety of differentiated cell types. Both intrinsic cell lineage information and environmental cues are thought to play a role in determining the fate of these cells. Early in development, these cells can be divided into distinct populations based on their axial level of origin. Cranial neural crest cells differentiate into facial cartilage and cranial ganglia, whereas vagal crest cells give rise to the enteric ganglia. Trunk neural crest cells normally give rise to melanocytes, neurons

The peripheral nervous system (PNS) is derived from a transitory embryonic structure, the neural crest with an additional contribution of the cephalic ectodermal placodes to the sensory ganglia of certain cranial nerves. In vivo grafting experiments, taking advantage of the quail/chick marker system, allowed the fate map of the neural crest to be constructed and the precise level of origin along the neuraxis of the component cells of PNS ganglia to be defined. It appeared that the neural crest is regionalized in different areas from which a limited number of phenotypes arise. However, if the p
