A closer look to neural pathways and psychopharmacology of obsessive compulsive disorder.

The intricate neural pathways involved in obsessive-compulsive disorder (OCD) affect areas of our brain that control executive functioning, organization, and planning. OCD is a chronic condition that can be debilitating, afflicting millions of people worldwide. The lifetime prevalence of OCD in the US is 2.3%. OCD is predominantly characterized by obsessions consisting of intrusive and unwanted thoughts, often with impulses that are strongly associated with anxiety. Compulsions with OCD encompass repetitive behaviors or mental acts to satisfy their afflicted obsessions or impulses. While these…

Four-dimensional tractography animates propagations of neural activation via distinct interhemispheric pathways.

OBJECTIVE: To visualize and validate the dynamics of interhemispheric neural propagations induced by single-pulse electrical stimulation (SPES). METHODS: This methodological study included three patients with drug-resistant focal epilepsy who underwent measurement of cortico-cortical spectral responses (CCSRs) during bilateral stereo-electroencephalography recording. We delivered SPES to 83 electrode pairs and analyzed CCSRs recorded at 268 nonepileptic electrode sites. Diffusion-weighted imaging (DWI) tractography localized the interhemispheric white matter pathways as streamlines directly c

Neural pathways involved in infection-induced inflammation: recent insights and clinical implications.

Although the immune and nervous systems have long been considered independent biological systems, they turn out to mingle and interact extensively. The present review summarizes recent insights into the neural pathways activated by and involved in infection-induced inflammation and discusses potential clinical applications. The simplest activation concerns a reflex action within C-fibers leading to neurogenic inflammation. Low concentrations of pro-inflammatory cytokines or bacterial fragments may also act on these afferent nerve fibers to signal the central nervous system and bring about earl…

Two neural pathways of face processing: A critical evaluation of current models.

The neural basis of face processing has been extensively studied in the past two decades. The current dominant neural model proposed by Haxby et al. (2000); Gobbini and Haxby (2007) suggests a division of labor between the fusiform face area (FFA), which processes invariant facial aspects, such as identity, and the posterior superior temporal sulcus (pSTS), which processes changeable facial aspects, such as expression. An extension to this model for the processing of dynamic faces proposed by O'Toole et al. (2002) highlights the role of the pSTS in the processing of identity from dynamic famil

Cortical hubs and subcortical cholinergic pathways as neural substrates of poststroke dementia.

BACKGROUND AND PURPOSE: A role of neural networks in the development of poststroke dementia has not been clearly established. We hypothesized that stroke-mediated disruption of subcortical cholinergic pathway or large-scale neural networks contributes to poststroke dementia. METHODS: A matched case-control study was conducted in a predetermined cohort with acute ischemic stroke. Cases were defined as newly developed dementia diagnosed >3 months after stroke using the Korean Vascular Cognitive Impairment Harmonization Standards. Each case was matched to 2 controls for age, education, and initi

Neural crest cell signaling pathways critical to cranial bone development and pathology.

Neural crest cells appear early during embryogenesis and give rise to many structures in the mature adult. In particular, a specific population of neural crest cells migrates to and populates developing cranial tissues. The ensuing differentiation of these cells via individual complex and often intersecting signaling pathways is indispensible to growth and development of the craniofacial complex. Much research has been devoted to this area of development with particular emphasis on cell signaling events required for physiologic development. Understanding such mechanisms will allow researchers

Optogenetic approaches for investigating neural pathways implicated in schizophrenia and related disorders.

Optogenetic approaches have been rapidly adopted by neuroscientists in order to control the activity of neurons with high temporal, spatial and genetic specificity. By expressing light-sensitive microbial opsins within a genetically-specified population of neurons, flashes of light can be used to activate these opsins and thereby modulate the targeted cells in a spatially and temporally defined manner. Thus, optogenetics can be used to activate very specific sets of neurons or projections at particular times, either within freely behaving animals, or in reduced preparations such as brain slice

Studies of the central neural pathways to the stomach and Zusanli (ST36).

The purpose of this morphological study was to investigate the relation between the meridian, meridian points and viscera using neuroanatomical tracers. The common locations of the spinal cord and brain projecting to the stomach and Zusanli were observed following injection of CTB (cholera toxin B subunit) and pseudorabies viruses (PRV-Ba, Bartha strain and PRV-Ba-Gal, galactosidase insertion) into the stomach and Zusanli (ST36). After 4-5 days of survival following injection into twelve rats, they were perfused, and their spinal cords and brains were frozen sectioned (30 microm). These sectio

Poliovirus spreads from muscle to the central nervous system by neural pathways.

A transgenic mouse model was used to address an unsolved question in the pathogenesis of poliomyelitis: how poliovirus invades the central nervous system (CNS). LD50 values for intramuscular and intracerebral inoculation of poliovirus in transgenic mice expressing poliovirus receptors (TgPVR mice) were similar. After intramuscular inoculation with poliovirus, paralysis was observed first in the inoculated limb. In contrast, localization of initial paralysis to the inoculated limb was not observed in normal mice inoculated intramuscularly with the mouse-adapted P2/Lansing poliovirus strain. Aft

Inhibitory neural pathway regulating gastric emptying in rats.

The relaxation of the pylorus is one of the most important factors for promoting gastric emptying. However, the role of inhibitory neurotransmitters in the regulation of pyloric relaxation and gastric emptying remains unclear. In this study, we investigated the effects of NO biosynthesis inhibitor, N(G)-nitro-L-arginine methyl ester (L-NAME), and calcium dependent potassium channel blocker, apamin, on vagal stimulation-induced pyloric relaxation and gastric emptying in rats. Sodium nitroprusside (SNP), adenosine 5'-triphosphate (ATP), vasoactive intestinal polypeptide (VIP) and pituitary adeny

Olfactory neural pathway in mouse hepatitis virus nasoencephalitis.

The mechanism of brain infection with mouse hepatitis virus-JHM was studied in BALB/cByJ mice following intranasal inoculation, and found to be a consequence of direct viral spread along olfactory nerves into olfactory bulbs of the brain. Infection was followed sequentially from nose to brain, using microscopy, immunohistochemistry and virus quantification. Lesions, antigen and virus were observed in the olfactory bulb and anterior brain as early as 2 days and posterior brain by 4 days after inoculation. Viral antigen extended through nasal mucosa into submucosa, then coursed along the olfacto

Human assembloid model of the ascending neural sensory pathway.

Somatosensory pathways convey crucial information about pain, touch, itch and body part movement from peripheral organs to the central nervous system1,2. Despite substantial needs to understand how these pathways assemble and to develop pain therapeutics, clinical translation remains challenging. This is probably related to species-specific features and the lack of in vitro models of the polysynaptic pathway. Here we established a human ascending somatosensory assembloid (hASA), a four-part assembloid generated from human pluripotent stem cells that integrates somatosensory, spinal, thalamic a

Neural Correlates of the Dual-Pathway Model for ADHD in Adolescents.

OBJECTIVE: The dual-pathway model has been proposed to explain the heterogeneity in symptoms of attention deficit hyperactivity disorder (ADHD) by two independent psychological pathways based on distinct brain circuits. The authors sought to test whether the hypothesized cognitive and motivational pathways have separable neural correlates. METHODS: In a longitudinal community-based cohort of 1,963 adolescents, the neuroanatomical correlates of ADHD were identified by a voxel-wise association analysis and then validated using an independent clinical sample (99 never-medicated patients with ADH

Neural inhibition of inflammation: the cholinergic anti-inflammatory pathway.

The innate immune system is activated by infection and injury to release pro-inflammatory cytokines, which activate macrophages and neutrophils and modulate specific cellular responses. The magnitude of the cytokine response is critical, because a deficient response may result in secondary infections, while an excessive response may be more injurious than the original insult. We recently described a neural pathway, termed the "cholinergic anti-inflammatory pathway", that reflexively monitors and adjusts the inflammatory response by inhibiting pro-inflammatory cytokine synthesis. The spleen-heart crosstalk in cardiovascular disease: integrating neural, immune, and secretory pathways.

Emerging evidence links cardiovascular disease (CVD) to organ-to-organ signaling that extends beyond the heart. The spleen is increasingly recognized as an innervated immune organ that rapidly remodels after cardiovascular injury and, in turn, regulates myocardial inflammation, repair, and remodeling. Human studies support clinical relevance through associations between splenic imaging phenotypes and cardiovascular risk or prognosis, while animal studies provide mechanistic support through spleen-targeted perturbations and mapping of immune trafficking and secreted mediators. This review synth

The neural mechanism and pathways underlying postoperative nausea and vomiting: a comprehensive review.

This scoping review aims to synthesize evidence regarding the neural mechanisms and pathways underlying postoperative nausea and vomiting (PONV). We systematically searched the PubMed, Embase and Cochrane Library databases up to 2024. The findings indicate that PONV is a common complications after surgeries involving anaesthesia and can also lead to severe postoperative discomfort. Various surgical procedures and anaesthesia methods can trigger PONV, and the underlying neural mechanisms and signalling pathways are complex. It is currently assumed that these pathways do not exist independently

Temporal and concentration-dependent effects of oestradiol on neural pathways mediating sexual receptivity.

The acceptance of oestradiol signalling through receptors found in the cell membrane, as well as, the nucleus, has provided for a re-examination of the timing and location of the actions of oestradiol on neural circuits mediating sexual receptivity (lordosis). Oestradiol membrane signalling involves the transactivation of metabotrophic glutamate receptors (mGluRs) that transduce steroid information through protein kinase C signalling cascades producing rapid activation of lordosis-regulating circuits. It has been known for some time that oestradiol initially produces an inhibition of the media
