Reversible S-nitrosation of creatine kinase by nitric oxide in adult rat ventricular myocytes.

We have previously demonstrated that the nitric oxide (NO) donor S-nitroso-N-acetylcysteine (SNAC) reversibly decreases the activity of creatine kinase (CK) in an isolated rat heart preparation, markedly suppressing myocardial contractile responsiveness to an inotropic challenge. We wished to further examine the role of exogenous and endogenous sources of NO species on S-nitrosation of CK and subsequent enzyme activity in adult rat ventricular myocytes (ARVM). Two S-nitrosothiol groups were formed in the CK dimer after nitrosation of rabbit skeletal muscle CK in solution. CK inactivation due t…

Construction of a modified Arrhenius equation for predicting drug nitrosation in solid dosage form.

To prevent contamination by nitrosamine drug substance-related impurities (NDSRIs) in solid dosage forms, the modeling of drug nitrosation reactions was investigated. The nitrosation reaction rate of desloratadine (DES) in the physical mixtures (PMs) was measured under various temperature, relative humidity, and alkaline excipient (AE) content conditions. The nitrosation reaction rate increased with increasing relative humidity and temperature. In contrast, an increase in AE content resulted in a decreased nitrosation reaction rate. The relationship between these three conditions and reaction …

Copper-Mediated Nitrosation: 2-Nitrosophenolato Complexes and Their Use in the Synthesis of Heterocycles.

A simple protocol yielding ortho-substituted nitrosophenols from phenols is demonstrated, in the form of copper(II) bis(nitrosophenolato) complexes. The developed methodology was applied to a range of substrates, confirming the role of the copper in both the formation and protection of the challenging 1, 2-substitution pattern. Using polymer supported thiourea, the Cu could be stripped from the complexes and thus enabled the isolation or identification of the uncoordinated ligands and their decomposition products, in yields generally low in line with the intrinsic high reactivity of 2-nitrosop

Mitochondrial Complex I Reversible S-Nitrosation Improves Bioenergetics and Is Protective in Parkinson's Disease.

AIMS: This study was designed to explore the neuroprotective potential of inorganic nitrite as a new therapeutic avenue in Parkinson's disease (PD). RESULTS: Administration of inorganic nitrite ameliorates neuropathology in phylogenetically distinct animal models of PD. Beneficial effects are not confined to prophylactic treatment and also occur if nitrite is administered when the pathogenic cascade is already active. Mechanistically, the effect is mediated by both complex I S-nitrosation, which under nitrite administration is favored over formation of other forms of oxidation, and down-strea…

Systemic Oxidative and Nitrosative Stress in Benign Prostatic Hyperplasia.

Benign prostatic hyperplasia (BPH) is an age-related disorder increasingly linked to chronic inflammation and redox imbalance, yet its systemic oxidative and nitrosative profile remains insufficiently characterized. In this cross-sectional study, fasting serum samples were collected from 47 men with clinically confirmed BPH scheduled for transurethral resection of the prostate and 40 healthy controls. We assessed antioxidant status (thiols, total antioxidant capacity), lipid peroxidation (malondialdehyde, 4-hydroxynonenal), protein nitration (3-nitrotyrosine), glycoxidation markers (Amadori pr

Obesity-associated lysosomal nitrosative stress promotes retinopathy.

BACKGROUND: Obesity and diabetes are major risk factors causing vision loss around the world, yet the molecular mechanisms linking obesity to retinal pathology remain incompletely understood. While chronic inflammation and dysregulated vascular endothelial growth factor (VEGF) signaling are established contributors to retinopathy, their integration within the context of obesity has not been fully elucidated. METHODS: Retinal transcriptomic RNA sequencing datasets from patients with type 2 diabetes mellitus (T2DM) and obese mouse models were retrieved from the GEO database. Meta-analysis was pe

Nitrosative modifications modulate the activity of plant aminoaldehyde dehydrogenase.

Reactive nitrogen species (RNS) are signalling molecules triggering several defence responses during plant stress. The highest importance has nitric oxide (NO), involved in the post-translational modifications, including protein S-nitrosation of target cysteine residues. Aminoaldehyde dehydrogenases (AMADHs) catalyze the NAD(P)+-dependent oxidation of various ω-amino aldehydes to the corresponding amino acids. Here, we focused on the dynamic redox regulation of AMADH activity in pea (Pisum sativum). We used the reversible S-nitrosation approach and studied the effect of NO donors on the activi…

Nitrite Causes Nitrosative Stress to Iron Sulfur Clusters.

Nitric oxide (NO), produced by nitrite reductases or nitric oxide synthases, performs vital roles in signaling and the immune response. Iron sulfur (FeS) clusters are known targets for NO induced degradation, serving as sensors to trigger cellular responses. However, this FeS reactivity is proposed as NO specific, with no demonstrated reactivity toward nitrite, a soluble NO storage molecule. We demonstrate that synthetic FeS clusters supported by various ligands undergo facile nitrosylation by nitrite in the presence of a reductant, evidencing the nitrite reductase reactivity for FeS clusters.

Nature and Implications of Oxidative and Nitrosative Stresses in Autoimmune Hepatitis.

Oxidative and nitrosative stresses can damage cellular membranes, disrupt mitochondrial function, alter gene expression, promote the apoptosis and necrosis of hepatocytes, and increase fibrosis in diverse acute and chronic liver diseases, including autoimmune hepatitis. The objectives of this review are to describe the mechanisms of oxidative and nitrosative stresses in inflammatory liver disease, indicate the pathogenic implications of these stresses in autoimmune hepatitis, and suggest investigational opportunities to develop interventions that counter them. The principal antioxidant defense

The plasma membrane of microaerophilic protists: oxidative and nitrosative stress.

The trans-plasma-membrane electrochemical potential of microaerophilic protists was monitored by the use of voltage-sensitive charged lipophilic fluorophores; of the many available probes, the anionic oxonol dye bis(1,3-dibarbituric acid)-trimethine oxonol [DiBAC(4)(3)] is an example of one which has been successfully employed using fluorescence microscopy, confocal laser-scanning microscopy and flow cytometry. Several microaerophilic protists have been investigated with this dye; these were Giardia intestinalis, Trichomonas vaginalis, Tritrichomonas foetus, Hexamita inflata and Mastigamoeba p

Nicotine-Mediated Redox and Nitrosative Stress in Cardiorenal Health.

Nicotine exposure is increasingly widespread due to the expansion of the use of noncombustible tobacco and nicotine-replacement products; yet nicotine is still being framed as an addictive but relatively benign compound. This review synthesizes clinical and experimental evidence linking nicotine exposure to cardiorenal disease, emphasizing that risks persist after cessation. We discuss here a central novel concept that damage caused by nicotine in tissues is not determined solely by receptor signaling, as nicotine exists in chemical forms capable of strong receptor-independent membrane diffusi

Fundamental Mechanisms of the Cell Death Caused by Nitrosative Stress.

Nitrosative stress, as an important oxygen metabolism disorder, has been shown to be closely associated with cardiovascular diseases, such as myocardial ischemia/reperfusion injury, aortic aneurysm, heart failure, hypertension, and atherosclerosis. Nitrosative stress refers to the joint biochemical reactions of nitric oxide (NO) and superoxide (O2 -) when an oxygen metabolism disorder occurs in the body. The peroxynitrite anion (ONOO-) produced during this process can nitrate several biomolecules, such as proteins, lipids, and DNA, to generate 3-nitrotyrosine (3-NT), which further induces cell

Oxygen and pulmonary vasodilation: The role of oxidative and nitrosative stress.

Respiratory failure complicates up to 2% of live births and contributes significantly to neonatal morbidity and mortality. Under these conditions, supplemental oxygen is required to support oxygen delivery to the brain and other organs, and to prevent hypoxic pulmonary vasoconstriction. However, therapeutic oxygen is also a source of reactive oxygen species that produce oxidative stress, along with multiple intracellular systems that contribute to the production of free radicals in pulmonary endothelium and vascular smooth muscle. These free radicals cause vasoconstriction, act on multiple sit

Variation of genes involved in oxidative and nitrosative stresses in depression.

The dominating hypothesis among numerous hypotheses explaining the pathogenesis of depressive disorders (DD) is the one involving oxidative and nitrosative stress. In this study, we examined the association between single-nucleotide polymorphisms of the genes encoding SOD2 (superoxide dismutase 2), CAT (catalase), GPx4 (glutathione peroxidase 4), NOS1 (nitric oxide synthase 1), NOS2 (nitric oxide synthase 2), and the development of depressive disorders. Our study was carried out on the DNA isolated from peripheral blood collected from 281 depressed patients and 229 controls. Using TaqMan probe

Nitrosative stress and nitrated proteins in trichloroethene-mediated autoimmunity.

Exposure to trichloroethene (TCE), a ubiquitous environmental contaminant, has been linked to a variety of autoimmune diseases (ADs) including SLE, scleroderma and hepatitis. Mechanisms involved in the pathogenesis of ADs are largely unknown. Earlier studies from our laboratory in MRL+/+ mice suggested the contribution of oxidative/nitrosative stress in TCE-induced autoimmunity, and N-acetylcysteine (NAC) supplementation provided protection by attenuating oxidative stress. This study was undertaken to further evaluate the contribution of nitrosative stress in TCE-mediated autoimmunity and to i

Redox active thiol sensors of oxidative and nitrosative stress.

SIGNIFICANCE: The reactivity of the thiol in the side chain of cysteines is exploited by bacterial regulatory proteins that sense and respond to reactive oxygen and nitrogen species. RECENT ADVANCES: Charged residues and helix dipoles diminish the pKa of redox active cysteines, resulting in a thiolate that is stabilized by neighboring polar amino acids. The reaction of peroxides with thiolates generates a sulfenic acid (-SOH) intermediate that often gives rise to a reversible disulfide bond. Peroxide-induced intramolecular and intermolecular disulfides and intermolecular mixed disulfides modu

Evidence for oxidative/nitrosative stress in the pathogenesis of hepatic encephalopathy.

Hepatic encephalopathy (HE) is a serious complication of liver failure. HE manifests as a series of neuropsychiatric and neuromuscular symptoms including personality changes, sleep abnormalities, asterixis and muscle rigidity progressing through stupor to coma. The pathophysiologic basis of HE remains unclear. There is general agreement that ammonia plays a key role. In recent years, it has been suggested that oxidative/nitrosative stress constitutes part of the pathophysiologic cascade in HE. Direct evidence for oxidative/nitrosative stress in the pathogenesis of HE has been demonstrated in e

Effect of combined oxidative and nitrosative stress on Neisseria meningitidis.

Reactive oxygen and nitrogen species are produced by the human immune system in response to infection. Methods to detoxify these reactive species are vital to the survival of human pathogens, such as Neisseria meningitidis, which is the major aetiological agent of bacterial meningitis. Following activation, macrophages produce superoxide (O(2)(-)), hydrogen peroxide (H(2)O(2)) and nitric oxide (NO). The toxicity of O(2)(-), generated using X/Xo (xanthine/xanthine oxidase), and H(2)O(2) was investigated in the presence and absence of the NO donor DEA-NONOate [2-(N,N-diethylamino)-diazenolate-2-
