Biological Evaluation of Molecular Spherical Nucleic Acids: Targeting Tumors via a Hybridization-Based Folate Decoration.

Folate receptors (FRs), membrane-bound proteins that bind specifically to folate with high affinity, are overexpressed by various cancer types and are therefore used as targets for delivery of therapeutic agents. Molecular spherical nucleic acids (MSNAs) are dendritic formulations of oligonucleotides (ONs) that may have advantages over linear parent ONs with respect to delivery properties. Here, we assembled folate-decorated MSNAs, site-specifically radiolabeled them, and then biologically evaluated their effects in mice bearing HCC1954 breast cancer xenograft tumors. The biodistribution of in

Loading of Extracellular Vesicles with Nucleic Acids via Hybridization with Non-Lamellar Liquid Crystalline Lipid Nanoparticles.

The translation of cell-derived extracellular vesicles (EVs) into biogenic gene delivery systems is limited by relatively inefficient loading strategies. In this work, the loading of various nucleic acids into small EVs via their spontaneous hybridization with preloaded non-lamellar liquid crystalline lipid nanoparticles (LCNPs), forming hybrid EVs (HEVs) is described. It is demonstrated that LCNPs undergo pH-dependent structural transitions from inverse hexagonal (HII) phases at pH 5 to more disordered non-lamellar phases, possibly inverse micellar (L2) or sponge (L3) phases, at pH 7.4, which

A novel hybrid platform of g-C3N4 nanosheets /nucleic-acid-stabilized silver nanoclusters for sensing protein.

The fabrication of nanomaterials-based sensing platform has attracted a great deal of interest due to their unique properties. Here, we report a novel hybrid platform of g-C3N4 nanosheets/DNA-stabilized Ag nanoclusters (CNNS/AgNCs) for sensing application. In this platform, the fluorescent AgNCs was synthesized using a pair of double-functional ssDNA sequence as a template, including the aptamer segment against thrombin and C-rich segment for AgNCs. Next, the interaction between the fluorescent Apt-AgNCs and CNNS was investigated. It is verified that DNA-stabilized AgNCs could absorb on the CN

Detection of pseudorabies virus DNA in the inner ear of intranasally infected BALB/c mice with nucleic acid hybridization in situ.

Evidence for the pathogenicity of pseudorabies virus for the auditory and vestibular organs of experimentally infected mice is presented. We demonstrate viral genomes in cells of the peripheral sensory organs, the nerve structures, and the affected areas of the brain in single sections from an entire cranium of an adult mouse. The data were obtained by an in situ hybridization technique adapted for use with fixed, plastic-embedded materials. In contrast to conventional methods which use frozen sections, we were able to analyze cartilaginous and bony materials with high resolution.

Atypical cytomegalovirus inclusions in gastrointestinal biopsy specimens from patients with the acquired immunodeficiency syndrome: diagnostic role of in situ nucleic acid hybridization.

Cytomegalovirus (CMV) infection of the gastrointestinal tract is a common cause of morbidity and mortality in the acquired immunodeficiency syndrome. The proper recognition of CMV-infected cells in gastrointestinal mucosal biopsies is critical so that effective therapy is not delayed, preventing further viral dissemination. Although the pathology criteria for classic CMV inclusions have been well described, the occurrence of morphologically atypical inclusions has been reported but the inclusions are not well characterized. This study prospectively examined the relative frequency of classic an

In situ detection of enteroviral genomes in myocardial cells by nucleic acid hybridization: an approach to the diagnosis of viral heart disease.

We have developed an in situ hybridization assay capable of detecting enteroviral RNA in myocardial cells, using molecularly cloned coxsackievirus B3 cDNA as a diagnostic probe. Because of the high degree of nucleic acid sequence homology among the numerous enteroviral serotypes, including the group A and B coxsackieviruses and the echoviruses, detection of these various agents commonly implicated in human viral heart disease is possible in a single hybridization assay. We demonstrate the considerable potential of this method for an unequivocal diagnosis of enteroviral heart disease as well as

Specific detection and semiquantitation of microorganisms in tissue by nucleic acid hybridization. II. Investigation of synovia from pigs with chronic Erysipelothrix arthritis.

Synovial tissues from animals with chronic Erysipelothrix arthritis were examined by a technique based on RNA-DNA hybridization in an attempt to detect the inducing organism, Erysipelothrix insidiosa (EI). Twelve synovial specimens from 5 animals whose joints lacked culturable EI also lacked EI detectable by this technique. Because the technique is capable of detecting approximately one organism per 50 mammalian cells, it is concluded that no more than this number were present in the involved tissue. Implications of these results for proposed pathogenetic mechanisms in this disorder and in hum

Hybrid Vernier Sensor Based on Fabry-Perot-Assisted Tapered Microfiber for Ultrasensitive Detection of Nucleic Acid Biomarkers.

Ultrasensitive detection of nucleic acid biomarkers is vital for early disease screening and clinical diagnosis. However, conventional tapered microfiber (TMF) interferometers are constrained by limited sensitivity and detection resolution. Here, we present a Fabry-Perot-assisted tapered microfiber (FP-TMF) biosensor that implements a hybrid Vernier architecture exploiting asynchronous phase evolution. In this design, an air-cavity Fabry-Perot interferometer (FPI) serves as a static and dispersion-stable reference, with its frequency intentionally detuned from that of the highly dispersive TMF

Employing a cluster transfer strategy in a hybridisation chain reaction-silver nanocluster hybrid sensor for nucleic acid detection.

The hybridisation chain reaction (HCR) is a robust isothermal amplification technique widely used for nucleic acid detection, often paired with silver nanoclusters (AgNCs) for fluorescence-based readouts. However, conventional HCR-AgNC assays are prone to circuit leakage and unpredictable AgNC formation, which can compromise assay reliability. In this study, we present a modular HCR-AgNC hybrid sensor employing a cluster transfer strategy, in which pre-formed AgNCs are introduced separately into the HCR system. This design minimises interference between AgNCs and HCR components, enhancing sign

Hybrid plasmonic fiber optic sensor enabling rapid and amplification-free nucleic acid determination for disease screening.

A tilted fiber Bragg grating-surface plasmon resonance biosensing platform is reported for rapid, amplification-free determination of viral nucleic acids in complex clinical matrices. The transducer integrates Au nanoparticles, Ta2C-MXene, and an Au film into a hybrid plasmonic architecture that creates a stereoscopic refractive index sensing region through localized gap-plasmon coupling. Finite element modeling reveals that the three-dimensional electromagnetic field extends the effective sensing volume beyond the classical near-field limit. The field also generates strong gradients that indu

Framework Nucleic Acids Combined with 3D Hybridization Chain Reaction Amplifiers for Monitoring Multiple Human Tear Cytokines.

Existing tear sensors are difficult to perform multiplexed assays due to the minute amounts of biomolecules in tears and the tiny volume of tears. Herein, the authors leverage DNA tetrahedral frameworks (DTFs) modified on the wireless portable electrodes to effectively capture 3D hybridization chain reaction (HCR) amplifiers for automatic and sensitive monitoring of multiple cytokines in human tears. The developed sensors allow the sensitive determination of various dry eye syndrome (DES)-associated cytokines in human tears with the limit of detection down to 0.1 pg mL-1, consuming as little a

EBV and HSV infections in a patient who had undergone bone marrow transplantation: oral manifestations and diagnosis by in situ nucleic acid hybridization.

The course of infections with herpes simplex virus and Epstein-Barr virus in an immunosuppressed patient who had undergone bone marrow transplantation and had tested seronegative for human immunodeficiency virus is described. The clinical oral manifestations were unusual, as they included hairy leukoplakia-like lesions and extensive mucosal ulceration. Histologic examination disclosed unique features consisting of both lichenoid and viral cytopathic changes. The association of the lesions with both Epstein-Barr virus and herpes simplex virus was confirmed by in situ hybridization histochemistr

Use of peptide nucleic acid fluorescent in situ hybridization technique in the detection of Mycobacterium avium paratuberculosis in fecal samples from dairy herds.

Novel peptide nucleic acid (PNA) probes were designed for direct detection of Mycobacterium avium paratuberculosis (MAP) by the Fluorescent in situ Hybridization technique from fecal samples. For this study, fecal samples (n = 218) were collected from cattle (n = 180) and buffaloes (n = 38) with a history or incidence of cases of chronic intermittent diarrhoea from organized dairy farms in the state of Punjab. Fluorescent in situ Hybridization was done on highly positive (acid-fast positive) fecal samples (n = 54) using a newly designed MAP probe. The designed probe was standardized for the st

Demonstration of human papillomavirus DNA by nucleic acid in situ hybridization in paired histologically abnormal cervical biopsies obtained at the same patient visit.

Fifty-four pairs of cervical biopsies ranging from minimal dysplasia to severe dysplasia were studied for the presence of human papillomavirus DNA by in situ hybridization. Two assays were performed on each biopsy. A 16 hour hybridization was used in one assay, while a 40 hour hybridization was utilized in the second assay. Increasing the hybridization time to 40 hours did not significantly increase the detection rate of HPV compared to the rate found using the 16 hour hybridization. Also, no difference in the detection rate of HPV was found by using one biopsy of the pair over the other biops

Demonstration of pseudorabies virus DNA in the mouse inner ear by an in situ nucleic acid hybridization technique in plastic embedded bony material.

This investigation is concerned with the possibility of identifying viral DNA using the in situ DNA hybridization method in methylmethacrylate-embedded material. As an experimental model we chose viral labyrinthitis produced by intranasal infection of the mouse with pseudorabies virus. Fixation and embedding methods specially adapted to this procedure and bony histology preparation technique (specimens by grinding or micromilling) made it possible to identify viral DNA directly morphologically and virologically in the inner ear. Quantitative microphotometric analyses of trans-sagittal sections

The presence of unique DNA sequences after viral induction of leukemia in mice. (RNA tumor virus-nucleic acid hybridization-insertion of viral DNA).

From previous studies, lymphocyte DNA from human leukemias and DNA from involved tissues of patients with Hodgkin's disease or Burkitt's lymphoma contain sequences that are absent from their normal counterparts. These sequences are related to those found in particulate elements associated with these neoplasias and possessing biochemical properties characteristic of RNA tumor viruses. Similar observations have been made of unique sequences related to those of the feline virus RD-114 and found in spontaneous mastocytomas in cats. Here we extend these results to the classical murine model of viru

Lateral flow nucleic acid biosensor for sensitive detection of microRNAs based on the dual amplification strategy of duplex-specific nuclease and hybridization chain reaction.

MicroRNAs (miRNAs) constitute novel biomarkers for various diseases. Accurate and quantitative analysis of miRNA expression is critical for biomedical research and clinical theranostics. In this study, a method was developed for sensitive and specific detection of miRNAs via dual signal amplification based on duplex specific nuclease (DSN) and hybridization chain reaction (HCR). A reporter probe (RP), comprising recognition sequence (3' end modified with biotin) for a target miRNA of miR-21 and capture sequence (5' end modified with Fam) for HCR product, was designed and synthesized. HCR was i
