Coronavirus contains three envelope proteins, M, E and S, and a nucleocapsid, which consists of genomic RNA and N protein, within the viral envelope. We studied the macromolecular interactions involved in coronavirus assembly in cells infected with a murine coronavirus, mouse hepatitis virus (MHV). Coimmunoprecipitation analyses demonstrated an interaction between N protein and M protein in infected cells. Pulse-labeling experiments showed that newly synthesized, unglycosylated M protein interacted with N protein in a pre-Golgi compartment, which is part of the MHV budding site. Coimmunoprecip

Coronaviruses are capable of inducing diverse infectious diseases that pose significant threats to the public health and the economic development. With a single positive-stranded RNA genome, coronaviruses utilize viral proteins to execute diverse immune escape strategies to facilitate their replication. Of all the identified structural proteins and non-structural proteins within the coronaviruses, nucleocapsid (N) protein is highly conserved and is the most abundant viral protein in infected host cells. N protein regulates the more complex and diverse mechanisms through which viruses suppress

Betacoronaviruses (β-CoVs) display divergent mechanisms to evade host antiviral responses, yet the evolutionary origin and functional relevance of their strategies remain unclear. Stress granules (SGs), central to host defenses, are disrupted by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) nucleocapsid (N) protein via its interaction with G3BP1 mediated by an 15ITFG18 motif. This interaction inhibits SG assembly, enhancing viral replication and immune evasion. Here, we analyzed 179 N protein sequences across β-CoVs subgenera and identified the ITFG motif in sarbecoviruses b

Mumps virus (MuV) is a highly contagious human pathogen and frequently causes worldwide outbreaks despite available vaccines. Similar to other mononegaviruses such as Ebola and rabies, MuV uses a single-stranded negative-sense RNA as its genome, which is enwrapped by viral nucleoproteins into the helical nucleocapsid. The nucleocapsid acts as a scaffold for genome condensation and as a template for RNA replication and transcription. Conformational changes in the MuV nucleocapsid are required to switch between different activities, but the underlying mechanism remains elusive due to the absence

We have previously provided evidence that a strong and precise interaction of the Sindbis virus nucleocapsid with the cytoplasmic tail of E2 is critical for the function of the mature virion and that changes in this association may affect infectivity (21). In this study, we examined the effects of temperature-sensitive defects in capsid protein on virus infectivity and glycoprotein function. The two Sindbis virus mutants chosen were ts2 and Ser180/Gly183, which contain amino acid substitutions in their capsid proteins. Heating these mutants to 60 degrees C results in a loss of infectivity sign

The SARS-CoV-2 nucleocapsid protein (Np) is essential for viral RNA replication and genomic RNA packaging. Phosphorylation of Np within its central Ser-Arg-rich (SRR) linker is proposed to modulate these functions. To gain mechanistic insights into these distinct roles, we performed in vitro biophysical and biochemical studies using recombinantly expressed ancestral Np and phosphomimetic SRR variants. Limited-proteolysis showed minor cleavage differences between wild-type (WT) and phosphomimetic Np, but no major structure or stability changes in the N- and C-terminal domains were observed by c

The gene for the nucleocapsid (N) protein of canine distemper virus was cloned into the pMD-18T vector, and positive recombinant plasmids were obtained by enzyme digestion and sequencing. After digestion by both EcoRI and KpnI, the plasmid was directionally cloned into the eukaryotic expression vector pcDNA; the positive clone pcDNA-N was screened by electrophoresis and then transfected into COS-7 cells. Immunofluorescence analysis results showed that the canine distemper virus N protein was expressed in the cytoplasm of transfected COS-7 cells. After emulsification in Freund's adjuvant, the r

Bunyamwera virus (BUNV), which belongs to the genus Orthobunyavirus, is the prototypical virus of the Bunyaviridae family. Similar to other negative-sense single-stranded RNA viruses, bunyaviruses possess a nucleocapsid protein (NP) to facilitate genomic RNA encapsidation and virus replication. The structures of two NPs of members of different genera within the Bunyaviridae family have been reported. However, their structures, RNA-binding features, and functions beyond RNA binding significantly differ from one another. Here, we report the crystal structure of the BUNV NP-RNA complex. The polyp

The nucleocapsid (N) protein of SARS-coronavirus (SARS-CoV) is the key protein for the formation of the helical nucleocapsid during virion assembly. This protein is believed to be more conserved than other proteins of the virus, such as spike and membrane glycoprotein. In this study, the N protein of SARS-CoV was expressed in Escherichia coli DH5alpha and identified with pooled sera from patients in the convalescence phase of SARS. A plasmid pCI-N, encoding the full-length N gene of SARS-CoV, was constructed. Expression of the N protein was observed in COS1 cells following transfection with pC

SARS-CoV-2 variants display substantial genetic and phenotypic diversity, yet the molecular mechanisms underlying their effects on host gene regulation need further investigation. We performed a transcriptomic analysis of host responses to multiple SARS-CoV-2 variants, including Delta and Omicron. Despite their divergence, all variants induced a conserved transcriptional program in human lung epithelial cells characterized by activation of interferon regulatory factors (IRFs), immediate-early genes, and downstream cytokines. We identify the viral nucleocapsid (N) protein as a central driver of

The single-stranded RNA genome of the SARS-CoV-2 virus is characterized by a complex secondary structure formed by patches of intramolecular RNA double-strands. Here, we show that the nucleocapsid (N) protein is not only the specific viral RNA packaging protein, but also acts as an RNA chaperone, facilitating RNA folding. RNA chaperones are classified by their non-specific RNA binding and the presence of intrinsically disordered regions (IDRs). N possesses three IDRs, separated by the structured RNA-binding domain (RBD) and the C-terminal domain (CTD). Our study identifies the amino acids 46-3

The emergence of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) as the causative agent of COVID-19 precipitated a global health crisis of unprecedented scale. SARS-CoV-2 has been shown to interfere specifically with S phase progression during early stages of infection. Nucleocapsid (N) is an important structural protein. The abundance and early presence of N suggest that the N protein may play a pivotal role in determining the fate of host cells post-infection, including in cell cycle regulation. Our observations reveal that the SARS-CoV-2 N protein actually induces S phase arres

Hantaviruses cause two severe diseases, haemorrhagic fever with renal syndrome in Eurasia and hantavirus pulmonary syndrome in the Americas. To understand more about the molecular mechanisms that lead to these diseases, the associations of Puumala virus nucleocapsid protein (PUUV-N) with cellular proteins were studied by yeast two-hybrid screening. Daxx, known as an apoptosis enhancer, was identified from a HeLa cDNA library and its interaction with PUUV-N was confirmed by GST pull-down assay, co-immunoprecipitation and co-localization studies. Furthermore, domains of interaction were mapped t

The nucleocapsid protein (NC) of all animal retroviruses, encoded by the gag gene, is the major structural protein of the core ribonucleoprotein complex, bound to genomic RNA in mature virions. NC is also thought to play one or more accessory roles in reverse transcription. Mature NC (p7NC) from human immunodeficiency virus type 1 (HIV-1) is a 71-amino acid, basic protein which contains two Cys3His Zn(II) retroviral-type zinc finger domains. Herein, we describe the subcloning and expression of HIV-1 NC, denoted NC71, from an inducible phage T7 RNA polymerase promoter in Escherichia coli. Purif

An inhibition radioimmunoassay and a two-site immunoradiometric (four-layer) assay for the quantification of measles virus nucleocapsid antigen were established and their sensitivities compared. Both assays exhibited threshold sensitivities of from 1 to 10 ng nucleocapsid antigen per ml. Concentrations of reagents used in the assays were shown to be an important factor determining the sensitivities of the assays. Purified mumps or parainfluenza 2 virus nucleocapsids did not compete with measles virus nucleocapsids in the inhibition assay but some degree of cross-reactivity with canine distempe

The nucleocapsid (N) protein of the severe acute respiratory syndrome coronavirus (SARSCoV) is the most abundant structural protein in virus-infected cells. Its primary function is to pack ~30 kb positive-sense viral RNA genome that includes a 5' cap into a ribonucleoprotein (RNP) complex known as capsid. Among the 4 major structural proteins of SARS-CoV, the N protein exhibits the highest expression levels in the host cells. We produced a recombinant SARS-CoV N protein (SARSN1) and used it to detect specific antibodies in mice vaccinated with a live probiotic vaccine containing the coronaviru

Porcine deltacoronavirus (PDCoV) is an emerging enteropathogenic coronavirus that causes acute diarrhea in piglets and poses a potential zoonotic risk. Effective antiviral agents against PDCoV are currently lacking. In this study, we identified a nanobody, designated 62Nb, that specifically targets the nucleocapsid (N) protein of PDCoV and potently inhibits viral replication. Stable expression of 62Nb in porcine ST cells significantly reduced N protein levels, viral RNA copies, and progeny virus titers without affecting cell viability, indicating a robust antiviral effect. Mechanistic analysis

IFI16 is an interferon-inducible protein that senses viral DNA and can also restrict RNA virus replication. Here, using IFI16 knockout cellular models, we identify IFI16 as a host restriction factor that limits SARS-CoV-2 replication. Upon SARS-CoV-2 infection, IFI16 relocalizes from the nucleus to the cytoplasm, where it binds both the nucleocapsid protein and the viral genome. This impairs SARS-CoV-2 replication by inhibiting viral RNA-induced condensate formation of the nucleocapsid protein. Finally, we extend our analysis to other human coronaviruses and observe that IFI16 depletion also e
