The nootropic drug piracetam was investigated in various experimental models of epilepsy. Generally, piracetam exhibits no or only moderate anticonvulsant properties against generalized tonic or clonic seizures. However, in many cases it did increase the anticonvulsant effectiveness of conventional antiepileptics, as shown in the maximal electroshock seizure (MES) threshold test, the traditional MES test or in DBA/2 mice. Sixty children with dyslexia (41 boys, 19 girls; ages 9 to 13) were enrolled in a 10-week summer tutoring program that emphasized word-building skills. They were randomly and blindly assigned to receive either placebo or piracetam, a purportedly memory-enhancing drug that has been reported to facilitate reading skill acquisition. Two randomized studies to test the efficacy of a combination of Piracetam and Ergotoxin on the destabilized brainstem are reported. In the first study, a pharmacological model using Minocycline is employed. A follow-up clinical study analysed the effect of the preparation in 5 patients suffering from vertigo and other related complaints. It was seen that there was a significant improvement in the nystagmus profiles of the pharmacological model volunteers. Similarly, the patient group showed a marked improvement in symptoms, and in orientation capability as tested by Cranio-Corpo-Graphy.

We describe a patient with cerebellar ataxia of degenerative nature who was administered high-dose piracetam in a single-blind trial. Piracetam was demonstrated to be highly effective on tandem gait and gait ataxia in daily doses of 60 g. We suggest piracetam has a potential anti-ataxic effect in human cerebellar ataxia when used in considerably higher doses than those indicated for other purposes.

In a double-blind trial according to a switchback design with 4 periods of one week each a comparison was made between the effects of piracetam and a placebo.
