Accelerated organelle movements can indirectly affect the motility of other organelles in plant cells.

Organelle movements in plants are driven by myosin XI motors moving along the actin filament cytoskeleton. Several experiments have suggeste … ing approach to modify the movements of specific organelles demonstrates that organelle motility in plant cells can occur independently but nevertheless is coordinated between different types of organelles.

Flavonol-based fluorescent probe for bisulfite detection in plant cells and environmental water samples.

Bisulfite (HSO3-), which exists in equilibrium with sulfite and sulfur dioxide derivatives, plays significant roles in food preservation and biological systems. However, elevated levels of bisulfite are associated with various physiological disorders. Therefore, the development of reliable methods for monitoring bisulfite is of considerable importance. Herein, we report a flavonol-based fluorescent probe, 4-(3-hydroxy-4-oxo-4H-chromen-2-yl) benzaldehyde (BSP), synthesized via a simple two-step reaction using 1-(2-hydroxyphenyl)ethan-1-one and 4-(diethoxy methyl)benzaldehyde. BSP exhibits excel

Comparative evaluation of plant extract effects on peritoneal, medullary and J774 cells. G8 macrophages.

The use of medicinal plants as raw material for extracts production and pure substances isolation and subsequence development of new drugs represents a constantly growing area. However, some stages are indispensable before pharmacologically evaluating natural products such as medicines. Toxicity tests in mammalian cells are essential to initiate new drugs development or verify the substance's biocompatibility. Thus, we verified the toxicity of crude extracts and fractions with different polarities obtained from the leaves and stems of eight plant species. The toxic effect was evaluated on macr

Plant extracts and betulin from Ligaria cuneifolia inhibit P-glycoprotein function in leukemia cells.

Overexpression of P-glycoprotein (P-gp), which is linked to multidrug resistance (MDR), is one of the underlying obstacles to the success of chemotherapy as it reduces the efficacy of anticancer drugs and the side effects of these increase as a result of any increased dose to achieve the therapeutic effect. To identify agents with P-gp inhibitory properties, ethanol extracts from 80 plants were screened for their ability to increase intracellular doxorubicin-associated fluorescence, and the extract of Ligaria cuneifolia was found to be the most effective. Its bioassay-guided isolation yielded

Expression of Multiple Taenia Solium Immunogens in Plant Cells Through a Ribosomal Skip Mechanism.

Taenia solium cysticercosis is a major parasitic disease that affects the human health and the economy in underdeveloped countries. Porcine cysticercosis, an obligatory stage in the parasite life cycle, is a suitable target for vaccination. While several recombinant and synthetic antigens proved to be effective as vaccines, the cost and logistic difficulties have prevented their massive use. Taking this into account, a novel strategy for developing a multi-epitope low-cost vaccine is herein explored. The S3Pvac vaccine components (KETc1, KETc12, KETc7, and GK1 [KETc7]) and the protective HP6/T

Plant-derived 3,3'-Diindolylmethane is a strong androgen antagonist in human prostate cancer cells.

3,3'-Diindolylmethane (DIM) is a major digestive product of indole-3-carbinol, a potential anticancer component of cruciferous vegetables. Our results indicate that DIM exhibits potent antiproliferative and antiandrogenic properties in androgen-dependent human prostate cancer cells. DIM suppresses cell proliferation of LNCaP cells and inhibits dihydrotestosterone (DHT) stimulation of DNA synthesis. These activities were not produced in androgen-independent PC-3 cells. Moreover, DIM inhibited endogenous PSA transcription and reduced intracellular and secreted PSA protein levels induced by DHT i

The alkaline comet test on plant cells: a new genotoxicity test for DNA strand breaks in Vicia faba root cells.

The alkaline comet assay (single cell gel electrophoresis assay) is a sensitive method for the detection of DNA damage. This paper describes the first application of this assay to plant cells for genotoxicological assessment. Germinating Vicia faba (field bean) seedlings were kept in water with either methyl methanesulfonate (MMS), ethyl methanesulfonate (EMS), mitomycin C (MMC), cycloheximide (CH), cadmium chloride (CdCl2), potassium dichromate (K2Cr2O7), or chromium trichloride (CrCl3). Nuclei were isolated from the root cells and evaluated for the extent of DNA migration. With the exception…

Cell-cell signaling controls Xylella fastidiosa interactions with both insects and plants.

Xylella fastidiosa, which causes Pierce's disease of grapevine and other important plant diseases, is a xylem-limited bacterium that depends on insect vectors for transmission. Although many studies have addressed disease symptom development and transmission of the pathogen by vectors, little is known about the bacterial mechanisms driving these processes. Recently available X. fastidiosa genomic sequences and molecular tools have provided new routes for investigation. Here, we show that a diffusible signal molecule is required for biofilm formation in the vector and for vector transmission to

Role of primary protectors of plant cells in salinity tolerance: molecular mechanisms and adaptive strategies.

Salinity stress is among the most pervasive abiotic factors limiting plant growth and agricultural productivity worldwide, currently affecting more than 20% of irrigated croplands. Elevated salt concentrations induce osmotic imbalance, ionic toxicity, and oxidative stress, leading to disruptions in photosynthesis, metabolism, and plant development. To counteract these deleterious effects, plants have evolved intricate, highly coordinated defense systems comprising structural and functional cellular protectors. At the structural level, the cell wall, plasma membrane, vacuole, and peroxisome pla

DEGAST3D: Learning Deformable 3D Graph Similarity to Track Plant Cells in Unregistered Time Lapse Images.

Tracking plant cells in three-dimensional (3D) tissue captured through light microscopy presents significant challenges due to the large number of densely packed cells, non-uniform growth patterns, and variations in cell division planes across different cell layers. In addition, images of deeper tissue layers are often noisy, and systemic imaging errors further exacerbate the complexity of the task. In this paper, we propose a novel learning-based method DEGAST3D: Learning Deformable 3D GrAph Similarity to Track Plant Cells in Unregistered Time Lapse Images exploits the tightly packed 3D cell …

Affordable oral proinsulin bioencapsulated in plant cells regulates blood sugar levels similar to natural insulin.

Diabetes Mellitus is a silent epidemic affecting >500 million, which claimed 6.7 million lives in 2021, a projected increase of >670% in <20 years old in the next two decades but insulin is unaffordable for the large majority of the globe. Therefore, we engineered proinsulin in plant cells to facilitate oral delivery. Stability of the proinsulin gene and expression in subsequent generations, after removal of the antibiotic-resistance gene, was confirmed using PCR, Southern and western blots. Proinsulin expression was high (up to 12 mg/g DW or 47.5% of total leaf protein), stable up to one year…

Plant immune responses - from guard cells and local responses to systemic defense against bacterial pathogens.

When plants are infected by pathogens two distinct responses can occur, the early being a local response in the infected area, and later a systemic response in non-infected tissues. Closure of stomata has recently been found to be a local response to bacterial pathogens. Stomata closure is linked to both salicylic acid (SA), an essential hormone in local responses and systemic acquired resistance (SAR), and absisic acid (ABA) a key regulator of drought and other abiotic stresses. SAR reduces the effects of later infections. In this review we discuss recent research elucidating the role of guar

Identification of antitumoral agents against human pancreatic cancer cells from Asteraceae and Lamiaceae plant extracts.

BACKGROUND: Pancreatic cancer is one of the most aggressive and mortal cancers. Although several drugs have been proposed for its treatment, it remains resistant and new alternatives are needed. In this context, plants and their derivatives constitute a relevant source of bioactive components which might efficiently inhibit tumor cell progression. METHODS: In this study, we have analyzed the potential anti-carcinogenic effect of different Asteraceae (Achillea millefolium and Calendula officinalis) and Lamiaceae (Melissa officinalis and Origanum majorana) plant extracts obtained by different g

Effective plague vaccination via oral delivery of plant cells expressing F1-V antigens in chloroplasts.

The chloroplast bioreactor is an alternative to fermentation-based systems for production of vaccine antigens and biopharmaceuticals. We report here expression of the plague F1-V fusion antigen in chloroplasts. Site-specific transgene integration and homoplasmy were confirmed by PCR and Southern blotting. Mature leaves showed the highest level of transgene expression on the third day of continuous illumination, with a maximum level of 14.8% of the total soluble protein. Swiss Webster mice were primed with adjuvant-containing subcutaneous (s.c.) doses of F1-V and then boosted with either adjuva

Decreased levels of heat shock proteins in gut epithelial cells after exposure to plant lectins.

BACKGROUND: The enterocytes of the intestinal epithelium are regularly exposed to potentially harmful substances of dietary origin, such as lectins. Expression of heat shock proteins (HSPs) by this epithelium may be part of a protective mechanism developed by intestinal epithelial cells to deal with noxious components in the intestinal lumen. AIM: To investigate if the lectins PHA, a lectin from kidney beans (Phaseolus vulgaris) and WGA, a lectin from wheat germ (Triticum aestivum) could modify the heat shock response in gut epithelial cells and to establish the extent of this effect. METHOD

PLASTID ENVELOPE ION CHANNELS (PEC1/2) link Ca2+ and jasmonic acid signaling in plant cells.

Plants constantly encounter adverse environmental interactions. One organelle is particularly specialized in stress signaling and phytohormone synthesis: the plastid. Calcium (Ca2+), a key second messenger, is known to intersect with cellular phytohormone signaling networks. While cytosolic Ca2+ dynamics have been studied extensively, the physiological relevance of stromal Ca2+ transients and the identity of channels mediating rapid Ca2+ flux into plastids remain largely unexplored. In this study, we provide evidence for PLASTID ENVELOPE ION CHANNELS (PECs) as long-sought mediators of fast-act
