Sexual Development in Non-Human Parasitic Apicomplexa: Just Biology or Targets for Control?

The phylum Apicomplexa is a major group of protozoan parasites including gregarines, coccidia, haemogregarines, haemosporidia and piroplasms, with more than 6000 named species. Three of these subgroups, the coccidia, hemosporidia, and piroplasms, contain parasites that cause important diseases of humans and animals worldwide. All of them have complex life cycles involving a switch between asexual and sexual reproduction, which is key to their development. Fertilization (i.e., fusion of female and male cells) results in the formation of a zygote that undergoes meiosis, forming a new generation

Differential effects of polyploidy and diploidy on fitness of apomictic Boechera.

The co-occurrence of apomixis (asexual reproduction) and polyploidy in plants has been the subject of debate in regard to the origin and evolution of asexuality. In recent years, polyploidy has been postulated as a maintenance and stabilization factor rather than as a source of apomixis origin. It is assumed polyploidy facilitates the compensation for mutation accumulation, and hence, the rare occurrence of diploid apomixis indirectly supports this finding. Nevertheless, diploid apomicts exist and are successful, especially in the genus Boechera. While comparing phenotypic traits, fitness-rela

Sexual structures in Aspergillus: morphology, importance and genomics.

The genus Aspergillus comprises a few hundred species sharing a common asexual spore forming structure, the aspergillum. Approximately one-third of these species also produce a sexual stage, all but five of which are known to be homothallic. Sexual stages associated with Aspergillus fall into approximately ten different genera, reflecting a tremendous degree of phylogenetic and biological diversity. Sexual stages in Aspergillus are plectomycetous, typical for the order in which it resides, the Eurotiales. Theoretically, a homothallic Aspergillus species can produce both asexual conidia and sex

Temperature during Aspergillus fumigatus conidiophore development primes spore transcriptome for asexual, parasexual or sexual development.

UNLABELLED: Aspergillus fumigatus is a thermotolerant saprobe found in soils and plant debris worldwide and an important pathogen of humans causing two million deaths annually. A. fumigatus makes abundant asexual spores (conidia) which are widely distributed by wind and can be inhaled from the environment. In susceptible individuals inhaled conidia break dormancy, germinate and grow in the lung leading to serious disease. Recent work has shown that conidia made at 37°C and 50°C have different morphologies and germination kinetics. While the asexual cycle is well-characterized at 37°C, much les

[Progress of researches on developmental processes and reproduction mode of Pneumocystis].

Pneumocystis, an important opportunistic fungal pathogen that parasitizes in multiple mammalian lungs, may cause life-threatening Pneumocystis pneumonia (PCP) and even death among immunocompromised individuals. With the rapid development of high-throughput sequencing and multi-omics technologies, systematic comparative analyses of genome, transcriptome, and whole-genome sequencing results demonstrate that Pneumocystis is a type of obligate biotrophic fungi, and requires obtaining nutrition from hosts. In addition, sexual reproduction is an essential process for Pneumocystis survival, productio

Regulatory basis for reproductive flexibility in a meningitis-causing fungal pathogen.

Pathogenic fungi of the genus Cryptococcus can undergo two sexual cycles, involving either bisexual diploidization (after fusion of haploid cells of different mating type) or unisexual diploidization (by autodiploidization of a single cell). Here, we construct a gene-deletion library for 111 transcription factor genes in Cryptococcus deneoformans, and explore the roles of these regulatory networks in the two reproductive modes. We show that transcription factors crucial for bisexual syngamy induce the expression of known mating determinants as well as other conserved genes of unknown function.

MHC genes and parasitism in Carassius gibelio, a diploid-triploid fish species with dual reproduction strategies.

BACKGROUND: The gibel carp is a fish species with dual reproduction modes, gynogenesis and sexual reproduction, coexisting in mixed diploid-polyploid populations. Following the Red Queen (RQ) assumption, asexual organisms are, due to their low genetic diversity, targets for parasite adaptation. Because MHC polymorphism is maintained by selection from parasites and sexual selection, MHC genes are considered as a suitable candidate for testing the RQ hypothesis. In this study, we investigated MHC variability and the selection pressure acting on MHC genes in sexual diploids and asexual triploids.

Endosome motility controls light-responsive reproductive development and secondary metabolite production in Aspergillus.

Filamentous fungi, such as Aspergillus species, use microtubule transport to move early endosomes. Other cargos, such as peroxisomes and mRNAs, "hitchhike" on early endosomes to move throughout the long hyphae of these organisms. In Aspergillus nidulans, peroxisomes hitchhike on early endosomes using the endosomal protein PxdA and the peroxisomal protein AcbdA. The HookA adaptor protein links endosomes to microtubule motors. Here, we set out to explore the physiological functions of peroxisome hitchhiking and endosome motility. A. nidulans has a complex life cycle that includes asexual and sex

Sexual partners for the stressed: facultative outcrossing in the self-fertilizing nematode Caenorhabditis elegans.

Sexual reproduction shuffles genetic variation, potentially enhancing the evolutionary response to environmental change. Many asexual organisms respond to stress by generating facultative sexual reproduction, presumably as a means of escaping the trap of low genetic diversity. Self-fertilizing organisms are subject to similar genetic limitations: the consistent loss of genetic diversity within lineages restricts the production of variation through recombination. Selfing organisms may therefore benefit from a similar shift in mating strategy during periods of stress. We determined the effects o

Unisexual reproduction reverses Muller's ratchet.

Cryptococcus neoformans is a pathogenic basidiomycetous fungus that engages in outcrossing, inbreeding, and selfing forms of unisexual reproduction as well as canonical sexual reproduction between opposite mating types. Long thought to be clonal, >99% of sampled environmental and clinical isolates of C. neoformans are MATα, limiting the frequency of opposite mating-type sexual reproduction. Sexual reproduction allows eukaryotic organisms to exchange genetic information and shuffle their genomes to avoid the irreversible accumulation of deleterious changes that occur in asexual populations, kno

Functional Characterization of AoFlbB and AoFlbD Reveals Their Roles in Sporulation and Trap Formation in Arthrobotrys oligospora.

Sporulation is essential for asexual reproduction and dispersal in filamentous fungi, but the roles of upstream sporulation regulators in nematode-trapping (NT) fungi remain poorly understood. In this study, we characterized two Flb homologs, AoFlbB and AoFlbD, in Arthrobotrys oligospora. AoFlbB contains a bZIP-type domain, whereas AoFlbD contains a Myb-like DNA-binding domain, and both proteins are conserved among NT fungi. Deleting AoflbB or AoflbD had little effect on hyphal growth but markedly reduced spore production and altered spore germination, and real-time quantitative PCR analysis s

Laboratory synthesis of an independently reproducing vertebrate species.

Speciation in animals commonly involves an extrinsic barrier to genetic exchange followed by the accumulation of sufficient genetic variation to impede subsequent productive interbreeding. All-female species of whiptail lizards, which originated by interspecific hybridization between sexual progenitors, are an exception to this rule. Here, the arising species instantaneously acquires a novel genotype combining distinctive alleles from two different species, and reproduction by parthenogenesis constitutes an effective intrinsic barrier to genetic exchange. Fertilization of diploid parthenogenet

Targeting protein-protein interactions in Plasmodium: from asexual replication to sexual development.

BACKGROUND: Malaria, a life-threatening protozoan disease caused by Plasmodium parasites and transmitted by mosquitoes, remains a global public health crisis. The 2025 World Malaria Report recorded 282 million malaria cases and 610,000 deaths in 2024, with global elimination goals severely hampered by widespread insecticide resistance and the rapid spread of artemisinin-resistant parasites. Conventional antimalarials primarily target enzyme catalytic sites, which are vulnerable to resistance via single point mutations with minimal parasite fitness cost. In contrast, core Plasmodium biological

Genomic confirmation of hybridisation and recent inbreeding in a vector-isolated Leishmania population.

Although asexual reproduction via clonal propagation has been proposed as the principal reproductive mechanism across parasitic protozoa of the Leishmania genus, sexual recombination has long been suspected, based on hybrid marker profiles detected in field isolates from different geographical locations. The recent experimental demonstration of a sexual cycle in Leishmania within sand flies has confirmed the occurrence of hybridisation, but knowledge of the parasite life cycle in the wild still remains limited. Here, we use whole genome sequencing to investigate the frequency of sexual reprodu

Identification of Mortierella wolfii, a causative agent of mycotic abortion in cattle.

Of several media tested for their ability to induce asexual reproduction in confirmed or suspected isolates of Mortierella wolfii obtained from cattle in various geographical locations, only silage extract agar worked consistently and rapidly. Its use should provide a simple reliable culturing procedure to assist with identification of clinical isolates of this fungus. Temperature growth response curves and electrophoretic patterns of soluble protein extracts of these isolates with characteristic morphological features of M. wolfii indicate substantial variation amongst them.

The private life of malaria parasites: Strategies for sexual reproduction.

Malaria parasites exhibit a complex lifecycle, requiring extensive asexual replication in the liver and blood of the vertebrate host, and in the haemocoel of the insect vector. Yet, they must also undergo a single round of sexual reproduction, which occurs in the vector's midgut upon uptake of a blood meal. Sexual reproduction is obligate for infection of the vector and thus, is essential for onwards transmission to new hosts. Sex in malaria parasites involves several bottlenecks in parasite number, making the stages involved attractive targets for blocking disease transmission. Malaria parasi

Antifungal Resistance in Pulmonary Aspergillosis.

Aspergilli may cause various pulmonary diseases in humans, including allergic bronchopulmonary aspergillosis (ABPA), chronic pulmonary aspergillosis (CPA), and acute invasive pulmonary aspergillosis (IPA). In addition, chronic colonization may occur in cystic fibrosis (CF). Aspergillus fumigatus represents the main pathogen, which may employ different morphotypes, for example, conidia, hyphal growth, and asexual sporulation, in the various Aspergillus diseases. These morphotypes determine the ease by which A. fumigatus can adapt to stress by antifungal drug exposure, usually resulting in one o

Circadian regulation of reproduction and neoblast cell division in the catenulid flatworm Stenostomum virginianum (Platyhelminthes).

The molecular circadian clock and melatonin secretion are pivotal for coordinating homeostasis pathways in many animals. Melatonin has been shown to regulate reproduction and neoblast proliferation in triclad flatworms, yet the extent to which these mechanisms operate in other platyhelminths remains unknown. We examined light sensing, endogenous melatonin production, and circadian control of asexual reproduction in the early-branching catenulid Stenostomum virginianum. Microscopy revealed refractile bodies with photoreceptor-like structures, blue-light autofluorescence, and PAX6 immunoreactivi

The influence of reproductive mode on resource competition and diversity patterns in Ediacaran early animal communities.

The appearance of the oldest known animals during the late Ediacaran period (~574 million years ago) was followed by a phase of slow diversification, until a later burst of rapid diversification known as the Ediacaran 'second wave'. The reasons behind the tempo of diversification are poorly understood. Here we investigate how reproductive mode mediated community dynamics and in turn Ediacaran macroevolutionary change. We show that widespread reproduction via stolon (namely via filaments connecting clones) limited within-species competition, leading to between-species competition acting at smal
