In the adult retina, ramifying microglia interact with the outer plexiform layer (OPL) monitoring the synaptic integrity between photoreceptors and post-synaptic target cells. Microglia are reactive during photoreceptor diseases, but their disease-related function(s) are not fully understood. Retinal explant cultures are model systems used to study degenerative events including photoreceptor degeneration and gliosis. Our culture paradigm, with adult porcine retinas subjected to coculture with human A-retinal pigment epithelia-19 (ARPE) cells, is an experimental approach resulting in improved p

The initial step in visual perception, i.e., the collection of light, is carried out by the outer segments of photoreceptor cells. These outer segments, specialized primary cilia, house the photopigments that absorb photons. Here, we report that EFA6A, an exchange factor for the small G protein Arf6, previously described as a regulator of early ciliogenesis by promoting distal appendage vesicle fusion, is essential for visual function. We demonstrate that EFA6A is present in various layers of the mouse retina, including the photoreceptors and the retinal pigment epithelium (RPE). Accordingly,

Cellular expression of the mRNA for peripherin/rds was studied in rat retinas by in situ hybridization histochemistry with antisense and sense probes prepared from polymerase chain reaction-amplified cDNA of the bovine peripherin/rds. Predominant mRNA signals were detected in the inner segments and distal region of the outer nuclear layer, but not in other layers of the retina including retinal pigment epithelium or in the choroid, giving evidence that the gene product, peripherin/rds, is synthesized specifically in the inner segment ribosomes to form outer segment disc membranes of photorecep

Retinal degeneration-3 protein (RD3) plays a dual role in photoreceptors - prevents their degeneration by suppressing aberrant activity of retinal membrane guanylyl cyclase (RetGC) in the inner segment and enables photoreceptor function by facilitating delivery of RetGC to the outer segment. Parts of RD3 structure supporting its dual function were evaluated in vivo using deletion mutants of a human RD3 transgenically expressed under the control of rod opsin promoter in Rd3-/- mouse rods lacking endogenous RD3. The human RD3 truncated after Gly148 or Arg158 not only inhibited RetGC activation b

BACKGROUND: Recent improvements in optical coherence tomographic (OCT) resolution and automated segmentation software have provided a means of relating visual pathway damage to structural changes in the retinal nerve fiber layer (RNFL) and corresponding soma of the ganglion cells in the inner layers of the macula and also in the outer photoreceptor layer in the macula.
EVIDENCE ACQUISITION: Studies correlating retinal structure with function are reviewed in the context of OCT in optic nerve and retinal disorders.
RESULTS: Recently published work provides evidence showing a strong relationshi

Inherited retinal diseases (IRDs) are a rare group of eye disorders characterized by progressive dysfunction and degeneration of retinal cells. In this study, we characterized the raifteirí (raf) zebrafish, a novel model of inherited blindness, identified through an unbiased ENU mutagenesis screen. A mutation in the largest subunit of the endoplasmic reticulum membrane protein complex, emc1 was subsequently identified as the causative raf mutation. We sought to elucidate the cellular and molecular phenotypes in the emc1-/- knockout model and explore the association of emc1 with retinal degener

The accumulation of lipofuscin in the cells of the retinal pigment epithelium (RPE) is thought to play an important role in the development and progression of degenerative diseases of the retina. The bulk of RPE lipofuscin originates in reactions of the rhodopsin chromophore, retinal, with components of the photoreceptor outer segment. The 11-cis retinal isomer is generated in the RPE and supplied to rod photoreceptor outer segments where it is incorporated as the chromophore of rhodopsin. It is photoisomerized during light detection to all-trans and subsequently released by photoactivated rho

Retinitis pigmentosa is a heterogeneous group of inherited retinal disorders in which the photoreceptor cells degenerate. A line of transgenic mice expresses a mutant opsin gene that encodes three missense mutations near the amino terminus, including P23H, which is the basis for a common form of dominant retinitis pigmentosa. By studying the photoreceptor cells of these mice and their normal littermates, we found that: (1) opsin was routed correctly, (2) the concentration of opsin in the disk membranes appeared normal by freeze fracture analysis, (3) the amount of disk membrane shedding was no

Fibroblast growth factor 2 (FGF2) is expressed in retinal Müller glia cells, and its expression increases in response to photoreceptor degeneration. To investigate the physiological relevance of FGF2, we analyzed retinal morphology and cellular responses in Fgf2-deficient (Fgf2-/-) mice. Loss of FGF2 did not affect photoreceptor survival, retinal vasculature, or retinal pigment epithelium (RPE) integrity. To further understand its role in retinal degeneration, Fgf2-/- mice were crossed with Pde6bSTOP/STOP mice, a model of retinitis pigmentosa (RP). We then analyzed outer nuclear layer thicknes

Accumulation of misfolded host proteins is central to neuropathogenesis of numerous human brain diseases including prion and prion-like diseases. Neurons of retina are also affected by these diseases. Previously, our group and others found that prion-induced retinal damage to photoreceptor cells in mice and humans resembled pathology of human retinitis pigmentosa caused by mutations in retinal proteins. Here, using confocal, epifluorescent and electron microscopy we followed deposition of disease-associated prion protein (PrPSc) and its association with damage to critical retinal structures fo

PURPOSE OF THE STUDY: Retinitis pigmentosa (RP) is a group of genetic disorders and a slow loss of vision that is caused by a cascade of retinal degenerative events. We examined whether these retinal degenerative events were reduced after cultured mixtures of adult olfactory ensheathing cells (OECs) and olfactory nerve fibroblasts (ONFs) were transplanted into the subretinal space of 1-month-old RCS rat, a classic model of RP.
MATERIALS AND METHODS: The changes in retinal photoreceptors and Müller cells of RCS rats after cell transplantation were observed by the expression of recoverin and gl

PURPOSE: To identify the distribution of creatine transporter (CRT) in the aged human retina and how this expression pattern is modified after retinal detachment.
METHODS: An affinity-purified antibody raised against the CRT was used in the immunohistochemical investigation. The anti-CRT antibody was colocalized with neuronal markers (calbindin, parvalbumin, Islet-1, calretinin, GAD₆₇, Go-alpha), glia markers (glutamine synthetase, glial fibrillary acid protein), and a blood vessel basal membrane marker (laminin). Confocal microscopy was used to visualize the labeling patterns in retinal sect

Light detection in vertebrate eyes is mediated through retinal photoreceptor rod and cone cells that transduce light signals into electrical responses. The differentiation and synaptogenesis of photoreceptor cells are especially important since these cells are the main targets of degeneration in retinitis pigmentosa. We produced transgenic mice that express enhanced green fluorescent protein (EGFP) under the control of the mouse rhodopsin gene promoter. In Western blot analyses of transgenic retinal homogenates, expression of the endogenous rhodopsin gene was detected from post-natal day (P)8;

The outer layers of the retinas of pigmented rats affected with hereditary retinal degeneration (rdy) were studied at an advanced stage in the degenerative process, ie, in 200 day old animals. At this age, most of the photoreceptors that survived the degenerative process were cones. The chromatin pattern of their nuclei clearly differentiated them from rods, displaced pigment cells and/or presumptive macrophages that also were found in the outer nuclear layer. None of the cones encountered had outer segments although structures resembling outer segment discs were found with a single cilium. Co

Complement factor 3 (C3) has emerged as a primary therapeutic target in age-related macular degeneration (AMD) supported by genetic, histologic, and clinical trial evidence. Yet, the site(s) of action are unclear. The purpose of this study was to test the effect of C3 knockout on photoreceptors and retinal pigment epithelial cells (RPE) in the sodium iodate (NaIO3) model, which mirrors some features of AMD. C3-/- and WT mice, both on a C57Bl/6J background, were injected intraperitoneally with 25 mg/kg NaIO3. Electroretinography and optical coherence tomography were performed 7 days later to as

Ciliopathies are a genetically and phenotypically heterogeneous group of human developmental disorders whose root cause is the absence or dysfunction of primary cilia. Joubert syndrome is characterized by a distinctive hindbrain malformation variably associated with retinal dystrophy and cystic kidney disease. Mutations in CC2D2A are found in ∼10% of patients with Joubert syndrome. Here we describe the retinal phenotype of cc2d2a mutant zebrafish consisting of disorganized rod and cone photoreceptor outer segments resulting in abnormal visual function as measured by electroretinogram. Retinitis Pigmentosa (RP) is a group of inherited retinal diseases characterized by progressive loss of rod followed by cone photoreceptors. An especially early onset form of RP with blindness in teenage years is caused by mutations in mertk, the gene encoding the clearance phagocytosis receptor Mer tyrosine kinase (MerTK). The cause for blindness in mutant MerTK-associated RP (mutMerTK-RP) is the failure of retinal pigment epithelial cells in diurnal phagocytosis of spent photoreceptor outer segment debris. However, the early onset and very fast progression of degeneration in mutMerTK-RP rema
