Abelmoschi Corolla, a traditional Chinese herbal material, is known for its pharmacological properties, including diuretic effects, urinary tract-cleansing, heat and dampness elimination, anti-oedema, and detoxification. Polysaccharides and flavonoids are the primary bioactive constituents of Abelmoschi Corolla. Although the structure-function relationships of flavonoids are well understood, polysa1ccharides, particularly those with potential immunotherapeutic applications, remain comparatively understudied. In this study, we isolated and purified a novel pectin polysaccharide from Abelmoschi

Oral delivery of natural antioxidants represents a promising therapeutic strategy for ulcerative colitis (UC), yet their therapeutic efficacy is hindered by instability and poor accumulation at inflamed sites. To address this, we developed Galectin-3 (Gal-3)-targeted nanoparticles (ZDP-NPs) by encapsulating diosmetin within zein complexes modified with a galactose- and rhamnogalacturonan-I (RG-I)-rich pectin (PMTP, Mw: 228.8 kDa, DM: 34.21 %, RG-I content: 43 %) extracted from Premna microphylla Turcz leaves. Surface plasmon resonance (SPR) analysis confirmed a strong binding affinity between

A novel polysaccharide SPS01-2 (87.5 kDa) was isolated from the roots of Scutellaria baicalensis Georgi. Monosaccharide composition revealed that SPS01-2 consists of rhamnose, arabinose, galactose, galacturonic acid, and glucuronic acid in ratio of 4.4: 67.1: 22.2: 6.3: 1.2. Further investigations using methylation, NMR, and mass spectrometry indicated that SPS01-2 is classified as a type II arabinogalactan (AG-II) with a minor presence of type I rhamnogalacturonan (RG-I). The core structure alternates between 1,2/1,2,4-α-L-Rhap and 1,4-α-D-GalpA, with branches including 1,3,6-β-D-Galp, 1,3-β-

Screening of commercially available fluoro monosaccharides as putative growth inhibitors in Arabidopsis thaliana revealed that 2-fluoro 2-l-fucose (2F-Fuc) reduces root growth at micromolar concentrations. The inability of 2F-Fuc to affect an Atfkgp mutant that is defective in the fucose salvage pathway indicates that 2F-Fuc must be converted to its cognate GDP nucleotide sugar in order to inhibit root growth. Chemical analysis of cell wall polysaccharides and glycoproteins demonstrated that fucosylation of xyloglucans and of N-linked glycans is fully inhibited by 10 μm 2F-Fuc in Arabidopsis s

Sugar beet pulp is rich in rhamnogalacturonan-I (RG-I) dominant pectin and promises functional properties. In this work, an integrated nuclear magnetic resonance (NMR) strategy combining diffusion ordered spectroscopy (DOSY) with qualitative and quantitative NMR was established. Then the molecular hydrodynamic properties and monosaccharide profile of an RG-I containing pectin from sugar beet pulp was characterized. DOSY was applied to measure translational diffusion coefficients, enabling calculation of the hydrodynamic radius (RH = 6.99 nm) and weight-average molecular weight (Mw = 4.66 × 104

Atractylodes polysaccharides (APs) are the core bioactive components responsible for the pharmacological effects of this genus, demonstrating immense potential in the fields of modern medicine, functional foods, and green animal feed development. However, due to the complex chemical structures of APs, the quantitative structure-activity relationships (SAR) between their fine structural features and specific biological functions remain insufficiently elucidated. Furthermore, most current studies lack a translational perspective oriented toward practical applications. This paper systematically r

The dried rhizomes and roots of Polygonum cuspidatum (PC) is a frequently used traditional Chinese medicine with the potential for treating gastrointestinal diseases. However, it is still uncertain whether the macromolecular polysaccharides, especially those of non-glucan types, play an important role in preventing intestinal inflammatory disease. In this study, an acidic polysaccharide (PCP-I) with a molecular weight of 134.45 kDa was isolated from PC, consisting of GalA, Gal, Ara, Rha, Glc and GlcA in a molar ratio of 50.4:25.8:14.0:5.9:3.4:0.5. Despite the well-established role of calcium in pectin gelation, the regulatory mechanisms by which Ca2+ concentration governs the microstructure and functionality of emulsion gels derived from citrus pectin remain poorly understood. This study bridges this knowledge gap by employing citrus pectin with distinct fine structures to fabricate emulsion gel system via Ca2+-mediated cross-linking, systematically elucidating the influence of Ca2+ on gel microstructure, rheology, and stability. The results demonstrated that low-methoxyl pectin (LMP) formed a dense gel network through Ca2+ cross-linkin

Industrial pectin production from citrus peels involves extended, multi-step acidic extraction at elevated temperatures. Pectin quality can be improved by reducing the extraction time or number of steps, but this comes at the expense of yield. A decrease in pectin yield revenue may be compensated for by developing new usage of the partially depectinized citrus residues. One option is to capitalize on the fucosylated xyloglucans present in the residues. Thus, the aim of this work was to maximize gentle extraction of fucosylated xyloglucan from partially depectinized citrus peel residues of oran

Ulcerative colitis (UC) poses a global health burden due to its refractoriness. Our previous study demonstrated that Hypecoum leptocarpum (H. leptocarpum) crude polysaccharides mitigate UC-related intestinal inflammation. In this study, we extracted, purified, and characterized a pectic polysaccharide (HLP-4-2, 19.7 kDa) from H. leptocarpum, which contained homogalacturonan (HG) and rhamnogalacturonan-I (RG-I). Specifically, RG-I comprises a repeating disaccharide unit of 1,4-α-GalpA and 1,2-α-Rhap (with arabinogalactan branches), whereas HG has partially methyl-esterified GalA residues. In DS

A key role of boron in plants is to cross-link the cell wall pectic polysaccharide rhamnogalacturonan-II (RG-II) through borate diester linkages. Phenylboronic acid (PBA) can form the same reversible ester bonds but cannot cross-link two molecules, so can be used as an antagonist to study the function of boron. This study aimed to evaluate the effect of PBA on apple (Malus domestica) pollen tube growth and the underlying regulatory mechanism. We observed that PBA caused an inhibition of pollen germination, tube growth and led to pollen tube morphological abnormalities. Fluorescent labeling, co

In Malian traditional medicine infusions of the roots of Vernonia kotschyana or Cochlospermum tinctorium in water are used for treating gastric ulcer. Helicobacter pylori is known to play a major role in gastric ulcer development, and it was of interest to evaluate a potential anti-adhesive activity towards H. pylori by crude water extracts and isolated polysaccharide fractions from the roots of V. kotschyana and C. tinctorium. The inhibitory effects were examined by an in vitro flow cytometric assay using human gastric adenocarcinoma epithelial cells, where fluorescent-labeled H. pylori were

The pectic polysaccharide WGPP-1a (46.8 kDa) has been obtained from wax gourd peel (WGP) which is a popular soup ingredient and Chinese herbal medicines in China. Monosaccharide composition, methylation analysis, FT-IR and NMR demonstrate that WGPP-1a is composed of rhamnogalacturonan I (RG-I) and homogalacturonan (HG) domains with mass ratios of 4:1. The RG-I domains contain a highly branched structure that is substituted primarily with β-D-1,4-galactan, α-L-1,5-arabinan and β-D-1,4-glucan. In ethanol-induced gastric ulcer mice, WGPP-1a reduced ulcer area, restored mucosal glycoproteins. In v

l-Arabinose is one of the main constituents of cell wall polysaccharides such as pectic rhamnogalacturonan I (RG-I), glucuronoarabinoxylans and other glycoproteins. It is found predominantly in the furanose form rather than in the thermodynamically more stable pyranose form. UDP-L-arabinofuranose (UDP-Araf), rather than UDP-L-arabinopyranose (UDP-Arap), is a sugar donor for the biosynthesis of arabinofuranosyl (Araf) residues. UDP-arabinopyranose mutases (UAMs) have been shown to interconvert UDP-Araf and UDP-Arap and are involved in the biosynthesis of polysaccharides including Araf. The UAM
