Rotenone selectively occludes sensitivity to hypoxia in rat carotid body glomus cells.

Carotid body glomus cells release transmitters in response to hypoxia due to the increase of excitability resulting from inhibition of O2 -regulated K+ channels. However, the mechanisms involved in the detection of changes of O2 tension are unknown. We have studied the interaction between glomus cell O2 sensitivity and inhibition of the mitochondrial electron transport chain (ETC) in a carotid body thin slice preparation in which catecholamine release from intact single glomus cells can be monitored by amperometry. Inhibition of the mitochondrial ETC at proximal and distal complexes induces ex

Involvement of 75NTR extracellular domain in rotenone-induced Parkinson's disease cell models.

OBJECTIVE: While the p75 neurotrophin receptor (p75NTR) is critically implicated in the aggregation of α-synuclein (α-syn), a defining pathological hallmark of Parkinson's disease, the distinct functional contributions of its structural domains remain largely unresolved. METHODS: To investigate this, we employed a rotenone-induced cellular Parkinson's disease model utilizing SH-SY5Y neuroblastoma cells transfected with plasmids encoding specific p75NTR truncation mutants. RESULTS: Overexpression of a mutant representing the p75NTR extracellular domain (HA-p75Δ151, lacking residues 277-427) s

PKM2-driven glycolysis mediates rotenone neurotoxicity via MG-Hs in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder lacking disease-modifying therapies. Rotenone (Rot) is widely used to model PD, but its neurotoxicity is not fully understood beyond mitochondrial complex I inhibition. Here, we identify a glycolytic mechanism that contributes to Rot-induced neuronal damage downstream of complex I inhibition. Our in vitro data demonstrate that Rot enhances glycolytic flux, leading to accumulation of methylglyoxal-derived hydroimidazolones (MG-Hs), which drive irreversible cellular damage. Shikonin effectively attenuates Rot-induced apoptosis

Glucagon-like Peptide-1 Boosts Plumbagin's Neuroprotection Against Rotenone-Induced Motor Deficits.

INTRODUCTION: Parkinson's disease (PD), a condition that involves neural degeneration, develops due to dopaminergic neuronal death in the substantia nigra pars compacta, resulting in reduced striatal dopamine levels. This shortage causes problems with movement and thinking. The neurodefensive response of GLP-1 is especially important in PD. Assessments have demonstrated the neuroprotective advantages of activating GLP-1 receptors in distinct models of PD, resulting in enhancements in motor as well as non-motor behaviour. These characteristics suggest that GLP-1 signalling could be a promising

Neuroprotective impacts of taurine nanoparticles against rotenone induced Parkinson's disease in mice.

Parkinson's disease (PD) is a progressive neurodegenerative disorder affects motor and cognitive functions in patients. The main pathology of this illness is the loss of dopaminergic neurons in the substantia nigra which leads to locomotor impairment such as tremors, bradykinesia, and muscular rigidity. In late stages of PD, non-motor symptoms like cognitive deficits develop, affecting the life quality. Recent studies indicated that these non-motor symptoms are attributed to neuronal loss in the hippocampus and impaired neurotransmission, due to oxidative stress and neuroinflammation. This stu

Rotenone-Induced Optic Nerve Damage and Retinal Ganglion Cell Loss in Rats.

Rotenone is a mitochondrial complex I inhibitor that causes retinal degeneration. A study of a rat model of rotenone-induced retinal degeneration suggested that this model is caused by indirect postsynaptic N-methyl-D-aspartate (NMDA) stimulation triggered by oxidative stress-mediated presynaptic intracellular calcium signaling. To elucidate the mechanisms by which rotenone causes axonal degeneration, we investigated morphological changes in optic nerves and the change in retinal ganglion cell (RGC) number in rats. Optic nerves and retinas were collected 3 and 7 days after the intravitreal inj

Pharmacological evaluation of vanillic acid in rotenone-induced Parkinson's disease rat model.

In the present study, we investigated the anti-Parkinson's effect of vanillic acid (VA) (12 mg/kg, 25 mg/kg, 50 mg/kg p.o.) against rotenone (2 mg/kg s.c.) induced Parkinson's disease (PD) in rats. The continuous administration of rotenone for 35 days resulted in rigidity in muscles, catalepsy, and decrease in locomotor activity, body weight, and rearing behaviour along with the generation of oxidative stress in the brain (rise in the TBARS, and SAG level and reduced CAT, and GSH levels). Co-treatment of VA and levodopa-carbidopa (100 mg/kg + 25 mg/kg p.o.) lead to a significant (P < 0.001) re

Coordinated Action of miR-146a and Parkin Gene Regulate Rotenone-induced Neurodegeneration.

Mitochondrial dysfunction is a common cause in pathophysiology of different neurodegenerative diseases. Elimination of dysfunctional and damaged mitochondria is a key requirement for maintaining homeostasis and bioenergetics of degenerating neurons. Using global microRNA (miRNA) profiling in a systemic rotenone model of Parkinson's disease, we have identified miR-146a as upmost-regulated miRNA, which is known as inflammation regulatory miRNA. Here, we report the role of activated nuclear factor kappa beta (NF-kβ) in miR-146a-mediated downregulation of Parkin protein, which inhibits clearance o

Rotenone protects against β-cell apoptosis and attenuates type 1 diabetes mellitus.

Type 1 diabetes mellitus (T1DM) is caused by pancreatic β-cell dysfunction and apoptosis, with consequent severe insulin deficiency. Thus, β-cell protection may be a primary target in the treatment of T1DM. Evidence has demonstrated that defective mitochondrial function plays an important role in pancreatic β-cell dysfunction and apoptosis; however, the fundamental effect of mitochondrial complex I action on β-cells and T1DM remains unclear. In the current study, the pancreas protective effect of complex I inhibitor rotenone (ROT) and its potential mechanism were assessed in a streptozotocin (

Parkinson's disease-like motor and non-motor symptoms in rotenone-treated zebrafish.

The pesticide rotenone is widely used to produce Parkinson's disease (PD)-like symptoms in rodents, but few studies have examined whether rotenone-treated zebrafish can serve as an animal model of PD. Here, we report that 4 weeks of rotenone treatment induced motor and non-motor PD-like symptoms in adult zebrafish. Compared with control fish, rotenone-treated fish spent less time swimming at a fast speed, indicating a deficit in motor function. In the light-dark box test, rotenone-treated fish exhibited longer latencies to enter the dark compartment and spent more time in the light compartment

Sestrin2 Protects Dopaminergic Cells against Rotenone Toxicity through AMPK-Dependent Autophagy Activation.

Dysfunction of the autophagy-lysosomal pathway (ALP) and the ubiquitin-proteasome system (UPS) was thought to be an important pathogenic mechanism in synuclein pathology and Parkinson's disease (PD). In the present study, we investigated the role of sestrin2 in autophagic degradation of α-synuclein and preservation of cell viability in a rotenone-induced cellular model of PD. We speculated that AMP-activated protein kinase (AMPK) was involved in regulation of autophagy and protection of dopaminergic cells against rotenone toxicity by sestrin2. The results showed that both the mRNA and protein

Neuroprotective effects of metallothionein against rotenone-induced myenteric neurodegeneration in parkinsonian mice.

Parkinson's disease (PD) is a neurodegenerative disease with motor symptoms as well as non-motor symptoms that precede the onset of motor symptoms. Mitochondrial complex I inhibitor, rotenone, has been widely used to reproduce PD pathology in the central nervous system (CNS) and enteric nervous system (ENS). We reported previously that metallothioneins (MTs) released from astrocytes can protect dopaminergic neurons against oxidative stress. The present study examined the changes in MT expression by chronic systemic rotenone administration in the striatum and colonic myenteric plexus of C57BL m

Dopamine and Rotenone Modulate α-Synuclein Phase Separation and Liquid to Solid Transition.

Liquid-liquid phase separation (LLPS) of α-Synuclein (α-Syn) is recognized as an early biophysical event driving pathological aggregation in Parkinson's disease (PD). Although 10%-15% of PD cases are due to familial mutations, the remaining cases are sporadic, often linked to various factors, like pesticides and metals. For example, rotenone and dopamine are known to be involved in PD pathology and are suggested to cause changes in α-Syn protein homeostasis, although the mechanism by which they influence α-Syn and cellular toxicity is largely unknown. In this work, we demonstrate that both dop

H2S protects hippocampal HT22 neurons against rotenone-induced ferroptosis by upregulating Isthmin 1.

BACKGROUND: Hydrogen sulfide (H2S) has been recognized for its potent neuroprotective effects in Parkinson's disease (PD) models, yet the underlying cellular and molecular mechanisms remain to be fully elucidated. Ferroptosis, an iron-dependent form of regulated cell death, is increasingly implicated in the pathogenesis of PD. Isthmin 1 (ISM1) is a secreted protein predominantly expressed in the brain with emerging roles in neuroprotection and metabolic regulation. This study aimed to investigate whether H2S protects against rotenone (ROT)-induced neuronal injury by modulating ferroptosis and

Parkinsonism Reversal and Dopaminergic Resilience: Lessons from a Rotenone-induced Parkinson's Disease Model.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by profound loss of dopaminergic (DA) neurons, yet the underlying mechanism remains incompletely defined. Mitochondrial toxins can induce acute degeneration of DA neurons and Parkinsonism-like phenotypes in animal models, and epidemiological studies have linked pesticide exposure to increased PD risk; however, the long-term effects of pesticide exposure remain elusive. Here, we examined both the acute and long-term effects of rotenone exposure in mice to understand PD onset, progression, and recovery. A 21-day r

Hydrogen Sulfide Inhibits Ferritinophagy-Mediated Ferroptosis in the Hippocampus of Rotenone-Exposed Rats.

Our previous research has established that hydrogen sulfide (H 2 S) exerts an antagonistic effect against the hippocampal neurotoxicity induced by Rotenone (ROT). However, the underlying mechanisms are so far poorly understood. Substantial evidence corroborates the involvement of ferroptosis in ROT-induced neurotoxicity. To elucidate the protective mechanism of H 2 S against ROT-induced hippocampal neurotoxicity, this study explores its regulatory role in ferroptosis and its underlying mechanisms. We used Fluoro-Jade B staining to detect dead neurons. The levels of ferrous ions and glutathione

N-acetylcysteine modulates rotenone-induced mitochondrial Complex I dysfunction in THP-1 cells.

Mitochondrial Complex I dysfunction and oxidative stress have been part of the pathophysiology of several diseases ranging from mitochondrial disease to chronic diseases such as diabetes, mood disorders and Parkinson's Disease. Nonetheless, to investigate the potential of mitochondria-targeted therapeutic strategies for these conditions, there is a need further our understanding on how cells respond and adapt in the presence of Complex I dysfunction. In this study, we used low doses of rotenone, a classical inhibitor of mitochondrial complex I, to mimic peripheral mitochondrial dysfunction in
