Brainstem Neural Circuits Triggering Vertical Saccades and Fixation.

Neurons in the nucleus raphe interpositus have tonic activity that suppresses saccadic burst neurons (BNs) during eye fixations, and that is inhibited before and during saccades in all directions (omnipause neurons, OPNs). We have previously demonstrated via intracellular recording and anatomical staining in anesthetized cats of both sexes that OPNs are inhibited by BNs in the medullary reticular formation (horizontal inhibitory BNs, IBNs). These horizontal IBNs receive monosynaptic input from the caudal horizontal saccade area of the superior colliculus (SC), and then produce monosynaptic inh

Freezing of saccades in dopa-responsive parkinsonian syndrome.

PURPOSE: Ocular motor abnormalities such as abnormal saccades are common in idiopathic Parkinson's disease (PD) and atypical parkinsonian syndrome, such as progressive supranuclear palsy (PSP) and corticobasal degeneration (CBD). In this study, we describe a case of patient with PD and show a video illustrating severe delay of reflexive saccades. OBSERVATIONS: A 68-year-old Caucasian woman with diagnosis of PD presented for evaluation of diplopia. Neuro-ophthalmic examination revealed good visual acuity in both eyes and normal optic nerves but prominent ocular motor abnormalities, including h

Rapid spatial oculomotor updating across saccades is malleable.

The oculomotor system uses a sophisticated updating mechanism to adjust for large retinal displacements which occur with every saccade. Previous studies have shown that updating operates rapidly and starts before saccade is initiated. Here we used saccade adaptation to alter life-long expectations about how a saccade changes the location of an object on the retina. Participants made a sequence of one horizontal and one vertical saccade and ignored an irrelevant distractor. The time-course of oculomotor updating was estimated using saccade curvature of the vertical saccade, relative to the dist

Saccades and handedness interact to affect scene memory.

Repetitive saccades benefit memory when executed before retrieval, with greatest effects for episodic memory in consistent-handers. Questions remain including how saccades affect scene memory, an important visual component of episodic memory. The present study tested how repetitive saccades affect working and recognition memory for novel scenes. Handedness direction (left-right) and degree (strong/consistent vs. mixed/inconsistent) was measured by raw and absolute laterality quotients respectively from an 8-question handedness inventory completed by 111 adults. Each then performed either 30 s

Slow saccades in bulbar-onset motor neurone disease.

Historical studies of eye movements in motor neurone disease (MND) have been conflicting although current findings suggest that eye movement abnormalities relate to frontal lobe impairment. Numerous case reports, however, describe slow saccades and supranuclear gaze palsies in patients with MND often associated with bulbar-onset disease. We performed a study of saccades and smooth pursuit in a large group of patients with MND to examine for any differences between bulbar-onset and spinal-onset patients. Forty-four patients (14 bulbar-onset and 30 spinal-onset patients) and 45 controls were rec

Poor binocular coordination of saccades in dyslexic children.

AIM: To examine the quality of binocular coordination of saccades in dyslexic children in single word reading and in a task requiring fixation of single LED. METHODS: Eighteen children with dyslexia (11.4 +/- 2 years old) and 13 non-dyslexic children of matched age were studied. Horizontal saccades from both eyes were recorded with a photoelectric system (Oculomotor-Bouis). RESULTS: Binocular coordination during and after the saccade in dyslexics is worse than that of non-dyslexic children; the disconjugacy does not depend on the condition. Moreover, dyslexics do not show the stereotyped pat

Saccadic intrusions in paediatric concussion.

Three paediatric patients post-mild traumatic brain injury (mTBI, also known as concussion) developed saccadic intrusions, which are irregular, involuntary saccades that interrupt fixation. Each patient presented to a mTBI clinic months post-injury, complaining of headaches, difficulty concentrating, and dizziness. Written informed consent from parents was obtained for use of these cases.

Saccadic fatigability in the oculomotor system.

BACKGROUND: Fatigue is one of the most common and disabling symptoms in multiple sclerosis (MS), but challenging to quantify. This prospective study investigated if repeated saccadic eye movements enable measurement of oculomotor fatigability and can reflect on perceived fatigue in MS. METHODS: A standardized infrared oculography protocol (DEMoNS) was used for quantifying saccades in MS patients and healthy controls which included a first and a repeated pro-saccadic task (FPT and RPT). Saccadic peak velocity, latency, gain, area under the curve (AUC) and peak velocity divided by amplitude (Pv

Saccadic trajectory in Huntington's disease.

Trajectories of saccadic eye movements can be modulated by the presence of a competing visual distractor. It is proposed that the superior colliculus (SC) controls the initial deviation through competitive lateral interactions. Given the ramifications of connections between basal ganglia (BG) thalamo-cortical circuitry and the SC, it was anticipated that this modulation would be differentially effected in those with Huntington's disease, which in its early stages is primarily a disorder of the BG. Horizontal deviation was determined for exogenously driven and endogenously driven vertical sacca

Voluntary saccadic control in dyslexia.

The role of eye-movement control in dyslexia is still unclear. Recent studies, however, confirmed that dyslexics show poor saccadic control in single and sequential target tasks. In the present study we investigated whether dyslexic subjects are impaired on an antisaccade task requiring saccades against the direction of a stimulus. Altogether, 620 subjects between the ages of 7 and 17 years were classified as dyslexics (N = 506) or control subjects (N = 114) on the grounds of the discrepancy between their intellectual abilities and reading/spelling achievements. All subjects performed an overl

Saccadic intrusions in internuclear ophthalmoplegia.

Saccadic intrusions disrupted forward fixation in four patients with bilateral internuclear ophthalmoplegia (INO). Three had multiple sclerosis and one had brainstem infarction. Sporadic bursts of monocular abducting saccades were recorded in either eye. Each saccade was followed by a slower return to midposition. The presence of the INO prevented conjugate saccades of the fellow eye. The adducting phases were faster than convergence eye movements or the glissadic smooth eye movements that sometimes follow saccades. Dynamic overshoot of the abducting saccadic phase may contribute to the relati

Saccadic velocity measurements in strabismus.

Traditional evaluation of strabismus has included cover test measurements, evaluation of the range of ocular rotations, and an array of subjective sensory tests. These studies could not always differentiate paresis of an extraocular muscle from restrictions and from various neuro-ophthalmic motility disorders. The measurement of horizontal and vertical saccadic movements can provide an objective test of rectus muscle function. Using EOG, saccades can be recorded easily, inexpensively, and repeatably at any age. In ocular muscle paresis or paralysis, saccadic speed is reduced mildly to markedly

Ipsilateral Saccade Hypometria and Contralateral Saccadic Pursuit in a Focal Brainstem Lesion: a Rare Oculomotor Pattern.

Eye movement examination may be used to rapidly differentiate peripheral and central vestibular syndromes in patients with acute unsteadiness. The analysis of oculomotor impairments may also support the accurate localization of cerebral lesions, particularly those in the brainstem, that are often loosely defined by cerebral MRIs. Saccades, smooth pursuit, and nystagmus were recorded with video-oculography in a patient who had developed sudden vertigo as a consequence of a focal lesion in the depth of the brachium pontis. The patient had shown a previously unreported pattern of eye movement imp

Direct gaze facilitates rapid orienting to faces: Evidence from express saccades and saccadic potentials.

Direct gaze is a crucial signal in human social communication, which is known to attract visual attention and modulate a wide range of behaviours. The present study investigated whether direct gaze facilitates rapid orienting to faces, which is important for adaptive on-line communication, and its neural correlates. Fifteen participants performed a rapid orienting task, in which they were instructed to saccade to peripherally presented buildings or faces containing direct or averted gaze as quickly as possible. Electroencephalographic recordings were made during the task. Shorter express sacca

Abnormal saccade profiles in hereditary spinocerebellar degeneration reveal cerebellar contribution to visually guided saccades.

OBJECTIVE: To study how the pathophysiology underlying hereditary spinocerebellar degeneration (spinocerebellar ataxia; SCA) with pure cerebellar manifestation evolves with disease progression using saccade recordings. METHODS: We recorded visually- (VGS) and memory-guided saccade (MGS) task performance in a homogeneous population of 20 genetically proven SCA patients (12 SCA6 and eight SCA31 patients) and 19 normal controls. RESULTS: For VGS but not MGS, saccade latency and amplitude were increased and more variable than those in normal subjects, which correlated with cerebellar symptom sev

The lateral intraparietal area codes the location of saccade targets and not the dimension of the saccades that will be made to acquire them.

Activity in the lateral intraparietal area (LIP) represents a priority map that can be used to direct attention and guide eye movements. However, it is not known whether this activity represents the location of saccade targets or the actual eye movement made to acquire them. We recorded single neurons from rhesus macaques (Macaca mulatta) while they performed memory-guided delayed saccades to characterize the response profiles of LIP cells. We then separated the saccade target from the saccade end point by saccadic adaptation, a method that induces a change in the gain of the oculomotor system

Characteristics of horizontal saccades in children with intermittent exotropia.

PURPOSE: To analyze the peak velocity and gain of horizontal saccades in children diagnosed with intermittent exotropia (IXT) and to compare these measurements with those obtained from children without strabismus. METHODS: Participants included children ≤12 years of age with IXT and no history of strabismus surgery and children without strabismus. The peak velocity and gain of horizontal saccades recorded using an eye tracker were analyzed. RESULTS: A total of 48 children (including 38 with IXT) were included. In the IXT group, the angles of deviation at near were 30.3Δ ± 13.5Δ; at distance,

Cortical functional anatomy of voluntary saccades in Parkinson disease.

In Parkinson Disease (PD) several aspects of saccades are affected. The saccade-generating brainstem neurons are spared, however, the signals they receive may be flawed. In particular voluntary saccades suffer, but the functional anatomy of the impairment of saccade-related cortical control is unknown. We measured blood-oxygenation-level-dependent (BOLD) activation with functional Magnetic Resonance Imaging (fMRI) while healthy participants and patients with PD performed horizontal voluntary saccades between peripheral visual targets or fixated centrally. We compared saccade-related BOLD-activ

Motor neuron disease presenting with slow saccades and dementia.

A 46-year-old man with a 2-year history of dementia presented with very slow saccades and tongue fasciculations. The diagnosis of amyotrophic lateral sclerosis (ALS) was established by electrophysiologic studies and ultimately on autopsy. Rarely, slowed saccades may be present in ALS, and slow saccades in a relatively young demented patient should prompt consideration of ALS.
