Molecular differentiation of Sarcocystis miescheriana and Sarcocystis suihominis using a new multiplex PCR targeting the mtDNA cox1 gene in wild boars in southern Italy.

The increase of wild boar populations density and their meat consumption across Europe could expose humans to a plethora of foodborne diseases as sarcocystosis, caused by the zoonotic protozoan Sarcocystis suihominis. Humans become infected by eating raw or undercooked pig (Sus scrofa domesticus) containing S. suihominis sarcocysts. Despite this, to date very few data are available on the risk of infection by this parasite to wild boar (Sus scrofa) meat consumers. Thus, the present study aimed to assess the occurrence of Sarcocystis spp. in wild boars from southern Italy, applying both histolo…

First molecular detection of Sarcocystis spp. in sheep from Aguascalientes, Mexico.

Sarcocystis spp. are widely distributed apicomplexan protozoa that infect sheep and other domestic animals. In Mexico, ovine sarcocystosis has been documented mainly through gross inspection and histopathology, while region-specific molecular data remain limited. In this study, we provide the first PCR-based molecular evidence of Sarcocystis spp. in sheep from Aguascalientes, Mexico. We processed 123 tissue samples (heart, esophagus, and liver) collected from 41 sheep slaughtered for human consumption. Parasite DNA was detected in 25/41 animals (60.9%). At the tissue level, PCR positivity was …

Sarcocystis calchasi Outbreak in Feral Rock Pigeons ( Columba livia) in California.

Twenty-two feral rock pigeons ( Columba livia) from 10 counties in California with ataxia, torticollis, and difficulty standing and flying w … smic bradyzoites typically without inflammation. Fifteen of the 22 birds tested positive using pan- Sarcocystis polymerase chain reaction. The sequence of the amplicon was most closely related to S. calchasi, and the 8 subtyped sequences had 100% homology with S. calchasi. This investigation demonstrated the transcontinental and North American spread of S. ca …

Genome-Wide Identification and Evolutionary Analysis of Sarcocystis neurona Protein Kinases.

The apicomplexan parasite Sarcocystis neurona causes equine protozoal myeloencephalitis (EPM), a degenerative neurological disease of horses. Due to its host range expansion, S. neurona is an emerging threat that requires close monitoring. In apicomplexans, protein kinases (PKs) have been implicated in a myriad of critical functions, such as host cell invasion, cell cycle progression and host immune response evasion. Here, we used various bioinformatics methods to define the kinome of S. neurona and phylogenetic relatedness of its PKs to other apicomplexans. We identified 97 putative PKs clust…

Clinical Sarcocystis neurona encephalomyelitis in a domestic cat following routine surgery.

Sarcocystis neurona is an important cause of equine protozoal myeloencephalitis (EPM) in horses in the Americas. An EPM-like neurological disease also has been reported from other mammals but it is difficult to induce this disease in the laboratory. A 4-month-old male domestic cat developed neurological signs 3 days following castration. The cat was euthanized 12 days later because of paralysis. Encephalomyelitis was the only lesion and was associated with numerous Sarcocystis schizonts and merozoites in the brain and spinal cord. The protozoa reacted positively with S. neurona-specific polycl…

Fatal Sarcocystis calchasi-associated meningoencephalitis in 2 captive vulturine guineafowl.

Two captive vulturine guineafowl (Acryllium vulturinum) were presented with lethargy, hyporexia, weight loss, and progressive neurologic signs. One of the guineafowl was seropositive for Sarcocystis falcatula (1:50 dilution). Both guineafowl died within 5 d of presentation. Histologic examination revealed nonsuppurative meningoencephalitis with gliosis, associated with occasional schizonts in the neuropil. Using fresh-frozen brain tissue, PCR was performed to amplify the ITS1 RNA region and portions of the 18S ribosomal RNA gene (18S gene) and the 28S ribosomal RNA gene (28S gene). Analysis of…

SARCOCYSTIS INFECTIONS IN TONGUES OF COUGARS (PUMA CONCOLOR COUGUAR) IN COLORADO.

Sarcocystis species are intracellular protozoan parasites that infect a wide range of intermediate and definitive hosts, including felids. This study reports Sarcocystis species in the tongues of cougars (Puma concolor couguar) in Colorado for the first time. By light microscope, sarcocysts were found in 4 of 30 cougars; they appeared to represent more than 2 morphologic types by light microscopy. One morphologic type had serrations on the sarcocyst wall and resembled Sarcocystis felis reported from domestic and wild felids. In the second type, serrations were not visible on the sarcocyst wall…

Possibility of Vertical Transmission of Sarcocystis spp. in Sika Deer in Japan.

In recent years, the wild deer population in Japan has grown exponentially, causing severe feeding damage to the agricultural and forestry industries. Therefore, the game meat industry is being promoted for effective utilization of hunted animals. Wild animals are not hygienically controlled and can serve as reservoirs for pathogenic microorganisms. However, epidemiological information on wild animals in Japan remains insufficient. Recently, food poisoning-like cases have occurred because of raw venison infection with Sarcocystis spp. As the prevalence of Sarcocystis spp. in sika deer is very …

The present status of Sarcocystis spp. and sarcocystosis in Iran: a literature review.

Sarcocystosis due to Sarcocystis species, is prevalent among livestock in most parts of Iran, the predominant species being S. cruzi, and then followed by S. hirsuta and S. hominis. Studies on the prevalence and geographic distribution pattern of infection in the most common sites of infection have revealed infection rates up to 100% in at least one of the examined tissues in the country. Although human intestinal and gallbladder Sarcocystis infections have been reported, nothing is known about muscular Sarcocystis infection in human in Iran. The main aim of this review is to estimate the prev…

Toxoplasmosis and Sarcocystis spp. infection in wild pinnipeds of the Brazilian coast.

The protozoans Toxoplasma gondii and Sarcocystis spp. (Sarcocystidae: Apicomplexa) affect a wide variety of vertebrates. Both have been reported to infect pinnipeds, with impacts on health ranging from inapparent to fulminant disease and death. However, little is known regarding their infections and associated pathology in South American pinnipeds. We used histological techniques to survey for the presence of T. gondii and Sarcocystis spp. in 51 stranded pinnipeds from Brazil. Immunohistochemical and molecular assays were employed in those cases consistent with Sarcocystidae infection. T. gond…

Fatal Sarcocystis cruzi-induced eosinophilic myocarditis in a heifer in Uruguay.

Sarcocystis spp. are causative agents of bovine eosinophilic myositis and/or myocarditis, which are chronic subclinical myopathies that are occasionally responsible for condemnation at slaughterhouses. Sarcocystis cruzi is a protozoan parasite of worldwide distribution transmitted by canids, most commonly associated with subclinical infection in cattle. Although S. cruzi infections can rarely lead to fatal systemic disease, fatal cardiac cases with confirmation of the etiologic diagnosis have not been reported, to our knowledge. We describe herein an unusual case of S. cruzi-induced fatal bovi…

First findings of Sarcocystis species in game deer and feral pigs in Australia.

Several wild game meat species, including deer and feral pigs are hunted and consumed in Australia. Feral pigs and deer are not indigenous to Australia, but they have proliferated extensively and established their presence in every state and territory. Following the report of a sambar deer displaying Sarcocystis like white cysts in its rump muscles, the present study was conducted to explore the prevalence of Sarcocystis infections in wild deer and feral pigs in the southeastern regions of Australia. Oesophagus, diaphragm, and heart tissue from 90 deer and eight feral pigs were examined visual…
