Antibiotic resistance of lactic acid bacteria isolated from Chinese yogurts.

The aim of this study was to evaluate the susceptibility of 43 strains of lactic acid bacteria, isolated from Chinese yogurts made in different geographical areas, to 11 antibiotics (ampicillin, penicillin G, roxithromycin, chloramphenicol, tetracycline, chlortetracycline, lincomycin, kanamycin, streptomycin, neomycin, and gentamycin). The 43 isolates (18 Lactobacillus bulgaricus and 25 Streptococcus thermophilus) were identified at species level and were typed by random amplified polymorphic DNA analysis. Thirty-five genotypically different strains were detected and their antimicrobial resist…

Safety assessment of two strains and anti-obese effects on mice fed a high-cholesterol diet.

Previous study documented that Lactobacillus paracasei S0940 and Streptococcus thermophilus ldbm1 have obvious cholesterol-lowering abilities in vitro. In this study, the safety of two strains were evaluated by nitroreductase test, hemolysis test and antibiotic sensitivity test and to evaluate the cholesterol-reducing abilities in vivo. The results indicated that two strains did not exhibit nitroreductase activities and were ɤ-hemolytic on blood agar plates. Further, both strains did not represent a health risk by antibiotic sensitivity test, and significantly reduced serum and liver cholester…

Sunflower wax-structured oil phase confers robustness to W/O/W emulsion gel beads for enhanced probiotic viability.

This study explored impact of natural waxes (beeswax, BW; rice bran wax, RBW; sunflower wax, SFW)-mediated oil-phase gelling on the physicochemical properties and probiotic-protective efficacy of W/O/W emulsion gel beads co-encapsulating Lactobacillus plantarum and Streptococcus thermophilus. Wax incorporation preserved "two-membrane, three-phase" emulsion architecture while increasing droplet sizes and viscosity, with SFW and RBW yielding denser, more uniform bead microstructures and reduced moisture mobility compared to controls (without wax, NW) and BW. XRD and DSC revealed β'-polymorphic c…

Temporal dynamics of the S. thermophilus phage population in a yogurt production facility.

Starter cultures are essential for milk fermentation, but phage infections can disrupt production, compromising product quality and causing economic losses. In this study, we investigated the dynamics, diversity, and persistence of 25 S. thermophilus phages isolated over a 15-year period from a large yogurt manufacturing plant. Phages were classified into Moineauvirus (16) and Brussowvirus (9), comprising ten distinct restriction profiles. Host range, genomic features, and resistance to thermal and chemical treatments were analyzed to identify factors underlying their long-term persistence. No

Curcumin-loaded extracellular vesicles derived from lactic acid bacteria for alleviation of inflammatory bowel disease.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder characterized by symptoms such as diarrhea, bleeding, and an elevated risk of colorectal cancer. Bacterial extracellular vesicles (EVs) have emerged as a promising delivery system for bioactive compounds, owing to their high biocompatibility, low immunogenicity, and cost-effectiveness. In this study, EVs were isolated from two Lactic acid bacteria (LAB)-Lactobacillus delbrueckii ssp. Bulgaricus and Streptococcus thermophilus, and curcumin was successfully encapsulated into these vesicles. Compared with those derived from S. th…

A phage-encoded anti-CRISPR protein co-opts host enolase to prevent type III CRISPR immunity.

CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) systems provide powerful adaptive immunity against phage infection. In response, phages use anti-CRISPR (Acr) proteins to evade CRISPR immunity. The few type III Acrs identified so far show conditional effectiveness in countering type III immunity or rely on unknown or poorly understood inhibitory mechanisms. Here we report the discovery of AcrIIIA2, a type III-A Acr encoded by Streptococcus thermophilus phages. Biochemical and structural analyses reveal that phage AcrIIIA2 co-opts host enolase, a highly abundant glycolysis enz…

Should yoghurt cultures be considered probiotic?

Probiotics are live micro-organisms that when administered in adequate amounts confer a health benefit on the host. Consumption of yoghurt has been shown to induce measurable health benefits linked to the presence of live bacteria. A number of human studies have clearly demonstrated that yoghurt containing viable bacteria (Streptococcus thermophilus and Lactobacillus delbrueckii sp. bulgaricus) improves lactose digestion and eliminates symptoms of lactose intolerance. Thus, these cultures clearly fulfil the current concept of probiotics.

Integrating Bacteriocins and Biofilm-Degrading Enzymes to Eliminate L. monocytogenes Persistence.

Listeria monocytogenes is a Gram-positive bacterium causing listeriosis, a severe infection responsible for significant morbidity and mortality globally. Its persistence on food processing surfaces via biofilm formation presents a major challenge, as conventional sanitizers and antimicrobials exhibit limited efficacy against biofilm-embedded cells. This study investigates a novel approach combining an engineered polysaccharide-degrading enzyme (CAase) with a bacteriocin (thermophilin 110) produced by Streptococcus thermophilus. Laboratory assays evaluated the effectiveness of this combination …
