Retrograde transduction of dopaminergic cells in substantia nigra of rhesus monkey.

Neuromodulatory systems regulate neural circuits across broad regions of the brain, and disruption of the dopaminergic system contributes to … he neuronal co-expression of fluorescent reporter protein and tyrosine-hydroxylase (TH) antibody in substantia nigra pars compacta (SNc). The rabies pseudotyped lentiviral vector, FuG-B2, produced superior retrograde transduction of DA cells to FuG-C or FuG-E. AAV2.retro was the least effective.

Neuromelanin Magnetic Resonance Imaging of the Substantia Nigra in Huntington's Disease.

BACKGROUND: Huntington's disease (HD) is an autosomal-dominant neurodegenerative disorder inducing motor, psychiatric changes and cognitive decline, characterized pathologically by striatal atrophy. Pathological changes in the extra-striatal structures, such as the substantia nigra (SN), and abnormalities in pre-synaptic striatal dopamine neurotransmission are also known to occur. Neuromelanin (NM)-sensitive magnetic resonance imaging (NM-MRI) is an innovative technique that was recently developed allowing the in vivo study of pathological changes in the dopaminergic neurons of the SN. OBJECT…

Complement activation in the Parkinson's disease substantia nigra: an immunocytochemical study.

BACKGROUND: Inflammatory processes are increased in the Parkinson's disease (PD) brain. The long-term use of nonsteroidal anti-inflammatory drugs has been associated, in retrospective studies, with decreased risk for PD, suggesting that inflammation may contribute to development of this disorder. The objective of this study was to determine the extent of complement activation, a major inflammatory mechanism, in PD. METHODS: Substantia nigra specimens from young normal subjects (n = 11-13), aged normal subjects (n = 24-28), and subjects with PD (n = 19-20), Alzheimer's disease (AD; n = 12-13),…

The actions of antipsychotic drugs on dopamine receptors in the rat substantia nigra.

The activity of neurones in the zona compacta of the rat substantia nigra was recorded extracellularly in vitro. Dopamine produced a dose-dependent depression of firing, threshold doses being in the 3 microM range. The inhibitory effects of dopamine were antagonized by (-)-sulpiride (pA2 7.5), haloperidol (pA2 8.4) and cis-flupenthixol (pA2 6.9). The actions of gamma-aminobutyric acid (GABA) were not affected by these compounds. The gradients of Schild plots of data for (-)-sulpiride were less than unity while those for haloperidol and cis-flupenthixol were greater than unity, which suggests t…

Iron Depletion in the Substantia Nigra of Children With Prenatal Alcohol Exposure.

Prenatal alcohol exposure (PAE) occurs in 10%-60% of pregnancies and can contribute to fetal alcohol spectrum disorders (FASD). FASD present … ge and sex. Compared with unexposed children, those with any PAE had significantly lower QSM in the substantia nigra (β = -18.93, p = 0.011). Stratified analyses revealed that substantia nigra QSM was lower even after first-trimester-restricted exposure (T1 β = -22.63 95% CI [-37.35, -7.91]; p = 0.004). Cortical analyses showed regionally variable alterations, wit …

A neuroimaging atlas of the nigrosomes in the substantia nigra based on 3D histology.

Nigrosomes are formed by clusters of pigmented dopaminergic cells in the substantia nigra that critically contribute to dopaminergic function. The ever-increasing resolution of ultra-high-field MRI brings clinical imaging of these clusters into reach, promising unprecedented insight into the functional role of the nigrosomes and their early degeneration in Parkinson's disease. However, due to the nigrosomes' small extents and intricate shapes, they are not included in current MRI brain atlases, preventing nigrosome-specific MRI data analysis. We provide a comprehensive 3D histological atlas of…

DOPAnization of α-synuclein exacerbates dopaminergic neurodegeneration in the substantia nigra.

Parkinson's disease (PD) is characterized by the selective and progressive loss of dopaminergic (DAergic) neurons in the substantia nigra (SN). Although abnormal forms and aggregates of the α-synuclein (αSyn) protein are considered to cause PD, the underlying mechanisms of the preferential death of dopaminergic neurons are largely unknown. We recently reported that a tyrosine hydroxylase post-translationally converted a tyrosine residue of αSyn to dihydroxyphenylalanine (DOPA), termed DOPAnization, in the DAergic neurons of PD patients, which facilitated the formation of αSyn oligomers and inc…

Variability in neuronal expression of dopamine receptors and transporters in the substantia nigra.

Parkinson's disease (PD) patients have increased susceptibility to impulse control disorders. Recent studies have suggested that alterations in dopamine receptors in the midbrain underlie impulsive behaviors and that more impulsive individuals, including patients with PD, exhibit increased occupancy of their midbrain dopamine receptors. The cellular location of dopamine receptor subtypes and transporters within the human midbrain may therefore have important implications for the development of impulse control disorders in PD. The localization of the dopamine receptors (D1-D5) and dopamine tran

Structural abnormalities in the substantia nigra and neighbouring nuclei in Tourette's syndrome.

Although midbrain nuclei (substantia nigra, ventral tegmental area, and periaqueductal grey) are considered candidate loci of pathology in Tourette's syndrome (TS), few imaging studies have examined midbrain structure. The objective of this study was to evaluate the presence of subtle structural abnormalities in the midbrain of patients with TS. High-field magnetic resonance imaging (MRI) (1.5- and 3-T) was used in 23 patients with TS and in 20 age- and sex-matched normal control subjects. Tics symptoms were rated using the Yale Global Tic Severity Scale and comorbid neuropsychiatric disorders…

LPS-induced degeneration of dopaminergic neurons of substantia nigra in rats.

In order to investigate the neurotoxicity of lipopolysaccharide (LPS) on the dopaminergic neurons of substantia nigra and the pathogenesis of Parkinson disease, LPS was stereotaxically infused into substantia nigra (SN). At different dosages and different time points with 5 microg LPS, the damage of the dopaminergic neurons in SN was observed by using tyrosine-hydroxylase (TH) immunohistochemical staining. The results showed that 14 days after injection of 0.1 microg to 10 microg LPS into the rat SN, TH-positive (TH+) neurons in the SN were decreased by 5%, 15%, 20%, 45 %, 96% and 99% respecti…

Influence of neuromelanin on oxidative pathways within the human substantia nigra.

Neuromelanin (NM) is a dark-coloured pigment produced in the dopaminergic neurons of the human substantia nigra (SN). The function of NM within the pigmented neurons is unknown but other melanins are believed to play a protective role via attenuation of free radical damage. Experimental evidence suggests that NM may also exhibit this characteristic, possibly by directly inactivating free radical species or via its ability to chelate transition metals, such as iron. Increased tissue iron, however, may saturate iron-chelating sites on NM and a looser association between iron and NM may result in…

Levodopa prolongs life expectancy and is non-toxic to substantia nigra.

The primary objective of the study was to determine the effect of levodopa (LD) on human substantia nigra. The study included patients seen at the Movement Disorder Clinic, Saskatoon over a 32 year period. The evidence provided is based on epidemiological observations of 934 consecutive Parkinson syndrome (PS) patients assessed during 22 years and detailed studies of six patients including two autopsies. Life expectancy increased significantly with LD therapy. The prolonged survival was evident when the patients were treated during early stage of the illness. One parkinsonian patient with subs…

MPTP induces alpha-synuclein aggregation in the substantia nigra of baboons.

1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) neurotoxicity reproduces many of the features of Parkinson's disease (PD). alpha-Synuclein has been identified as a prominent component of the Lewy body (LB), the pathological hallmark of PD. MPTP-treated primates have been reported to develop intraneuronal inclusions but not true Lewy bodies. We administered MPTP to baboons and used a monoclonal alpha-synuclein antibody to define the relationship between neuronal degeneration and alpha-synuclein immunoreactivity in the substantia nigra. MPTP-induced neuronal degeneration was associated with …
