The incidence of anogenital warts (condyloma acuminatum) is rapidly increasing while there is still no totally satisfactory treatment available. In light of the emphasis of experimental approaches toward the prevention of viral replication and evidence of the antiviral action of lithium salts it was proposed to investigate the efficacy of Topical Lithium Succinate cream (LSC) in the treatment of anogenital warts. A total of 101 patients (42 women, 59 men) were randomized to receive either active or placebo treatment for a period of 4 weeks. Assessment of the number, location, size and area of

For more effective chemotherapy and targeted treatment of colorectal cancer, this study seeks to develop chitosan (CH)-human serum albumin (HAS)-D-α-tocopheryl polyethylene glycol 1000 (TPGS) nanoparticles (BOS-CH-HSA-TPGS-NPs) coated with Bosutinib (BOS). Nuclear magnetic resonance (NMR) indicated that chitosan's structure was modified by carbodiimide coupling with HSA. We used a Box-Behnken design to find the ideal region for particle size, zeta potential, and entrapment efficiency, eventually emerging at a formulation with these values: 187.14 ± 3.2 nm, 76.2 ± 0.96 %, and 21.1 ± 2.3 mV. Metabolic remodeling, defined as changes in the energy substrate preferences and energy production efficiency of the myocardium, is one of the pathogenic characteristics of failing hearts, though its molecular mechanisms remain incompletely understood. Through untargeted metabolomics coupled with targeted mass spectrometry, we report that plasma succinate levels are significantly elevated in patients with impaired cardiac function (ejection fraction, EF<50%) compared to those with preserved EF (EF⩾50%). Succinate is a key metabolite intermediate in the tricarboxylic acid cycle. Mechanistically

Succinic acid is an important industrial platform chemical with broad applications, and its sustainable production has become increasingly critical. Converting carbon dioxide (CO2) into succinic acid through integrated electrothermal catalytic routes offers a promising alternative to fossil-based and biological methods while contributing to carbon mitigation. This paper summarizes emerging pathways that utilize key CO2-electrocatalytic products, including gas-phase (C2H4/CO/H2), liquid-phase (acetic acid/acetate), and gas-liquid-phase (alcohols), as intermediates for thermocatalytic upgrading

This work performs techno-economic assessment (TEA) and cradle-to-gate life cycle assessment (LCA) for a cyanobacterial system producing succinic acid (SA). Experimental studies of cyanobacteria cultivation producing disodium succinate (Na2Succ) in culture were used to build a scaled-up process producing 100 tonnes/year of SA. High-temperature (HT) and low-temperature (LT) crystallization of SA (Process I) and purification using methanol addition (Process II) were considered for downstream separation. A combination of upstream experimental data, ecoinvent® 3.9 database, literature, and simulat

Bone-related diseases, including osteoporosis (OP), osteoarthritis (OA), rheumatoid arthritis (RA), fracture, and periodontitis, significantly impact human health. Succinate, primarily known as a metabolic intermediate in the tricarboxylic acid (TCA) cycle, has emerged as a regulator of cellular functions beyond its metabolic role. Under stress, succinate accumulates in mitochondria and acts as a signaling molecule, modulating cellular processes. Notably, succinate activates angiogenesis and inflammation by stabilizing hypoxia-inducible factor-1α (HIF-1α). Moreover, it influences various patho

Patients with metabolic syndrome and liver dysfunction demonstrate elevated levels of succinate in the circulation. Succinate has been causally associated with nonalcoholic fatty liver disease (NAFLD) in multiple animal models and via different mechanisms including interaction with succinate receptor-1 (SUCNR-1) in hepatic stellate cells (HSCs), activity of AMP-activated protein kinase (AMPK) and others. Although skeletal muscle is a major source of endogenous succinate, here, using a transgenic mouse with muscle-specific ablation of succinate dehydrogenase complex subunit C (SDHC knockout ani

The objective of the study is to evaluate the evolving phenotype and genetic spectrum of patients with succinic semialdehyde dehydrogenase deficiency (SSADHD) in long-term follow-up. Longitudinal clinical and biochemical data of 22 pediatric and 9 adult individuals with SSADHD from the patient registry of the International Working Group on Neurotransmitter related Disorders (iNTD) were studied with in silico analyses, pathogenicity scores and molecular modeling of ALDH5A1 variants. Leading initial symptoms, with onset in infancy, were developmental delay and hypotonia. Year of birth and specif

Succinate dehydrogenase (SDH), formed by four subunits SDHA, SDHB, SDHC, SDHD, and an assembly factor SDHAF2, functions as a key respiratory enzyme. Biallelic inactivation of genes encoding any of the components, almost always in the presence of a germline mutation, causes loss of function of the entire enzyme complex (so-called SDH deficiency) and subsequent development of SDH-deficient neoplasms which include pheochromocytoma/paraganglioma, gastrointestinal stromal tumor, and renal cell carcinoma (RCC). These tumors may occur in the same patient or kindred. SDH-deficient RCC shows distinctiv

AIMS: Succinate accumulates several-fold in the ischaemic heart and is then rapidly oxidized upon reperfusion, contributing to reactive oxygen species production by mitochondria. In addition, a significant amount of the accumulated succinate is released from the heart into the circulation at reperfusion, potentially activating the G-protein-coupled succinate receptor (SUCNR1). However, the factors that determine the proportion of succinate oxidation or release, and the mechanism of this release, are not known.
METHODS AND RESULTS: To address these questions, we assessed the fate of accumulate

Succinate dehydrogenase (SDH) mutations lead to the accumulation of succinate, which acts as an oncometabolite. Germline SDHx mutations predispose to paraganglioma (PGL) and pheochromocytoma (PCC), as well as to renal cell carcinoma and gastro-intestinal stromal tumors. The SDHx genes were the first tumor suppressor genes discovered which encode for a mitochondrial enzyme, thereby supporting Otto Warburg's hypothesis in 1926 that a direct link existed between mitochondrial dysfunction and cancer. Accumulation of succinate is the hallmark of tumorigenesis in PGL and PCC. Succinate accumulation

Succinate dehydrogenase (SDH), also named as complex II or succinate:quinone oxidoreductases (SQR) is a critical enzyme in bioenergetics and metabolism. This is because the enzyme is located at the intersection of oxidative phosphorylation and tricarboxylic acid cycle (TCA); the two major pathways involved in generating energy within cells. SDH is composed of 4 subunits and is assembled through a multi-step process with the aid of assembly factors. Not surprisingly malfunction of this enzyme has marked repercussions in metabolism leading to devastating tumors such as paraganglioma and pheochro

We investigated the mechanisms underlying the vascular effects of succinate. Vascular reactivity experiments were performed in aortic rings isolated from male Wistar rats and C57BL/6 wild type (WT) or GPR91(-/-) mice. Nitrate/nitrite (NOx) was measured colorimetrically whereas 6-keto-prostaglandin F1α (stable product of prostacyclin) was measured by enzyme immunoassay (EIA). Phosphorylation of endothelial nitric oxide synthase (eNOS) was assessed by western immunoblotting. Functional assays revealed that the direct effect of succinate in the vasculature is biphasic. At lower concentrations suc

Buccal administration of sumatriptan succinate might be an interesting alternative to the present administration routes, due to its non-invasiveness and rapid onset of action, but because of its low permeability, a permeation enhancement strategy is required. The aim of this work was then to study, in-vitro, buccal iontophoresis of sumatriptan succinate. Permeation experiments were performed in-vitro across pig esophageal epithelium, a recently proposed model of human buccal mucosa, using vertical diffusion cells. The iontophoretic behavior of the tissue was characterized by measuring its isoe
