Sunitinib is a multitargeted kinase inhibitor that inhibits many receptor tyrosine kinases and has been used in the treatment of gastrointestinal stromal tumors, metastatic renal cell carcinoma, and pancreatic neuroendocrine tumors. In this study, the effects of sunitinib given to rats, both alone and after stress with cisplatin, were investigated. The animals were divided into four groups - (1) control group (C) administered interperitoneally with a single dose 0.9 % saline, (2) Cis group administered a single dose (7 mg/kg) of cisplatin, (3) Sun group administered 10 mg/kg sunitinib for seve

Mast cell activation disease typically presents as chronic multisystem polymorbidity of generally inflammatory ± allergic theme. Presently, treatment of the rare, cytoproliferative variant systemic mastocytosis employs empirically selected therapies to impede mast cell mediator production and action and, when necessary, inhibition of proliferation. Some tyrosine kinase inhibitors (TKIs) have been used successfully in uncommon cases of systemic mastocytosis not bearing that disease's usual imatinib-resistant KIT D816V mutation. Recently, sunitinib, a multi-targeted TKI, had been successful in

PURPOSE: To evaluate the effects of topical everolimus and sunitinib on corneal neovascularization (CNV).
METHODS: CNV was induced by application of silver nitrate to the cornea for all groups. Rats were divided into four groups of 10 rats each, and two corneas were obtained from each rat. Group I received 1 mg/ml everolimus, Group II received 0.5 mg/ml sunitinib, Group IV received no treatment (control group) and Group IV received 1% Dimethylsulfoxide (DMSO). All treatments were administrated twice daily for 2 weeks. The right corneas were used for extracellular signal-regulated kinase 1/2 (

Pyoderma gangrenosum is a painful, noninfectious, ulcerative, cutaneous disorder that most commonly affects the lower extremities. The diagnosis is made by recognizing the characteristic clinical manifestations in the setting of a causative systemic disease and excluding other causes of similar-appearing ulcerations. Pyoderma gangrenosum often evolves in the setting of systemic illnesses, such as inflammatory bowel disease, rheumatological disorders, and hematological conditions. However, several medications have rarely been linked to pyoderma gangrenosum. The authors report the second case of

BACKGROUND: Pazopanib and sunitinib provided a progression-free survival benefit, as compared with placebo or interferon, in previous phase 3 studies involving patients with metastatic renal-cell carcinoma. This phase 3, randomized trial compared the efficacy and safety of pazopanib and sunitinib as first-line therapy.
METHODS: We randomly assigned 1110 patients with clear-cell, metastatic renal-cell carcinoma, in a 1:1 ratio, to receive a continuous dose of pazopanib (800 mg once daily; 557 patients) or sunitinib in 6-week cycles (50 mg once daily for 4 weeks, followed by 2 weeks without tre

CONTEXT: Sunitinib malate is an oral tyrosine kinase inhibitor used in the treatment of renal cell carcinoma (RCC) and gastrointestinal stromal tumours. Hypothyroidism has been observed in patients treated with sunitinib, but the mechanism whereby sunitinib induces hypothyroidism is unknown.
OBJECTIVE: To describe a series of six patients who developed thyrotoxicosis while on sunitinib for metastatic RCC.
SETTING: The study was conducted at Austin Health, a tertiary teaching hospital in Melbourne, Australia.
RESULTS: Two patients developed severe thyrotoxicosis within 10 weeks after commenc

A 66-year-old previously healthy female was diagnosed with renal cell carcinoma (RCC) metastic to the 4th, 6th and 12th thoracic vertebrae. Despite external beam radiotheraphy (20G in 5 fractions), she progressed to spinal cord compression within 4 weeks and was wheelchair bound. Radiation and surgery were not considered suitable, but sunitinib was initiated. After two 6-week sunitinib cycles, she is fully ambulatory. She reports grade I fatigue and occasional epistaxis, as sole side effects. This case demonstrates that despite the rapid progression of the disease, which escaped the effect of

The combined use of sunitinib and other tyrosine kinase inhibitors (TKIs) is discouraged because of the increased risk of adverse events (AEs). Furthermore, plasma sunitinib levels are affected by drugs that affect CYP3A4 activity; therefore, caution should be exercised when using CYP3A4 inhibitors. In the present study, a 59-year-old Japanese man with metastatic renal cell carcinoma (RCC) was diagnosed with chronic myeloid leukemia (CML) while on sunitinib treatment and was simultaneously treated with sunitinib and dasatinib, a multi-TKI used for CML with moderate CYP3A4 inhibitory activity.

BACKGROUND: Sunitinib resistance remains a major clinical challenge in renal cell carcinoma, and the underlying molecular mechanisms are incompletely understood. SP5, a member of the specificity protein transcription factor family, has been implicated in tumor progression, but its role in sunitinib resistance and angiogenesis in RCC is unclear.
METHODS: We integrated bulk RNA-seq datasets (GSE172165, GSE173572, GSE216494) and single-cell transcriptomics (GSE156632) to identify sunitinib resistance-associated differentially expressed genes. A prognostic model was constructed using 117 machine-

Hand-foot skin reaction (HFSR) is a common dermatological adverse effect of multikinase inhibitors like sunitinib that can impair quality of life and disrupt treatment continuity. We report a 52-year-old male with metastatic renal cell carcinoma who developed Common Terminology Criteria for Adverse Events (CTCAE v5.0) Grade 2 sunitinib-induced HFSR presenting as painful palmar-plantar hyperpigmented lesions and bullae during his third treatment cycle (50 mg/day; two weeks on, one week off). Sunitinib was temporarily held, and multimodal supportive management (including topical corticosteroids,

Pancreatic neuroendocrine tumors (PanNETs) are rare malignancies of the pancreas, but their incidence is steadily increasing. Standard therapy with antiangiogenic inhibitors, including sunitinib, has shown clinical benefit for advanced PanNETs; however, its long-term effectiveness is limited due to the development of resistance. In this study, we demonstrate that targeting the CD93-IGFBP7 axis enhances the efficacy of sunitinib by normalizing tumor vasculature in PanNETs. Both CD93 and its ligand IGFBP7 are enriched in the tumor microenvironment (TME) of PanNETs. Disrupting the CD93/IGFBP7 int

Corneal disease is a leading cause of blindness globally, and is commonly associated with corneal neovascularization, which results in the loss of optical clarity and visual impairment. A wide variety of factors contribute to abnormal growth of new blood vessels into the cornea, including contact lens wear, infectious keratitis, corneal graft rejection, inflammatory disorders and trauma. In each case, proangiogenic factors such as vascular endothelial, platelet-derived, and fibroblast growth factors fuel new blood vessel growth and proliferation. Topical corticosteroids are a front-line treatm

BACKGROUND: Sunitinib has been the standard of care for patients with metastatic renal cell carcinoma (mRCC). However, nearly all patients will eventually develop resistance. Before the introduction of novel agents, few treatment options remained after sunitinib failure. Sunitinib rechallenge is a strategy based on the presumption that resistance might be only temporary. The aim of this analysis was to evaluate the efficacy and safety of sunitinib rechallenge in patients with mRCC.
PATIENTS AND METHODS: Patients who had undergone sunitinib rechallenge (SU2) at the Medical University of Vienna

Adrenocortical carcinoma (ACC) is a rare malignancy with poor prognosis. Palliative chemotherapy can be considered in patients with metastatic disease. Although mitotane- or cisplatin-based chemotherapy regimens are widely used, the effects of these agents have been limited. We have experienced a case that showed a partial response with sunitinib after failure of mitotane-based cytotoxic chemotherapy. The clinical benefit from sunitinib persisted 7.5 months in this case. To our knowledge, this is the first reported case showing the effects of sunitinib on metastatic ACC.

Mast cell (MC) activation syndrome (MCAS) is a recently recognized, likely prevalent collection of heterogeneous illnesses of inappropriate MC activation with little to no MC neoplasia likely driven by heterogeneous patterns of constitutively activating mutations in MC regulatory elements including various tyrosine kinases (TKs, dominantly KIT). MCAS typically presents as chronic multisystem polymorbidity of generally inflammatory ± allergic theme. As with indolent systemic mastocytosis (SM), treatment of MCAS focuses more against MC mediators than MC neoplasia, but some cases prove refractory
