BACKGROUND AND PURPOSE: Results from several studies point to voltage-gated Na(+) channels as potential mediators of the immobility produced by inhaled anaesthetics. We hypothesized that the intrathecal administration of tetrodotoxin, a drug that blocks Na(+) channels, should enhance anaesthetic potency, and that concurrent administration of veratridine, a drug that augments Na(+) channel opening, should reverse the increase in potency.
EXPERIMENTAL APPROACH: We measured the change in isoflurane potency for reducing movement in response to a painful stimulus as defined by MAC (minimum alveola…

High performance liquid chromatography (HPLC) using fluorescent detection following post-column alkaline degradation and a sample preparation procedure for the analysis were established to detect tetrodotoxins (TTXs) in seafood. In south Taiwan Prefectures, each specimen of shellfishes and gobies, collected from Chiayi, Tainan, and Pingtung from January 1997 to May 1998, was analyzed by HPLC to detect the presence and quantity of TTXs. Overall results showed that only 5 specimens out of 557 specimens contained TTXs; the toxic species were gastropod Natica lineata and Nassarius livescens. The h…

The role of neuronal activity in the refinement of the retinotectal projection in chick embryos was studied using tetrodotoxin (TTX) which blocks sodium channels and grayanotoxin I (GTX) which opens the channels. Optic nerve fibers were traced by the fluorescent dye DiI. The toxins were injected into the eyeball during the refinement phase of retinotectal projection (either the 13th, 14th or 15th day of incubation). A tiny crystal of DiI was placed in the temporal part of the retina. In the control embryos, fibers caudally overshooting and arborizations outside the terminal zone were found bef…

The developing uncrossed retinocollicular projection in the Syrian hamster undergoes a characteristic set of changes during the first 2 postnatal weeks. The retinal fibres, which initially project across the whole superior colliculus, withdraw from the caudal part and their terminals become clustered into deep, discrete clumps rostrally. Coincident with these afferent changes, there is substantial retinal ganglion cell death. To examine whether neuronal activity plays a role in these changes, we made daily injections of the sodium channel blocker tetrodotoxin (TTX) into one or both eyes from p…

Behavioral effects of bilateral intracranial infusions of tetrodotoxin (1, 3.3 or 10 ng/rat), 50% procaine (2 microliters/rat) or phosphate-buffered saline (PBS-2 microliters/rat) into the dorsal midbrain of conscious, lightly-restrained female rats were evaluated. High levels of lordotic responsiveness were induced in ovariectomized animals treated with estradiol (E2) capsules or subcutaneous injections of estradiol benzoate (EB) followed by progesterone (P). The effect of each of the 3 infusates on lordosis was determined using manual stimulation and lordosis quotient determinations. In addi…

The action potential Na+ ionophore of excitable cells can be activated either by alkaloid compounds such as veratridine, or by small polypeptide toxins extracted from scorpion venom or sea anemone. One of the main features of this Na+ channel is that it is blocked by tetrodotoxin (TTX). However, we report here that during analysis of Na+ influx in resting fibroblasts, we found that a variety of fibroblast lines also possess a TTX receptor. Veratridine and sea anemone toxin act synergistically to stimulate Na+ influx 7 to 10-fold in hamster and rat fibroblasts. As in excitable cells, this toxin…

Propranolol, a representative adrenergic β-receptor antagonist, is widely used to prevent migraine attacks. Although propranolol is well known to inhibit tetrodotoxin-resistant (TTX-R) Na+ channels in cardiac myocytes, it is unclear whether the drug modulates these channels expressed in dural afferent neurons. In this study, we examined the effects of propranolol on TTX-R Na+ channels in medium-sized dural afferent neurons identified by the fluorescent dye DiI. The TTX-R Na+ currents (INa) were recorded from acutely isolated DiI-positive neurons using a whole-cell patch clamp technique under v…

Granule cell dispersion (GCD) has been associated as a pathological feature of temporal lobe epilepsy (TLE). Early-life epileptiform activit … heat-shock induced GCD could be significantly reduced by treatment with the sodium channel blocker tetrodotoxin (TTX), suggesting that electrical activity is an important factor involved in heat-shock induced GCD.

1. Extracellular recording techniques have been used to study nerve impulses in single sensory nerve terminals in guinea-pig cornea isolated in vitro. 2. Nerve impulses occurred spontaneously and were evoked by electrical stimulation of the ciliary nerves. 3. The nerve impulses were identified as originating in polymodal receptors, mechano-receptors or 'cold' receptors. All three types are believed to be nociceptors. 4. Tetrodotoxin (TTX, 1 microM) blocked nerve impulses evoked by electrical stimulation of the ciliary nerves. However, ongoing and/or naturally evoked nerve impulses persisted in…

BACKGROUND: The Suez Canal permits migration of fish from the Indo-Pacific Ocean to the Mediterranean Sea. This phenomenon (Lessepsian migration) has enabled poisonous fish species to colonize the Mediterranean Sea.
OBJECTIVE: To report clinical tetrodotoxin poisoning after consumption of the Lessepsian immigrant fish Lagocephalus sceleratus caught on the Israeli coast of the eastern Mediterranean.
CASE SERIES: Thirteen patients aged 26-70years were admitted after consuming L. sceleratus. Signs of toxicity appeared within 1h. The main manifestations included vomiting, diarrhea, headache, par…

BACKGROUND: In isolated nerves, tetrodotoxin (TTX) blocks nerve conduction longer than bupivacaine. In vivo, however, both substances block nerve conduction to an equal duration, presumably because the hydrophilic TTX binds only weakly to the perineural tissue. High molecular weight hyaluronic acid (HA) prolongs the action of local anaesthetics several-fold. We tested whether admixture of HA enhances the binding of TTX to the perineural tissue and thus induces an ultra-long conduction block after a single application.
METHODS: In 12 anaesthetized rabbits, the minimal blocking concentrations o…

A study in cats was designed to investigate the blocking of sensory nerve terminals by i.v. injection of lipid-soluble local anaesthetics such as lignocaine. I.v. injection of lignocaine 15--30 mg kg-1 did not completely block sensory nerve endings in the tooth of the cat, whilst tetrodotoxin (TTX) in large non-toxic doses did so. Based on the i.v. doses of TTX and the relationship between the local anaesthetic potency of the two substances when injected intra-arterially, it was calculated that lethal doses of i.v. administered lignocaine are necessary to block sensory nerve endings. It seems …

Tetrodotoxin (TTX) is one of the most potent naturally occurring neurotoxins and continues to attract considerable scientific, clinical, forensic, and regulatory attention. Historically associated with pufferfish poisoning in East and Southeast Asia, TTX has recently emerged as a global food safety concern due to its detection in an increasing number of marine and terrestrial organisms and its expanding geographical distribution, including European coastal waters. Despite decades of research, important questions regarding its biosynthesis, ecological origin, toxicokinetics, and environmental d…

Pufferfishes (Tetraodontidae) accumulate tetrodotoxin, a potent neurotoxin, primarily through microbial and trophic pathways rather than endogenous biosynthesis, linking their toxicity directly to marine microbial ecology and food-web structure. Anthropogenic climate change, ocean warming, deoxygenation, and acidification are reshaping the environmental conditions that govern TTX production, transfer, and accumulation. This review synthesizes evidence from 62 studies (28 with quantitative data) identified through a PRISMA-ScR-compliant search. We organize findings around three mechanistic path…

Pacific oysters (Crassostrea gigas) may bio-accumulate high levels of paralytic shellfish toxins (PST) during harmful algal blooms of the ge … X sensitivity compared to other bivalve species. However, theses nerves were much less sensitive to tetrodotoxin. The STX-sensitivity of compound nerve action potential (CNAP) recorded from oysters experimentally fed with Alexandrium minutum (toxic-alga-exposed oysters), or Tisochrysis lutea, a non-toxic microalga (control oysters), revealed that oysters could …

Tetrodotoxin (TTX) is a selective blocker of voltage-gated Na+ channels that is used to block action potentials in vitro and in vivo. Maintaining a sufficiently high local concentration of TTX in vivo to block conduction in a peripheral nerve is technically demanding and carries a risk of systemic toxicity. We report that slow diffusion of TTX out of a microcapsule (glass capillary) inserted beneath the epineurium of the sciatic nerve, with a loose cuff around the nerve, combines high blocking efficacy with low systemic toxicity in rats and mice. The local anaesthesia and motor paralysis was s…

We hypothesize that the accumulation of tetrodotoxin (TTX) sensitive sodium channels in injured dorsal root ganglion (DRG) neurons plays a critically important role in the generation of ectopic discharges and mechanical allodynia after peripheral nerve injury. Using the segmental spinal nerve (L5) ligation model of neuropathic pain, this hypothesis was tested by examining the effect of TTX on the mechanical sensitivity of the affected hind paw. Various concentrations of TTX were applied topically to the L5 DRG by using chronically implanted polyethylene tubing. The data showed that application…

1. Relaxation responses were produced by vagal and field stimulation, respectively, of the whole oesophagus preparation from the rat and of the isolated tunica muscularis mucosae (t.m.m.) preparation from the rat. These relaxation responses persisted in the presence of antagonists of histamine, serotonin, noradrenaline and acetylcholine. 2. Unlike vagally evoked relaxation, that evoked by low-frequency field stimulation, i.e. field-stimulated relaxation (f.s.r.) was generally resistant to tetrodotoxin (TTX). 3. Both types of relaxations exhibited remarkable temperature sensitivity and were abo…
