PURPOSE: Hypothyroidism is an endocrine disorder characterised by decreased T3, T4 and increased TSH levels. This study aims to examine the potential effects of Ferulic acid (FA) on rats with hypothyroidism induced by propylthiouracil through the estimation of biochemical parameters and histopathological studies.

In this preliminary study, the potential of an in utero-lactation assay to detect thyroid effectors was evaluated by treating three dams/group with 6-n-propyl-2-thiouracil (PTU), a known thyroid antagonist, by oral gavage at doses of 0, 0.0032, 0.016, 0.08 and 0.4 mg/kg/day during fetal organogenesis and lactation. The proposed mechanism of iodothyronine deiodinase inhibition by the thiourea-derived drugs 6-n-propylthiouracil (PTU) and 6-methylthiouracil is supported by experimental evidence. Model reactions with sterically or coordinatively stabilized organoselenyl iodides as enzyme-mimetic substrates (E-SeI; see scheme) support the proposal that PTU reacts not with the enzyme but with the enzyme-SeI intermediate containing a covalent Se-I bond, and suggest that the Se-I bond is kinetically activated by basic amino acid groups such as histidine.

Protamone supplementation did not correct growth depression due to feeding rapeseed meal although inclusion levels up to 0.1% may have induced a hyperthyrotic condition which in itself leads to a growth depressing effect. Metal pyrimidine complexes (MPCs) including cadmium-barbiturate (Cd-BA), zinc-barbiturate (Zn-BA), cadmium-thiouracil (Cd-TU) and mercury-thiouracil (Hg-TU) were prepared and their analysis was carried out. These MPCs were evaluated as monoamine oxidase-B (MAO-B) inhibitors. Rat brain MAO-B was inhibited (in vitro) by Cd-BA, Zn-BA, Cd-TU and Hg-TU complexes.
