Exploring the biocatalytic potential of wild-type split transketolases.

Split transketolases (split TKs) are an underexplored class of carboligases with potential for application in biocatalysis. Here, we investigate ten split TKs and reveal substantial functional heterogeneity, with marked differences in substrate scope, activity, enantioselectivity, and robustness. The enzymes accepted diverse acceptors, aliphatic and aromatic, while retaining the canonical (S)-selectivity of full-length TKs. Pyruvate acceptance further revealed distinct catalytic behaviours, including cross-coupling and self-coupling specificity. Selected enzymes also displayed high organic sol…

Erythrocyte transketolase activity in patients with diabetic and alcoholic neuropathies.

BACKGROUND: Clear cell renal cell carcinoma (ccRCC) is characterized by aggressive behavior and poor prognosis. Ubiquitin-conjugating enzyme E2C (UBE2C), a key component of the ubiquitin-proteasome system, has been implicated in tumorigenesis; however, its role in ccRCC remains unclear. This study aimed to systematically investigate the clinical relevance, genetic associations, and biological functions of UBE2C in ccRCC. METHODS: Multi-omics analyses of The Cancer Genome Atlas Kidney Renal Clear Cell Carcinoma (TCGA-KIRC) and Gene Expression Omnibus (GEO) datasets were performed to evaluate U

Relationship of nervous tissue transketolase to the neuropathy in chronic uremia.

Patients with chronic uremia develop neurologic defects which are similar to the demyelinating lesions seen in thiamine deficiency. The present study describes inhibitory effects of uremic material on nervous tissue transketolase, a thiamine-dependent enzyme of the pentose phosphate pathway which has been reported to have functional importance in the metabolism of myelinated nervous structures. Transketolase activity (TKA) of normal human brain and spinal cord was measured by the conversion of ribose-5-phosphate (R5P) to sedoheptulose-7-phosphate (S7P). TKA was significantly inhibited by plasm…

Transketolase activity modulates glycerol-3-phosphate levels in Escherichia coli.

Transketolase activity provides an important link between the metabolic pathways of glycolysis and pentose phosphate shunt and catalyzes inter-conversions between pentose phosphates and glycolytic intermediates. It is widely conserved in life forms. A genetic screen for suppression of the growth defect of Escherichia coli tktA tktB mutant in LB medium revealed two mutations, one that rendered the glpK expression constitutive and another that inactivated deoB. Characterizing these mutations aided in uncovering the role of ribose-5-P (a transketolase substrate) as an inhibitor of glycerol assimi…

Altered heavy metals and transketolase found in autistic spectrum disorder.

Autism and autism spectrum disorder (ASD) are developmental brain disorders with complex, obscure, and multifactorial etiology. Our recent clinical survey of patient records from ASD children under the age of 6 years and their age-matched controls revealed evidence of abnormal markers of thiol metabolism, as well as a significant alteration in deposition of several heavy metal species, particularly arsenic, mercury, copper, and iron in hair samples between the groups. Altered thiol metabolism from heavy metal toxicity may be responsible for the biochemical alterations in transketolase, and are…

Transketolase haploinsufficiency reduces adipose tissue and female fertility in mice.

Transketolase (TKT) is a ubiquitous enzyme used in multiple metabolic pathways. We show here by gene targeting that TKT-null mouse embryos are not viable and that disruption of one TKT allele can cause growth retardation ( approximately 35%) and preferential reduction of adipose tissue ( approximately 77%). Other TKT(+/-) tissues had moderate ( approximately 33%; liver, gonads) or relatively little ( approximately 7 to 18%; eye, kidney, heart, brain) reductions in mass. These mice expressed a normal level of growth hormone and reduced leptin levels. No phenotype was observed in the TKT(+/-) co…

Molecular evolutionary analysis of the thiamine-diphosphate-dependent enzyme, transketolase.

Members of the transketolase group of thiamine-diphosphate-dependent enzymes from 17 different organisms including mammals, yeast, bacteria, and plants have been used for phylogenetic reconstruction. Alignment of the amino acid and DNA sequences for 21 transketolase enzymes and one putative transketolase reveals a number of highly conserved regions and invariant residues that are of predicted importance for enzyme activity, based on the crystal structure of yeast transketolase. One particular sequence of 36 residues has some similarities to the nucleotide-binding motif and we designate it as t…

Reassessing Transketolase Assays: Methodological Considerations for Detecting Functional Thiamine Deficiency.

Thiamine (vitamin B1) deficiency due to degradation, inadequate intake, impaired absorption, or increased metabolic demand remains prevalent in both human and animal populations. In its diphosphate form (thiamine diphosphate, TDP), thiamine serves as an essential cofactor for metabolic enzymes, including transketolase (TKT), a rate-limiting enzyme of the pentose-phosphate pathway. Measurement of TKT activity with and without exogenous TDP provides a functional assessment of thiamine utilization that serves as a surrogate for or complements direct quantification of thiamine forms. Beyond its hi…

An updated transketolase activity assay to probe thiamine utilization in fish.

Thiamine (vitamin B1) deficiency is causative of reproductive failures and population declines in lake trout, Atlantic salmon, and other predatory fish species. Transketolase (TKT) is the rate-limiting enzyme of the non-oxidative phase of the pentose-phosphate pathway. TKT is critical for mediating the availability of sugars to return to glycolysis and for synthesizing NADPH and R5P, which are needed to maintain the cellular oxidation state and to produce biomolecules necessary for successful growth and reproduction. TKT activity provides a measure of functional thiamine availability since it …

Validation of the robust performance of novel transketolase epitopes for tuberculosis immunodiagnostics.

Tuberculosis (TB) is a major global health concern, contributing to millions of deaths annually. Mycobacterium tuberculosis (M.tb) is the causative agent for TB. The dormancy/non-replicative phase of M.tb is characterized by a lower cell division and decreased metabolic rate. Transketolase (TKT) plays a crucial role in bacterial growth and M.tbTKT plays a key role in switch from the dormancy to proliferative phase. Despite advances in molecular TB-diagnostics, there is an unmet need for a point-of-care TB-diagnostics. Through immunoscreening of a novel T7 phage display library, we identified a…

Characterisation of plasmodial transketolases and identification of potential inhibitors: an in silico study.

BACKGROUND: Plasmodial transketolase (PTKT) enzyme is one of the novel pharmacological targets being explored as potential anti-malarial drug target due to its functional role and low sequence identity to the human enzyme. Despite this, features contributing to such have not been exploited for anti-malarial drug design. Additionally, there are no anti-malarial drugs targeting PTKTs whereas the broad activity of these inhibitors against PTKTs from other Plasmodium spp. is yet to be reported. This study characterises different PTKTs [Plasmodium falciparum (PfTKT), Plasmodium vivax (PvTKT), Plasm…

[Transketolase activity and the TDP effect in tissues of animals with experimental tumors].

Activity of transketolase and the TDP-effect were studied in blood and liver subcellular fractions of mice with Erlich ascites carcinoma and of rats with sarcoma 45 which were maintained on a synthetic diet containing either all the vitamins or devoid of thiamine. As compared with other mice liver subcellular fractions the microsomal fraction proved to be the most sensitive to thiamine deficiency: inhibition of transketolase activity reached 75%. Decrease in TDP-effect found in microsomes might reflect the most distinct terminal steps of B1 avitaminosis. As a result of vitamin B1 deprivation o…

Transketolase drives the development of aortic dissection by impairing mitochondrial bioenergetics.

AIM: Aortic dissection (AD) is a disease with rapid onset but with no effective therapeutic drugs yet. Previous studies have suggested that glucose metabolism plays a critical role in the progression of AD. Transketolase (TKT) is an essential bridge between glycolysis and the pentose phosphate pathway. However, its role in the development of AD has not yet been elucidated. In this study, we aimed to explore the role of TKT in AD. METHODS: We collected AD patients' aortic tissues and used high-throughput proteome sequencing to analyze the main factors influencing AD development. We generated a…

Effect of chronic alcohol administration on transketolase in the brain and the liver of rats.

To estimate the nutritional and the pathological states in thiamin-deficiency-related diseases, especially Wernicke-Korsakoff syndrome, we studied the relationship among transketolase activity, transketolase concentration, and thiamin phosphate esters in rats chronically fed alcohol. In the brain of alcohol-fed rats, the enzyme activity and concentration decreased although there was no positive correlation between the two. On the contrary, transketolase activity in the liver correlated positively with concentration, and both transketolase activity and concentration were decreased in the thiami…
