Antigenic cross-reactivity between the human whipworm, Trichuris trichiura, and the mouse trichuroids Trichuris muris and Trichinella spiralis.

Cross-reactivity was demonstrated between circulating antibodies from Trichuris trichiura-infected humans and T. muris-infected mice for heterologous antigen preparations. Mouse immune sera raised against excretory/secretory (E/S) products and anterior end homogenate from adult T. muris showed marked affinity for T. trichiura adult homogenate in ELISA, and 35S-methionine-labelled adult T. muris E/S products were precipitated by T. trichiura infection sera. Monoclonal antibodies recognizing a 48 kD Trichinella spiralis muscle larval beta-stichocyte granule antigen also showed avidity for T. tri…

Emodepside for Trichuris trichiura and Hookworm Infection.

BACKGROUND: Current treatments for soil-transmitted helminth infections in humans have low efficacy against Trichuris trichiura. Emodepside - a drug in veterinary use and under development for the treatment of onchocerciasis in humans - is a leading therapeutic candidate for soil-transmitted helminth infection. METHODS: We conducted two phase 2a, dose-ranging, randomized, controlled trials to evaluate the efficacy and safety of emodepside against T. trichiura and hookworm infections. We randomly assigned, in equal numbers, adults 18 to 45 years of age in whom T. trichiura or hookworm eggs had…

Trickle infection and immunity to Trichuris muris.

The majority of experiments investigating the immune response to gastrointestinal helminth infection use a single bolus infection. However, in situ individuals are repeatedly infected with low doses. Therefore, to model natural infection, mice were repeatedly infected (trickle infection) with low doses of Trichuris muris. Trickle infection resulted in the slow acquisition of immunity reflected by a gradual increase in worm burden followed by partial expulsion. Flow cytometry revealed that the CD4+ T cell response shifted from Th1 dominated to Th2 dominated, which coincided with an increase in …

Immunity to Trichuris muris in the laboratory mouse.

Of all the laboratory models of intestinal nematode infection, Trichuris muris in the mouse is arguably the most powerful. This is largely due to the fact that the ability to expel this parasite is strain dependent. Thus, most mouse strains readily expel T. muris. However certain mouse strains, and indeed some individuals within particular mouse strains, are unable to mount a protective immune response and harbour long term chronic infections. This unique model thus presents an opportunity to examine the immune events underlying both resistance to infection and persistent infection within the …

The public health significance of Trichuris trichiura.

An estimated 1049 million persons harbour T. trichiura, including 114 million preschool-age children and 233 million school-age children. The prevalence of T. trichiura is high and may reach 95% in children in many parts of the world where protein energy malnutrition and anaemias are also prevalent and access to medical care and educational opportunities is often limited. The Trichuris dysentery syndrome (TDS) associated with heavy T. trichiura, which includes chronic dysentery, rectal prolapse, anaemia, poor growth, and clubbing of the fingers constitutes an important public health problem, a…

The adverse consequences of heavy Trichuris infection.

Sixty-seven children with heavy Trichuris trichiura infection (Group A) were compared to 73 control children of similar socio-economic status but with light or no T. trichiura (Group B), for nutritional status, rates of concomitant bacterial and protozoal, and symptoms and clinical signs associated with heavy T. trichiura infection. Anoscopy was used to determine heavy T. trichiura infection. Measurements and physical examination were done on Group A on admission to and discharge from hospital; 46% were seen on follow-up visit two to eight months later. Children in Group B were seen only once.…

Visceral larva migrans caused by Trichuris vulpis.

Two brothers with visceral larva migrans caused by Trichuris vulpis were diagnosed after they had been investigated for an eosinophilia. Both patients were almost asymptomatic. The diagnosis of visceral larva migrans was based on the results of immunoelectrophoretic studies and no liver biopsy was performed. After administration of thiabendazole, the number of eosinophils and serum total IgE levels gradually decreased, and the patients have remained well.

Oxantel pamoate-albendazole for Trichuris trichiura infection.

BACKGROUND: Infections with soil-transmitted helminths (Ascaris lumbricoides, hookworm, and Trichuris trichiura) are widespread and often occur concomitantly. These parasitic-worm infections are typically treated with albendazole or mebendazole, but both drugs show low efficacy against T. trichiura. Albendazole is the drug of choice against hookworm. METHODS: In this double-blind trial conducted on Pemba Island, Tanzania, we randomly assigned children, 6 to 14 years of age, to receive one of four treatments: oxantel pamoate at a dose of 20 mg per kilogram of body weight, plus 400 mg of albend…
