The leucine-responsive regulatory protein (lrp) gene for characterization of the relationship among Xanthomonas species.

Characterization of strains of Xanthomonas axonopodis pv. citri by using DNA fingerprints that were generated from primers for enterobacterial repetitive intergenic consensus (ERIC) elements led to the discovery of differential sequences for a leucine-responsive regulatory protein (lrp) gene in two subgroups of strains with different host ranges on Citrus spp. DNA hybridization and PCR-based assays that used different sets of primers were designed to detect the core sequence, as well as to obtain the entire sequence of the lrp gene for several Xanthomonas species and pathovars. Higher variabil…

DrdR Negatively Modulates the Expression of Flagellar Genes via Interaction With FleQ in Xanthomonas campestris.

Response regulators (RRs) of two-component signalling systems (TCSs) containing tandem receiver (REC) domains are widespread in bacteria, yet their functions and regulatory mechanisms remain poorly understood. In our previous study, DrdR, one such RR in the cruciferous black rot disease pathogen Xanthomonas campestris pv. campestris (Xcc) was demonstrated to positively regulate pilus-dependent motility and negatively regulate flagellum-dependent motility. We showed that DrdR modulates the ATPase activities of pili motor proteins PilT and PilB, thereby enhancing bacterial pilus-dependent swarmi…

The Transcriptional Regulator Crt1 is Involved in The Pathogenic Lifestyle of Xanthomonas campestris pv. campestris B100.

The γ-proteobacterium Xanthomonas campestris pv. campestris B100 is the causal agent of black rot disease in a wide range of economically important crops. In addition, X. campestris pv. campestris is commercially relevant due to the synthesis of the exopolysaccharide xanthan. In this work, we first introduce a novel transcriptional regulator, termed Crt1, and present the effect that the deletion of the crt1 gene exerts on growth, xanthan production, virulence, and transcriptome and proteome profiles. Differential transcriptome analysis of the deletion mutant X. campestris pv. campestris B100 Δ…

Synthesis of carbazole and naphthyl-pyrimidine-based inhibitors of Xanthomonas for tomato bacterial spot management.

UNLABELLED: Bacterial spot of tomato (BST), caused by Xanthomonas species, is a devastating disease in tomato production, leading to severe yield losses and reduced crop quality. Current control strategies, including cultural management and chemical applications, are increasingly challenged by the emergence of copper- and antibiotic-resistant strains. To address this issue, we are exploring the development of novel small molecule antibacterial as a promising alternative control strategy. Through structural expansion and modification of previously identified anti-Xanthomonas leads (X2, X5, and …

The dual role of Xanthomonas effector XccXynB in plant cell wall deconstruction and immunity activation.

The GH10 endoxylanase XccXynB from Xanthomonas campestris pv. campestris (Xcc) functions as a PAMP, triggering PTI responses and enhancing disease resistance in plants, in addition to its polysaccharide degradation capability. Plant pathogens of the Xanthomonas genus are capable of infecting a wide array of economically important plant species. Among their virulence factors, the GH10 family xylanase, XynB, is typically responsible for the predominant extracellular xylanase activity. However, the detailed biochemical characteristics of this enzyme in most Xanthomonas pathogens remain insufficie…

Biochemical and structural insights of a new endolysin encoded by the Xanthomonas citri lytic phage CP2.

The Gram-negative bacterium Xanthomonas citri subsp. citri (X. citri) is a phytopathogen causal agent of citrus canker, a severe disease affecting citrus crops worldwide. Disease control relies primarily on copper biocides, which pose toxicity risks to humans and the environment. Endolysins are enzymes that catalyze bacterial cell wall degradation and could serve as environmentally sustainable alternatives for the control of X. citri. Here, we conducted structural in silico studies with the endolysin CP2-07 encoded by the X. citri lytic phage CP2, followed by its heterologous expression and pu…

The OsICS1 is directly regulated by OsWRKY6 and increases resistance against Xanthomonas oryzae pv. oryzae.

OsICS1 but not OsICS1-L mediates the rice response to Xoo inoculation, with its overexpression increasing resistance against this pathogen. OsICS1 but not OsICS-L is directly upregulated by OsWRKY6. Rice (Oryza sativa) is a staple crop for about half of the global population and is particularly important in the diets of people living in Asia, Latin America, and Africa. This crop is continually threatened by bacterial leaf blight disease caused by Xanthomonas oryzae pv. oryzae (Xoo), which drastically reduces yields; therefore, it is needed to elucidate the plant's resistance mechanisms against…

A Comprehensive Review of Bacterial Leaf Spot (Xanthomonas spp.) Host Resistance in Pepper (Capsicum spp.).

Bacterial leaf spot (BLS) is one of the most destructive diseases of pepper (Capsicum spp.) worldwide, causing significant yield and quality losses under warm and humid conditions. The disease is caused by multiple Xanthomonas species, whose rapid evolution continues to challenge effective management. This review synthesizes current knowledge on BLS in pepper, including pathogen taxonomy, global distribution, and the historical progression of disease research and resistance breeding. We summarize advances in the identification and deployment of resistance loci, including major race-specific ge…

Antibacterial activity and mechanism of novel berberine derivative HBBR-F4 against Xanthomonas oryzae pv. Oryzae.

BACKGROUND: Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of rice bacterial blight, poses a significant threat to global crop security by means of systemic vascular colonization. In order to address emerging resistance challenges, a rational drug design strategy was employed, which involved structurally optimizing the natural alkaloid berberine (BBR). Through a C-9/C-13 dual-site modification approach, we developed a novel berberine derivative, 9-benzoyloxy-13-(4-(tert-butyl)benzyl)-berberine bromide (HBBR-F4). This compound demonstrated exceptional anti-Xoo efficacy, achieving protect…

A recently collected Xanthomonas translucens isolate encodes TAL effectors distinct from older, less virulent isolates.

Xanthomonas translucens, the causal agent of bacterial leaf streak disease (BLS) in cereals, is a re-emerging pathogen that is becoming increasingly destructive across the world. While BLS has caused yield losses in the past, there is anecdotal evidence that newer isolates may be more virulent. We observed that two X. translucens isolates collected from two sites in Colorado, USA, are more aggressive on current wheat and barley varieties compared to older isolates, and we hypothesize that genetic changes between recent and older isolates contribute to the differences in isolate aggressiveness.…

Antibacterial Activities and Underlying Mechanisms of the Compound SYAUP-491 against Xanthomonas oryzae pv. oryzae.

SYAUP-491 is a novel alkyl sulfonamide. In this study, in vivo and in vitro tests were performed along with a proteomic analysis to determine the effects and underlying mechanisms of the antibacterial activity of SYAUP-491 against the causative agent of bacterial leaf blight in rice. The antibacterial test results suggested that SYAUP-491 exhibited significant activities against Xanthomonas oryzae pv. oryzae (Xoo) in vitro and in vivo. The minimal EC50 values reached 6.96 μg/mL and the curative activity reached 74.1%. Detailed studies demonstrated that SYAUP-491 altered membrane permeability a…

Fvchli deficiency impairs ABA-mediated stomatal closure and enhances susceptibility to Xanthomonas fragariae in strawberry.

The magnesium chelatase subunit CHLI is well-characterized in chlorophyll biosynthesis, but its role in plant immunity remains unclear. In this study, we generated chimeric Fvchli mutant strawberries via CRISPR/Cas9 and obtained T1-T3 progeny with a wide range of editing efficiencies through self-pollination. Mutants with higher editing efficiency exhibited more leaf spots. Following inoculation with Xanthomonas fragariae, Fvchli mutants showed increased susceptibility, with both lesion area and disease incidence positively correlating with editing efficiency. We further demonstrated that stom…

RNA-seq pinpoints a Xanthomonas TAL-effector activated resistance gene in a large-crop genome.

Transcription activator-like effector (TALE) proteins of the plant pathogenic bacterial genus Xanthomonas bind to and transcriptionally activate host susceptibility genes, promoting disease. Plant immune systems have taken advantage of this mechanism by evolving TALE binding sites upstream of resistance (R) genes. For example, the pepper Bs3 and rice Xa27 genes are hypersensitive reaction plant R genes that are transcriptionally activated by corresponding TALEs. Both R genes have a hallmark expression pattern in which their transcripts are detectable only in the presence and not the absence of…

Risks Posed by Intercrops and Weeds as Alternative Hosts to Xanthomonas campestris pv. musacearum in Banana Fields.

Alternative host plants are important in the survival and perpetuation of several crop pathogens and have been suspected to play a role in the survival of Xanthomonas campestris pv. musacearum (Xcm) and perpetuation of Xanthomonas wilt (XW) disease of banana and enset. This study determined the potential risk posed by two weeds (Canna spp. and wild sorghum) and common banana intercrops (maize, millet, sorghum, taro, and sugarcane) as alternative hosts to Xcm. The study employed screenhouse experiments, laboratory procedures and diagnosis of banana fields in XW-affected landscapes. Typical XW s…

Screening and characterization of Xanthomonas oryzae pv. oryzae strains with resistance to pheazine-1-carboxylic acid.

Xanthomonas oryzae pv. oryzae (Xoo) causes bacterial leaf blight (BLB) and can be effectively controlled by phenazine-1-carboxylic acid (PCA), an antibiotic secreted by Pseudomonas spp. PCA resistance in Xoo was investigated in this research. Only four PCA-resistant strains were obtained by extensive screening, and the resistance was genetically stable in only one of them (P4). P4 was also resistant to phenazine and 1-hydroxyphezine but not to captan, bismerthiazol, or streptomycin. The following were reduced in P4 relative to the parental wild type: growth, virulence, EPS production, extracel…

Tandem receiver domain response regulator DrdR modulates swarming by controlling PilB/PilT activity in Xanthomonas campestris.

Like many other bacterial organisms, Xanthomonas campestris pv. campestris (Xcc) employs two motility appendages, a single polar flagellum and type IV pili, for sensing environmental cues and traversing liquids and solid surfaces. However, compared to the flagellum, the molecular mechanisms underlying the function and regulation of the pilus in Xcc and other xanthomonads are not fully understood. In this study, we demonstrated that a special conserved orphan response regulator DrdR, which contains double receiver (REC) domains but lacks an output domain, modulates the activities of motor prote…

Isolation of a complementary bacteriophage from a phage-resistant mutant expands control strategies against Xanthomonas campestris.

Bacterial resistance to bacteriophages represents a major limitation for the durable use of phage-based biocontrol strategies in agriculture. Here, we used a phage-resistance derivative of Xanthomonas campestris pv. campestris (Xcc) as a selective host to isolate an additional phage with activity against resistant bacteria. A spontaneous mutant, designated Xcc 8004R, was isolated following exposure of Xcc 8004 to phage X1. Xcc 8004R exhibited a growth profile comparable to that of the wild-type strain, but differed in colony appearance, X1 adsorption efficiency, and motility phenotypes. Whole-…

Xanthomonas phaseoli pv. Manihotis effector XopAG impairs cassava immunity by hijacking MeHSP26 to suppress flavonoid biosynthesis.

Cassava bacterial blight (CBB), caused by Xanthomonas phaseoli pv. manihotis (Xpm), is a major disease constraining cassava production. Although overexpression of the Xpm effector XopAG enhances Arabidopsis susceptibility to Pseudomonas syringae pv. tomato DC3000, its virulence mechanism in cassava remains unclear. The study showed that deletion of XopAG did not affect morphology or extracellular enzyme production of Xpm but significantly attenuated its virulence on cassava. Quantitative proteomic analysis revealed that 267 differentially accumulated proteins (DAPs) at 6 h post-inoculation (hp…

Stringent response regulators (p)ppGpp and DksA positively regulate virulence and host adaptation of Xanthomonas citri.

The bacterial stringent response is a response to nutrition deprivation and other stress conditions. In Gram-negative bacteria, this process is mediated by the small signal molecules guanosine pentaphosphate pppGpp and guanosine tetraphosphate ppGpp (collectively referred to as (p)ppGpp), and the RNA polymerase-binding transcription factor DksA. The (p)ppGpp synthetase RelA and the bifunctional (p)ppGpp synthase/hydrolase SpoT are responsible for cellular (p)ppGpp levels. Here, we investigated the roles of DksA and (p)ppGpp in the virulence traits of Xanthomonas citri subsp. citri (Xcc), the c…

Copper chloride induces antioxidant gene expression but reduces ability to mediate H2O2 toxicity in Xanthomonas campestris.

Copper (Cu)-based biocides are currently used as control measures for both fungal and bacterial diseases in agricultural fields. In this communication, we show that exposure of the bacterial plant pathogen Xanthomonas campestris to nonlethal concentrations of Cu(2+) ions (75 µM) enhanced expression of genes in OxyR, OhrR and IscR regulons. High levels of catalase, Ohr peroxidase and superoxide dismutase diminished Cu(2+)-induced gene expression, suggesting that the production of hydrogen peroxide (H2O2) and organic hydroperoxides is responsible for Cu(2+)-induced gene expression. Despite high …
