Serum ferritin, alpha-tocopherol, beta-carotene and retinol levels in lymphatic filariasis.

Based on a cross-sectional study conducted among 100 adults in 1993 in Tanga, Tanzania, the relationship between Wuchereria bancrofti infect … microfilaria intensity was a positive predictor of serum ferritin and a negative predictor of serum alpha-tocopherol. In contrast, negative relationships observed between W. bancrofti microfilaria intensity and serum beta-carotene and retinol were not significant. Neither antigen concentration nor clinical manifestations were predictors of micronutrient status. In …

Prevention of ethanol-induced changes in reactive oxygen parameters by alpha-tocopherol.

Rats were given a 200 mg/kg body weight daily dose of alpha-tocopherol by i.p. injection for 15 days. This resulted in elevated levels of glutathione in both liver and brain, and in a reduced hepatic rate of generation of reactive oxygen species. The depression of hepatic and cerebral glutathione levels in ethanol-consuming rats was prevented by simultaneous treatment with alpha-tocopherol. Other putative indices of hepatic pro-oxidant events, namely levels of mixed function oxidase and proteolytic activity, were elevated by alpha-tocopherol both in the presence and absence of ethanol. In addi…

Selective block of tetramethrin-modified sodium channels by (+/-)-alpha-tocopherol (vitamin E).

Pyrethroids exert their hyperexcitatory effects by prolonging the open time of individual neuronal sodium channels. Occupational exposure to … eriments were performed using rat dorsal root ganglion neurons and cerebellar Purkinje cells. (+/-)-alpha-Tocopherol (vitamin E) selectively blocked the tetramethrin(type I pyrethroid)-modified sodium channels in a dose-dependent, but voltage-independent manner without affecting normal sodium channels. The concentration-response curves for tetramethrin modificatio …

Alpha-tocopherol reduces doxorubicin-induced toxicity in rats--histological and biochemical evidences.

The beneficial effect of alpha-tocopherol on doxorubicin-induced toxicity was studied in rats. alpha-Tocopherol (400 mg/kg/day) was administered orally, daily for a period of 2 months along with/without doxorubicin (2.5 mg/kg, i v weekly once for 8 weeks). Histology showed liver necrosis, heart myocyte degeneration, glomerular and tubular degeneration, cellular infiltration and desquammation of intestinal mucosa in doxorubicin treated animals. There was a significant increase in lipid peroxide levels measured in terms of "TBA reactants" in all these organs. These changes were associated with e…

Reduction of benzo(a)pyrene induced chromosomal aberrations by DL-alpha-tocopherol.

The antioxidant, DL-alpha-tocopherol (vitamin E), has been demonstrated to significantly reduce the percentage of benzo(a)pyrene (BP) induced chromosomal aberrations in vitro. Chinese hamster lung (Don) and Chinese hamster ovary (CHO) cells were treated with either 1 microgram/ml or 5 micrograms/ml BP for 4 to 28 h; some cultures were treated with S9 mix activated BP. Additional cultures of Don and CHO were treated simultaneously with 100 micrograms/ml of DL-alpha-tocopherol and BP. In CHO cells 1 microgram/ml non-activated BP significantly increased the chromosomal aberration percentage above…

Alpha-Tocopherol from People to Plants Is an Essential Cog in the Metabolic Machinery.

Significance: Protection from oxygen, a diradical, became a necessity with the evolution of photosynthetic organisms about 2.7 billion years. α-Tocopherol plays an essential, protective role in organisms from plants to people. An overview of human conditions that result in severe vitamin E (α-tocopherol) deficiency is provided. Recent Advances: α-Tocopherol has a critical role in the oxygen protection system by stopping lipid peroxidation, its induced damage, and cellular death by ferroptosis. Recent findings in bacteria and plants support the concept of why lipid peroxidation is so dangerous

In vivo adverse effects of alpha-tocopherol on the semen quality of male bucks.

Oxidative stress has detrimental effects on semen quality during spermatogenesis and semen processing for artificial insemination. This work was conducted to study the effect of different levels of vitamin E on the semen traits, oxidative status and trace minerals in Beetal bucks. Thirty-six bucks of similar body weight and age (1 year) were randomly divided into four groups. One group was kept as control with no supplementation (group 1), and the others were supplemented with 200 (group 2), 400 (group 3) and 800 IU (group 4) vitamin E/animal/day for 2 months. At the end of the experiment, sem

Enhancement of the antibiotic activity of aminoglycosides by alpha-tocopherol and other cholesterol derivates.

Alpha-tocopherol, one of the most abundant isoforms of vitamin E, is a biologically active liposoluble vitamin and potent antioxidant. It occurs naturally in foods of plant and animal origin. Because of its lipophilic character, it can cause perturbations in the bacterial cell membrane, resulting in damage to components essential for the integrity of the membrane, thereby allowing an increase in permeability. This is the first report of the modulatory effect of alpha-tocopherol in multiresistant bacteria. We evaluated alpha-tocopherol against multiresistant strains of Staphylococcus aureus, Ps…

Correlation of vitamin A nutritional status on alpha-tocopherol in the colostrum of lactating women.

The adequate supply of vitamins A and E to newborns is essential. However, factors such as maternal nutritional status and nutrient interaction may limit its bioavailability. The aim of this study was to establish nutritional status for vitamins A and E and evaluate the correlation of retinol on colostrum alpha-tocopherol in lactating women. A total of 103 lactating women were recruited at a Brazilian public maternity hospital. Fasting serum and colostrum samples were collected in the immediate post-partum. Retinol and alpha-tocopherol levels were determined by high-performance liquid chromato…

[The effect of alpha-tocopherol on blood lipid peroxidation in patients with hepatic echinococcosis].

Patients with hydatid disease of the liver were found to have disorders of lipid metabolism in the liver which led to increase in the amount of lipid peroxidation products and simultaneous decrease of blood serum antioxidant activity. An operation sharply induces the noted disturbances of lipid metabolism. A single administration of 1 ml of 10% alpha tocopherol solution in the preoperative period has a beneficial effect on the above-noted shifts.

Effects on neural function of repleting vitamin E-deficient rats with alpha-tocopherol.

A severe and chronic deficiency of vitamin E (alpha-tocopherol) is associated with a characteristic neurological syndrome with typical "clinical," neuropathological, and electrophysiological abnormalities in both humans and experimental animals. Repletion of vitamin E-deficient human subjects with alpha-tocopherol typically halts the progression of the neural signs and symptoms, and in some cases, can result in objective improvement. Electrophysiological parameters provide an objective measure of neural and visual function and improvement of some of these measures has been reported after reple…

Protective role of ascorbic acid and alpha-tocopherol on arsenic-induced microsomal dysfunctions.

Arsenic, a naturally occurring element, is present in food, soil, air and water. All human populations are exposed to arsenic and its compounds through occupational or environmental processes. Since arsenic compounds have been shown to exert their toxicity chiefly by generating reactive oxygen species, we have evaluated the effect of ascorbic acid and alpha-tocopherol on oxidative damage, antioxidant status and on xenobiotic metabolizing systems in arsenic-exposed rat liver and kidney microsomes. Arsenic exposure increases oxidative damage to lipids and proteins and decreases the levels of ant…

Anti-ulcer effects of antioxidants, quercetin, alpha-tocopherol, nifedipine and tetracycline in rats.

When free radical-scavenging activities of quercetin, alpha-tocopherol, nifedipine and tetracycline were measured by an electron spin resonance technique, all test compounds (10(-5) to 10(-3) M) scavenged both superoxide anions and hydroxyl radicals. The oral administration of quercetin (50 and 100 mg/kg), alpha-tocopherol (8 and 16 mg/kg), nifedipine (20 and 40 mg/kg) or tetracycline (10 and 20 mg/kg) markedly prevented the HCl plus ethanol-induced gastric mucosal injury and the increase in the content of thiobarbituric acid-reactive substances in the injured mucosa in rats. In addition, quer…

Cytoprotective effects of alpha tocopherol against liver injury induced by extrahepatic biliary obstruction.

There are several important aetiologies of extrahepatic biliary obstruction (EBO). If EBO is surgically reconstructed in the critical time period, liver damage can be halted or reversed. In this golden period, lipid peroxidations significantly intensify liver defects. We hypothesised that alpha tocopherol (alpha-T) could protect the liver from the damage caused by response to EBO. In standard conditions, albino rats of Wistar strain were divided into two groups. All rats underwent double ligations and divisions of common bile ducts (CBD). One of these groups received alpha-T (CBDL-alpha-T). Th

alpha-Tocopheryl hemisuccinate administration increases rat liver subcellular alpha-tocopherol levels and protects against carbon tetrachloride-induced hepatotoxicity.

Rats were administered a series of tocopherol analogs 18 h prior to a hepatotoxic dose of carbon tetrachloride (CCl4). Of the compounds tested, only d-alpha-tocopheryl hemisuccinate (TS) provided significant protection against CCl4-induced hepatotoxicity. No protection was observed with either d-alpha-tocopherol (alpha-T) or a tocopherol succinate ether derivative, d-alpha-tocopheryloxybutyric acid (TSE). None of the tocopherol analogs significantly inhibited CYP2E1 activity as measured by oxidation of p-nitrophenol. Liver homogenates and subcellular fractions (cytosol, nuclei, plasma membrane…
