We measured CSF levels of beta-endorphin, an opioid hormone, in 19 patients with infantile autism and in 3 patients with Rett syndrome, and compared them with control values. In infantile autism, CSF levels of beta-endorphin did not differ significantly from those of age-matched controls. There was no significant correlation between CSF levels and clinical symptoms, including self-injurious behavior, pain insensitivity, and stereotyped movement. In an attempt to determine whether plasma beta-endorphin (beta-ED) concentrations correlate with occurrence of apnea in preterm infants, measurements were made in three groups of infants. The effect of chronic haloperidol on brain beta-endorphin immunoreactivity was examined in rats. Chronic haloperidol resulted in significant reductions of beta-endorphin immunoreactivity within the striatum, but other brain regions were uneffected. When a variety of neuroleptic drugs were tested, fluphenazine, chlorpromazine and haloperidol resulted in comparable reductions in beta-endorphin immunoreactivity in the striatum. These findings suggest a functional interaction between beta-endorphin and dopamine systems in the striatum.

We describe a sensitive beta-endorphin (beta-EP) radioimmunoassay specific for beta-EP 1-31 applied to human ventricular and lumbar cerebrospinal fluid (CSF).
