The adenosine A(2A) receptor knockout mouse: a model for anxiety?

The main behavioural features of the adenosine A(2A) receptor knockout mouse include anxiety, aggressiveness in males and a paradoxical response to caffeine. These behavioural characteristics caused by the lack of adenosine A(2A) receptor function in mice correspond to the effects of functional antagonism of adenosine A(2A) receptors in humans and rodents. Increased anxiety in patients with panic disorder and increased psychotic symptomatology in patients with schizophrenia have been observed after caffeine administration. Several hypotheses have been developed suggesting a reduced adenosine A…

Response to prejunctional adenosine receptors is dependent on stimulus frequency.

PURPOSE: To characterize the adenosine receptor modulation of norepinephrine (NE) release from sympathetic neurons in the isolated rabbit iris/ciliary body. METHODS: Iris/ciliary bodies were isolated from New Zealand White rabbits and incubated in the presence of 3H-NE. Norepinephrine release was elicited by field stimulation with varied frequencies from 5 to 30 Hz. The effects of adenosinergic and alpha 2 adrenergic compounds on NE release were determined and compared. RESULTS: At a stimulation frequency of 5 Hz, the addition of the adenosine A1 agonist CHA did not significantly alter 3H-NE…

Role of adenosine in the diagnosis and treatment of tachyarrhythmias in pediatric patients.

Tachyarrhythmias are common rhythm disturbances in infants and children. Despite the availability of diagnostic criteria arrhythmias are sometimes commonly misdiagnosed. Recent reports suggest that an endogenous purine nucleoside, adenosine, has a diagnostic effect in narrow QRS complex tachycardias, in addition to terminating supraventricular tachycardia involving the atrioventricular node. This report reviews the authors' experience with the use of adenosine for diagnosis of narrow and wide complex tachyarrhythmias in children. Adenosine was administered to 43 patients with several types of …

[Cardiovascular effects and underlying mechanisms of adenosine and its analogues].

The present work was aimed to define the cardiovascular effects and underlying mechanisms of adenosine and its analogues. The results showed that (1) adenosine and 2-chloroadenosine could induce an initial increase in MAP mediated by the adenosine A2 receptors in carotid body chemoreceptor, and a subsequent decrease in MAP, being attributed to the adenosine A1 receptors in heart and A2 receptors in blood vessels; (2) N6-cyclopentyladenosine (CPA, a selective adenosine A1 receptor agonist) inhibited the electrophysiological activity of pacemaker cells in sinoatrial node; (3) CPA markedly attenu…

Therapeutic use of theophylline to antagonize renal effects of adenosine.

Experiments in laboratory animals clearly show that adenosine acts as a vasoconstrictive metabolite in the kidney. Adenosine receptor antagonists like theophylline can inhibit renal vasoconstriction in response to exogenous and endogenous adenosine. Based on these findings a number of experiments have been performed to test whether the vasoconstrictive action of adenosine in the kidney might be important also in pathophysiological states. In various animal models theophylline and other methylxanthine derivatives have been successfully employed to improve renal function after induction of acute…

Metabolism of exogenous cyclic AMP to adenosine in the rat kidney.

Although adenosine contributes importantly to the regulation of renin release, renal vascular resistance and renal tubular reabsorption, the metabolic pathways that control the intrarenal production rate of adenosine remain ill defined. The objective of this study was to determine whether extracellular metabolism of cyclic AMP to AMP by extracellular phosphodiesterase and hence to adenosine by ecto-5'-nucleotidase can occur in the intact kidney. To test this hypothesis, five experimental series were conducted in kidneys from male Sprague-Dawley rats perfused in a nonrecirculating system (5 ml/…

Adenosine analogues modulate the incidence of sleep apneas in rats.

The effects of adenosine A1 and A2 agonists on spontaneous central sleep apneas in rats have been examined by simultaneously monitoring sleep and respiration in freely moving unanesthetized animals. Intraperitoneal administration of 1.0 mg/kg of the A1 receptor agonist R(-)N6-L-(2-phenyl-isopropyl)adenosine (L-PIA) and 150 and 300 micrograms/kg of 2-p-(2-carboxyethyl)phenethylamino-5'-N-ethylcarboxamido-adenosine hydrochloride (CGS 21680), a selective A2 agonist, decreased the apnea index (AI) during sleep. Within a sleep period, AI increased over time in both control and drug-treated animals.…

Use of adenosine deaminase as a diagnostic tool for tuberculous pleurisy.

BACKGROUND: A statistical audit of adenosine deaminase (ADA) in pleural effusions was undertaken. METHODS: ADA analysis, cytological and microbiological examinations, and differential cell counts were performed on 462 pleural fluid samples. RESULTS: ADA activity in tuberculous effusions was higher than in any other diagnostic group. At a level of 50 U/l the sensitivity and specificity for the identification of tuberculosis was 90% and 89%, respectively. CONCLUSIONS: ADA activity remains a useful test in the evaluation of pleural effusions.

Use of adenosine for diagnosis and treatment of tachyarrhythmias in pediatric patients.

This report reviews our experience with the use of adenosine for diagnosis and treatment of narrow QRS complex tachyarrhythmias in children. All electrocardiograms obtained since the introduction of adenosine for clinical use at one pediatric tertiary care institution during an 18-month period were reviewed, and those patients receiving adenosine were included for study. Of the 24 patients who received adenosine, the median age was 4 years; four neonates were included. Adenosine produced atrioventricular block in 21 (88%) of 24 patients. It terminated the tachyarrhythmia in 11 patients and pro…

Therapeutic and diagnostic efficacy of adenosine in wide-complex tachycardia.

Wide-complex tachycardia is one of the most challenging problems faced by emergency physicians. Adenosine is an endogenous nucleoside that has gained recent popularity as an anti-arrhythmic drug in the setting of supraventricular tachycardia. We present the cases of two patients with stable wide-complex tachycardia in which adenosine was administered for therapeutic and diagnostic effects. Both patients quickly converted into sinus rhythm with subsequent diagnoses of atrioventricular reciprocating tachycardia secondary to Wolff-Parkinson-White syndrome. The role of adenosine in the acute manag…

Adenosine: a new drug for acute termination of supraventricular tachycardia.

Adenosine, recently released for the acute termination of supraventricular tachycardia, is potentially the drug of first choice for this indication. Its notable effects are transient slowing of the sinus rate and atrioventricular nodal block. Adverse effects are transient and clinically insignificant, primarily because of the ultra-short half-life of the drug. Preliminary investigations suggest that adenosine may also be useful in the differential diagnosis of wide QRS complex tachycardias.

Adenosine and its analogs stimulate phosphoinositide hydrolysis in the kidney.

Renal blood flow, glomerular filtration rate and sodium excretion are known to be affected by adenosine. The present studies were undertaken to investigate the actions of adenosine and its analogs (both agonists and antagonists) on phosphoinositide (PI) hydrolysis in the outer medullary slices. Adenosine was found to cause a dose-dependent stimulation of PI hydrolysis (ED50, 2.8 microM) in renal slices from outer medulla. The adenosine analogs 5'-(N-cyclopropyl)-carboxamidoadenosine (NCCA) and 5'-N-ethylcarboxamidoadenosine (NECA) also stimulated PI hydrolysis in renal medulla. Stimulation of …

Role of endogenous adenosine in postdefibrillation bradyarrhythmia and hemodynamic depression.

In halothane-nitrous oxide-anesthetized pigs, the effect of the competitive adenosine antagonist, BW-A1433U (a derivative of 1,3-dipropyl-8-phenylxanthine), on postdefibrillation bradyarrhythmia and hemodynamic depression was investigated. In protocol 1, repetitive episodes of ventricular fibrillation lasting 15 seconds before transthoracic DC shock were performed in five animals, before (control) and after the administration of BW-A1433U (5 mg/kg i.v.). An unsuccessful initial shock was immediately followed by a rescue shock of 40 A. In ventricular fibrillation episodes requiring rescue shock…
