Neural regulation and manipulation of pancreatic β cells.

Pancreatic β cells are regulated by integrative neural signals beyond simple autonomic efferent control. Emerging evidence highlights the critical roles of afferent nerve signals and the gut-liver-brain-pancreas network in regulating β cells. Harnessing these intrinsic neural pathways may offer innovative therapeutic strategies for restoring β cell function and mass.

Renal interoception in health and disease.

Catheter based renal denervation has recently been FDA approved for the treatment of hypertension. Traditionally, the anti-hypertensive effects of renal denervation have been attributed to the ablation of the efferent sympathetic renal nerves. In recent years the role of the afferent sensory renal nerves in the regulation of blood pressure has received increased attention. In addition, afferent renal denervation is associated with reductions in sympathetic nervous system activity. This suggests that reductions in sympathetic drive to organs other than the kidney may contribute to the non-renal

Responsiveness of afferent renal nerve units in renovascular hypertension in rats.

Previous data suggest that renal afferent nerve activity is increased in hypertension exerting sympathoexcitatory effects. Hence, we wanted to test the hypothesis that in renovascular hypertension, the activity of dorsal root ganglion (DRG) neurons with afferent projections from the kidneys is augmented depending on the degree of intrarenal inflammation. For comparison, a nonhypertensive model of mesangioproliferative nephritis was investigated. Renovascular hypertension (2-kidney, 1-clip [2K1C]) was induced by unilateral clipping of the left renal artery and mesangioproliferative glomerulonep

Evidence for multiple bulbar and higher brain circuits processing sensory inputs from the respiratory system in humans.

KEY POINTS: Unpleasant respiratory sensations contribute to morbidity in pulmonary disease. In rodents, these sensations are processed by no … jugular pathways in respiratory sensory processing. However, it is unknown whether bronchopulmonary afferent pathways are similarly arranged in humans. We set out to investigate this using high resolution brainstem and whole brain functional magnetic resonance imaging in healthy human participants when they were inhaling stimuli known to differentially activate nod …

THE ANATOMY OF OROFACIAL INNERVATION.

The whole human body receives rich sensory innervation with only one exception and that is the brain tissue. The orofacial region is hence no exception. The head region consequently receives a rich network of sensory nerves making it special because the two types of sensory fibres, visceral and somatic overlap, especially in the pharynx. Also, different pain syndromes that affect this region are rather specific in comparison to their presentation in other body regions. With this review article we wanted to show the detailed anatomy of the peripheral sensory pathways, because of its importance

Sensory neuronopathies.

The sensory neuronopathies (or ganglionopathies) are a small subcategory of neuropathies characterized by primary degeneration of the dorsal root ganglia and trigeminal ganglion sensory neurons, resulting in a distinctive clinical presentation. Patients typically have subacute onset of asymmetric, non-length-dependent sensory impairment and early ataxia. The etiologies of acquired sensory neuronopathies are rather limited. Early identification is imperative, as they may herald an underlying malignancy or an autoimmune condition such as Sjögren syndrome. This review addresses the various causes

Central control of autonomic function and involvement in neurodegenerative disorders.

The central autonomic network includes the insular cortex, anterior cingulate cortex, amygdala, hypothalamus, periaqueductal gray, parabrachial nucleus, nucleus of the solitary tract, ventrolateral reticular formation of the medulla, and medullary raphe. These areas: are reciprocally interconnected; receive converging visceral and somatosensory information; generate stimulus-specific patterns of autonomic, endocrine, and motor responses; and are regulated according to the behavioral state, including the sleep-wake cycle. Several components of the central autonomic networks are affected in neur

Caffeine ingestion causes detrusor overactivity and afferent nerve excitation in mice.

PURPOSE: We examined the effect of caffeine (Sigma®) on voiding patterns in mice and characterized potential changes in bladder function and sensory signaling. MATERIALS AND METHODS: A total of 12 mice were fed high dose (150 mg/kg) caffeine daily for 2 weeks. Micturition frequency and volume were recorded at baseline and at the end point. The effects of chronic low dose (10 mg/kg) caffeine on voiding patterns were examined in 7 mice, which were subsequently studied using awake cystometry. In a separate study to characterize the effects of acute caffeine consumption on bladder function and se

Sensing pulmonary oxidative stress by lung vagal afferents.

Oxidative stress in the bronchopulmonary airways can occur through a variety of inflammatory mechanisms and also following the inhalation of environmental pollutants. Oxidative stress causes cellular dysfunction and thus mammals (including humans) have developed mechanisms for detecting oxidative stress, such that defensive behavior and defensive biological mechanisms can be induced to lessen its potential damage. Vagal sensory nerves innervating the airways play a critical role in the detection of the microenvironment in the airways. Oxidative stress and associated compounds activate unmyelin

Compartmentalization of the deep cerebellar nuclei based on afferent projections and aldolase C expression.

The distribution of aldolase C (zebrin II)-positive and -negative Purkinje cells (PCs) can be used to define about 20 longitudinally extended compartments in the cerebellar cortex of the rat, which may correspond to certain aspects of cerebellar functional localization. An equivalent compartmental organization may exist in the deep cerebellar nuclei (DCN). This DCN compartmentalization is primarily represented by the afferent projection pattern in the DCN. PC projections and collateral nuclear projections of olivocerebellar climbing fiber axons have a relatively localized terminal arbor in the

Perception of gastro-oesophageal reflux.

Gastro-oesophageal reflux disease (GORD) is common and often associated with unpleasant symptoms requiring utilisation of health care resource. While in the majority of patients symptom resolution occurs with acid suppressant therapy, in a proportion this treatment is ineffective in resolving symptoms. This is particularly the case in patients with non-erosive reflux disease (NERD) and functional heartburn (FH). It is increasingly being recognised that the presence of acid in the oesophagus can cause dilated intercellular spaces (DIS) which increases the exposure of the sub-epithelial nerves t

Why is the neurobiology of nausea and vomiting so important?

Nausea and vomiting are important as biological systems for drug side effects, disease co-morbidities, and defenses against food poisoning. Vomiting can serve the function of emptying a noxious chemical from the gut, and nausea appears to play a role in a conditioned response to avoid ingestion of offending substances. The sensory pathways for nausea and vomiting, such as gut and vestibular inputs, are generally defined but the problem of determining the brain's final common pathway and central pattern generator for nausea and vomiting is largely unsolved. A neurophysiological analysis of brai

Internally driven control of reaching movements: a study on a proprioceptively deafferented subject.

We investigated the possibility of controlling reaching movements on the sole basis of central mechanisms, i.e., without peripheral feedback on hand and target positions. A deafferented subject (GL) and control subjects reached with the unseen hand for a straight-ahead target that could be displaced laterally at movement onset. The shifted target was continuously or briefly lit, or not visible. In this latter condition, a beep from either side of subjects' head single-handedly signaled the change in the movement goal, so that movements could only be controlled through an internal representatio

Electrophysiological studies on the effects on single cells of silver electrodes implanted in the visual cortex of cats.

We have studied whether the presence of an artificial metal conductor in the visual cortex has an effect on the responsiveness pattern of the cells there. The physiological properties of single cells have been characterized in the mammalian primary visual cortex, following the acute and chronic implantation of silver wire electrodes. The Experimental group was Chronic and Acute operated and implanted adult cats. The Control group consisted of operated, but not implanted, adult cats and intact (Normal) adult cats. A sagittal incision was unilaterally made in the primary visual cortex (area 17)

Neonatal damage to the rat's infraorbital nerve upregulates both galanin and neuropeptide Y in individual vibrissae-related primary afferent axons.

Previous studies in adult animals have suggested that the peptides galanin and neuropeptide Y (NPY) may be upregulated in the same primary afferent neurons after peripheral axotomy. The present study was undertaken to determine whether such upregulation occurred in vibrissae-related primary afferent neurons and their axons after damage to the infraorbital nerve [ION; the trigeminal (V) branch that innervates the vibrissae follicles]. Double-labelling experiments demonstrated that approximately 75% of axotomized V ganglion cells and the central arbors of vibrissae-related primary afferents expr

Spinal projections to the parabrachial nucleus are substance P-immunoreactive.

Substance P plays an important role in nociceptive processing in the spinal cord. Substance P is also present in several supraspinal regions, such as the pontine parabrachial nucleus, a major relay for autonomic regulation. In this study we examined in the cat with an immunogold method the presence of substance P-like immunoreactivity in spinoparabrachial terminals that were labelled by anterograde transport of unconjugated and lectin-conjugated horseradish peroxidase. We found that dense core vesicles in anterogradely labelled terminals were substance P-immunoreactive. Taken together with pre

Effect of multiple denervations on the exercise hyperpnea in awake ponies.

In three previously reported studies, we had documented that the normal exercise hyperventilation in ponies is accentuated by carotid body denervation (CBD), not affected by hilar nerve pulmonary vagal denervation (HND), and mildly attenuated by spinal cord ablation of the dorsal lateral columns at L2 (SA). In the present study, we hypothesized that if redundancy of control existed in exercising ponies, then multiple denervations of theoretically important pathways in the same animal might attenuate the ventilatory response to exercise in a way not predictable by the individual lesion experime

Neurons projecting rostrally in the hypogastric nerve of the cat.

Afferent, pre- and postganglionic nerve cell bodies projecting rostrally in the hypogastric nerve have been labelled retrogradely with horseradish peroxidase, in order to study number and location of these neurons. Labelled somata were found in the ganglia of the pelvic plexus, in sacral spinal ganglia and in the sacral spinal cord. They were located mainly ipsilateral with a maximum of afferent and preganglionic neurons at the segmental level S2. The numbers of labelled cells were small (less than 3.5% of all axons in the hypogastric nerve) and varied considerably between animals. Therefore t

Sexual behavior of male dairy goats: effects of deafferentation of the genitalia.

The following study examines the effects of deafferentation of the penis, scrotum, prepuce and adjacent cutaneous surfaces on the development of mounting behavior in male dairy goats. Ten males underwent surgical deafferentations and 10 males received sham operations at 8 to 12 weeks of age. Mounting and other sexual behaviors were monitored while subjects were cohabiting with male and female penmates and when individually exposed to estrous females. Denervated males mounted penmates less frequently than sham controls and lacked vigorous thrusting behavior. When exposed to estrous females dene
