Comparative trials using albendazole and mebendazole in the treatment of soil-transmitted helminths in schoolchildren on Penang, Malaysia.

Trials using albendazole and mebendazole, as single 400 mg dose treatments, against soil-transmitted helminths, were carried out in 7-9 and 10-12 years-old schoolchildren living in urban and rural environments in Penang, Malaysia. Both drugs were equally effective in treating trichuriasis and ascariasis in both age groups and environments. However, mebendazole is not so effective in the treatment for hookworms when compared to albendazole. It is suggested that albendazole should be considered the drug of choice for mass chemotherapy for Penang.

Vitamin A status of Indonesian children infected with Ascaris lumbricoides after dosing with vitamin A supplements and albendazole.

In developing countries, both marginal vitamin A status and intestinal helminths are common among children. Indonesian children (n = 309, 0.6-6.6 y), known to be infected with Ascaris lumbricoides, were randomized into six different treatment groups (A-F). The treatments included 210 mumol vitamin A supplement and a dose of 400 mg albendazole (5-propylthio-1H-benzimidazol-2-yl carbamic acid methyl ester) administered orally either at the same health visit (Groups B and F) or at different contact times during a 1-mo period (groups A, C, D and E). Vitamin A status was assessed both before and 3-…

Development of an injectable formulation of albendazole and in vivo evaluation of its efficacy against Echinococcus multilocularis metacestode.

The loading of poly (D, L-lactide) nanoparticles with ABZ has led us to evaluate the potential of this new colloidal drug delivery system against E. multilocularis, using a murine model of hepatic alveolar echinococcosis. ABZ-loaded nanoparticles had a monodisperse size distribution between 100 and 150 nm. The efficiency of drug loading to nanoparticles was over 97%. In vitro, at an ABZ concentration of 1.0 microgram ml-1, the formulation had no toxicity for peritoneal macrophages harvested from uninfected mice. In vivo, the ABZ-loaded nanoparticles exhibited no signs of toxicity at any of the

Rate of reinfection with intestinal nematodes after treatment of children with mebendazole or albendazole in a highly endemic area.

The comparative efficacy of albendazole and mebendazole in the treatment of intestinal nematode infections were compared 3 weeks after treatment in a randomized trial among schoolchildren on Pemba Island, Tanzania. Egg counts were compared 3 weeks, 4 months and 6 months after treatment of 731 children seen on each occasion. Differences in the efficacies were apparent with some nematodes 21 d after treatment, but these were no longer apparent 4 months after treatment, and by 6 months intensities of infection were similar to pre-treatment levels. These findings suggest that treatment of schoolch…

Treatment of Strongyloides stercoralis infection with ivermectin compared with albendazole: results of an open study of 60 cases.

Ivermectin is highly effective against animal intestinal nematodes and is used in the treatment of onchocerciasis in humans. A study was undertaken to compare the efficacy of the drug with that of albendazole in the treatment of uncomplicated strongyloidiasis. Sixty patients with confirmed Strongyloides stercoralis infection were enrolled in an open randomized study and given either albendazole, 400 mg/d for 3 d or ivermectin, 150-200 micrograms/kg in a single dose. Efficacy and tolerance were evaluated on days 7, 30 and 90. Each visit included a parasitological examination of 3 stool specimen…

Comparative efficacy of a single 400 mg dose of albendazole or mebendazole in the treatment of nematode infections in children.

The relative efficacy of a single 400 mg dose of albendazole or mebendazole in the treatment of nematode infections was assessed in 2- to 9-year-old children living in two different Bolivian rural communities. Both agents were equally very effective (100% cure rate) in treating ascariasis. Albendazole was clearly more active than mebendazole against hookworm infections, both in terms of egg reduction rate (92.8% vs. 62.4%) and cure rate (81.8% vs. 17.2%). As far as trichuriasis is concerned, albendazole produced a higher egg reduction rate than mebendazole (45.7% vs 15%), but a lower cure rate…

Efficacy of albendazole against Strongyloides ratti and S. stercoralis in vitro, in mice, and in normal and immunosuppressed dogs.

In view of conflicting clinical reports, the effects of albendazole on Strongyloides were examined in vitro and in experimentally-infected animals. Albendazole inhibited the hatching and moulting of S. ratti eggs and larvae and prevented the development in vitro of S. stercoralis first-stage larvae. Pre-exposure to the drug did not impair the infectivity of either S. ratti or S. stercoralis third-stage larvae. Albendazole had a dose-dependent inhibitory effect on S. ratti migrating larvae in mice when measured in terms of the numbers of larvae recovered from the skin or lungs or the subsequent…

Amorphization of Albendazole Using Organic Acid-based Solid Dispersions for Improved Dissolution: A Solvent-free Approach.

Albendazole (ABZ), a broad-spectrum anthelmintic drug, exhibits poor aqueous solubility and low oral bioavailability (< 5%), limiting its therapeutic effectiveness. This study introduces an organic solvent-free strategy to enhance ABZ dissolution by preparing solid dispersions (SDs) with weak organic acids-citric acid (CA), tartaric acid (TA), and glutaric acid (GA). pH-dependent solubility analysis showed that all three acids improved ABZ solubility, with CA achieving the most pronounced effect-a 188,000-fold increase compared to intrinsic solubility of ABZ (0.25 µg/mL). SDs were formulated a…

Synthesis, Molecular Docking Studies and ADME Prediction of Some New Albendazole Derivatives as α-Glucosidase Inhibitors.

A series of novel 2-(substituted arylidene)-N-(5-(propylthio)-2,3-dihydro-1H-benzo[d]imidazol-2-yl)hydrazine-1-carboxamide derivatives 3a-i were synthesized via condensation of N-(5-(propylthio)-1H-benzo[d]imidazol-2-yl) hydrazinecarboxamide (2), with the corresponding ketone or aldehydes. The chemical structures of the compounds prepared were confirmed by analytical and spectral data. The compounds were screened for their α-glucosidase inhibitory activity and all of them showed better inhibition than acarbose, except 3h. In particular, compound 3a proved to be the most active compound among a

Copper nanoparticles: Biosynthesis, characterization, and protoscolicidal effects alone and combined with albendazole against hydatid cyst protoscoleces.

BACKGROUND: Surgery remains the preferred treatment option for hydatid cyst (cystic echinococcosis); however, recent studies have demonstrated that the current protoscolicidal agents used during surgery are associated with some adverse side effects such as biliary fibrosis, hepatic necrosis, and cirrhosis. The present study aims to evaluate the in vitro and ex vivo anti-parasitic effects of copper nanoparticles (CuNPs) alone and combined with albendazole on hydatid cyst protoscoleces. METHODS: CuNPs was green synthesized using C. spinosa extract. Various concentrations of CuNPs (250, 500, and…

Post-market surveillance of albendazole tablet brands in Kenya: Implications for deworming programs and clinical outcomes.

BACKGROUND: Albendazole is a broad-spectrum anthelminthic extensively used in clinical settings and national deworming programs. It is the cornerstone of the preventive chemotherapy for soil-transmitted helminthiases prevalent in low-resource settings. The high proliferation of albendazole generic products, with poor post-market surveillance capacity in the low- and middle-income countries, poses dire risk of substandard and falsified medicines, which may predispose patients to treatment failures, adverse drug reactions, morbidity, and mortality, with consequent loss of public confidence in he…

Efficacy of two rounds of albendazole treatment on soil-transmitted helminths in schoolchildren, Yunnan Province, China.

Mass drug administration (MDA) of albendazole to at-risk populations remains the primary strategy for controlling soil-transmitted helminths (STH). Despite its widely use, its efficacy varies among different STH species and remains sub-optimal, particularly in the treatment of T. trichiura. Currently, studies investigating the optimal dose and regimens for albendazole are lacking. A longitudinal cohort study was conducted to assess the efficacy of two single-dose albendazole 400 mg treatments given four weeks apart targeting STH infections compared with just one single-dose albendazole 400 mg …
