Delta-opioid receptor antagonists attenuate motor activity induced by amphetamine but not cocaine.

Naloxone and naltrindole attenuate the locomotor response to amphetamine, implicating delta-opioid receptors in the opioid-antagonist/amphetamine interaction. To determine the role of delta-opioid receptor subtypes in this phenomenon, rats were pretreated with the following selective antagonists administered intracisternally: naltrindole, [D-Ala2,Leu5,Cys6]enkephalin (DALCE, delta 1 receptor selective), naltrindole-5'-isothiocyanate (delta 2 receptor selective). Cumulative dose-response curves to amphetamine were constructed (saline, 0.1, 0.4, 1.6 and 6.4 mg/kg s.c.), with injections every 30 …

Opioid modulation of amphetamine-stimulated dopamine release and concentration in rat striatal slices.

The effects of morphine and naltrexone on amphetamine-stimulated release and total concentration of dopamine (DA) from rat striatal slices in vitro were examined in this study. Adult male Sprague-Dawley rats were sacrificed and the striata were dissected, sliced, and then incubated in buffer solution at 37 degrees C with amphetamine in the presence or absence of various concentrations of morphine, naltrexone (or both), or the dihydroxyphenylalanine (DOPA) decarboxylase inhibitor (NSD-1015). The concentrations of DOPA, DA, and dihydroxyphenylacetic acid (DOPAC) in the tissue slices and buffer m…

Endocrine, immune, and neurochemical changes in rats during withdrawal from chronic amphetamine intoxication.

In humans who chronically abuse amphetamine (AMPH), sudden abstinence often precipitates an organic mood disorder that mimics many symptoms of major depression. We report that AMPH exposure and withdrawal in rats modifies hypothalamic-pituitary-adrenal axis endocrine responses, peripheral immune functions, and regional brain catecholamine levels. Compared to vehicle-treated animals, rats treated with AMPH for 10 days exhibit significantly decreased physostigmine-induced release of adrenocorticotropin hormone (ACTH). During AMPH withdrawal, these animals show a loss of the normal correlation be…

Variable-ratio schedules of timeout from avoidance: effects of d-amphetamine and morphine.

Rats were trained on concurrent schedules in which pressing one lever postponed shock and pressing the other lever produced periods of signaled timeout from avoidance on variable-ratio schedules. These procedures generated high rates of timeout-reinforced responding and provided a baseline for studying the effects of drugs on behavior maintained by different types of negative reinforcement (shock postponement vs. timeout). Morphine (2.5 to 10.0 mg/kg) reduced behavior maintained by timeout at doses that increased or had no effect on avoidance responding. In contrast, d-amphetamine (0.125 to 2.…

Self-administration of phenylpropanolamine (PPA) by rats previously trained to self-administer amphetamine.

Central nervous system stimulation, similar to that observed for amphetamine, has been attributed to phenylpropanolamine (PPA). However, formal tests, using evaluation of locomotion or of self-administration, fail to reveal that PPA is a stimulant. Self-administration studies have trained rats to self-administer cocaine and then have attempted to switch rats to PPA with no success of transfer. The present study further examined the reinforcing properties of PPA (0.08, 0.16, and 0.32 mg/infusion, IV) in rats that were initially trained to self-administer d-amphetamine (0.005-0.08 mg/infusion) i…

Opioid influence on some aspects of stereotyped behavior induced by repeated amphetamine treatment.

Rats were administered repeated IP injections of dl-amphetamine (AMPH) according to a chronic escalating dose schedule (three doses per 24 hr, for four days, two days or one day). Animals treated for four days exhibited a diminished oral stereotypy in response to a challenge of 12 mg/kg AMPH or 2 mg/kg SC apomorphine (APO), 72 hr after withdrawal. Pretreatment with 2 mg/kg IP naloxone (NAL) during the period of chronic AMPH administration prevented the reduction in oral stereotypy induced by AMPH or APO. No differences were detected among the mean of stereotypy scores from the different treatm…

Discriminative stimulus properties of intragastrically administered d-amphetamine and pentobarbital in rhesus monkeys.

Rhesus monkeys were trained to discriminate intragastrically administered d-amphetamine (AMPH) or pentobarbital (PENTO) from saline using a signaled shock-avoidance trail procedure. All monkeys maintained criterion levels (greater than 90% drug-appropriate responding) throughout the duration of the study during training sessions. In the AMPH experiment, the anorectics diethylpropion, mazindol, phendimetrazine, phenmetrazine and phentermine completely substituted for the training dose of AMPH. The atypical antidepressant bupropion and the psychomotor stimulant methylphenidate also completely su…

Drug interactions on spontaneous locomotor activity in rats. Neuroleptics and amphetamine-induced hyperactivity.

The locomotor activity of female rats was recorded during 10-min sessions in a circular open-field apparatus after the administration of vehicle or drug. Dose-response curves were obtained for seven neuroleptic drugs both alone (spontaneous activity) and in combination with 1.0 mg/kg of d-amphetamine. Haloperidol, pimozide, loxapine, thiothixene, molindone and chlorpromazine all produced graded decreases in spontaneous locomotor activity. Haloperidol, pimozide, loxapine, thiothixene and molindone also produced graded reversals of the hyperactivity produced by d-amphetamine, while chlorpromazin…

A rare presentation of gastric ischemia manifesting as gastric pneumatosis in the context of amphetamine use.

Gastric pneumatosis is a rare condition characterized by gas within the stomach wall and can be associated with gastric ischemia. Gastric ischemia is uncommon due to the stomach's rich vascular supply but carries an increased risk of mortality. We present a rare case of a patient with amphetamine use disorder who was found to have gastric pneumatosis in the setting of gastric ischemia.

Locomotor and discriminative stimulus effects of fluorinated analogs of amphetamine and methamphetamine in mice and rats.

The prevalence of fluorinated amphetamine and methamphetamine analogs on the illicit market has continued to increase in the past decade. The perceived ability of these compounds to bypass legal regulation has resulted in an increasing popularity among drug users; however, their use produces significant adverse effects, including heart failure, cerebral hemorrhage, and death. This study aimed to investigate the effects of phenyl ring fluorination on the abuse potential of 5 synthetic stimulant compounds: 2- and 3-fluoroamphetamine (FA) and 2-, 3-, and 4-fluoromethamphetamine (FMA). The open-fi…

MDMA as well as amphetamine and alcohol increase feelings of social closeness in healthy adults.

Psychoactive drugs such as alcohol and stimulants are typically used in social settings such as bars, parties or small groups. Yet, relative … connectedness. To examine these relations, we investigated the effects of several drugs (MDMA, methamphetamine, alcohol) on feelings of connection between two strangers engaged in a conversation. We also investigated feelings of connection between two participants who discussed either 'shallow' or deeper topics in two conversations, without any drugs. All fo …

Expression and role of cocaine-amphetamine regulated transcript (CART) in the proliferation of biliary epithelium.

Cholangiocytes, the epithelial cells that line the biliary tree, can proliferate under the stimulation of several factors through both autocrine and paracrine pathways. The cocaine-amphetamine-regulated-transcript (CART) peptide has several physiological functions, and it is widely expressed in several organs. CART increases the survival of hippocampal neurons by upregulating brain-derived neurotrophic factor (BDNF), whose expression has been correlated to the proliferation rate of cholangiocytes. In the present study, we aimed to evaluate the expression of CART and its role in modulating chol…

Acute Myocardial Infarction Associated With Amphetamine Use and Smoking in a Young Healthy Individual.

One of the primary causes of death among methamphetamine users is cardiovascular disease, which is a result of the narrowing and spasm of blood vessels caused by the drug. This leads to increased blood pressure and heart rate, which can damage the heart muscle at the molecular level. The most common forms of chronic cardiovascular disease associated with methamphetamine use are coronary artery disease and cardiomyopathy. Here, we report a case of myocardial infarction (MI) due to amphetamine use and smoking in a young healthy male who developed ST-elevation myocardial infarction, ventricular f…

A rare case of renal infarction due to heroin and amphetamine abuse: case report.

BACKGROUND: Renal infarctions as a result of recreational drug use are rare and are commonly associated with cocaine use. Although amphetamines have a similar mechanism of action as cocaine, there are few reports linking them to ischemic events, and only one to renal infarction. Similarly, few reports link heroin use with infarcts, but never in the kidney. Although uncommon, several mechanisms have been implicated in heroin and amphetamine-induced infarction, including vasculopathy, vasculitis and the activation of the coagulation cascade. CASE PRESENTATION: 47-year-old female with a past med…

Wastewater analysis for nicotine, cocaine, amphetamines, opioids and cannabis in New York City.

According to current surveys and overdoses data, there is a drug crisis in the USA. Wastewater-based epidemiology (WBE) is an evolving discipline that analyses wastewater samples to detect drugs and metabolites to estimate drug consumption in a certain community. This study demonstrates how drug relative presence could be tracked by testing wastewater, providing real-time results, in different boroughs in New York City throughout 1 year. We developed and fully validated two analytical methods, one for 21 drugs and metabolites, including nicotine, cocaine, amphetamines, opioids and cannabis mar…
