Prevascularized Tracheal Scaffolds Using the Platysma Flap for Enhanced Tracheal Regeneration.

OBJECTIVES: One of the greatest hurdles in tracheal tissue engineering is insufficient vascularization, which leads to delayed mucosal regeneration, inflammation, and restenosis. This study investigated whether a prevascularized segmental tracheal substitute using platysma can enhance tracheal mucosal regeneration. METHODS: Three-dimensional (3D) printed scaffolds with (group M) or without (group S) Matrigel coating were implanted under the feeding vessels of the platysma in New Zealand White rabbits (n = 3) to induce vascularization. After 1 or 2 weeks, tracheal defects were created and vasc

Experimentally induced polyps in the sinus mucosa: a structural analysis of the initial stages.

To document polyp formation in the sinus mucosa, the authors of this study subjected New Zealand white rabbits to different modes of manipulation intended to induce inflammation of the maxillary sinus. These manipulations included a combination of bacterial infection and mechanical trauma, the deposition of agarose into the sinus cavity, and the deposition of N-formyl-methionyl-leucyl-phenylalanine, a chemotactic peptide, into the sinus cavity. A majority of animals developed polyps, which were examined by light and electron microscopy. Polyp formation appears to involve epithelial disruption

Echinococcus granulosus sensu lato in animal intermediate hosts: What is with the organ location?

Organ or tissue tropism/preference in many parasites is essential for their establishment, survival and pathogenesis. In protozoan parasites e.g. Trypanosoma and Plasmodium, tissue tropism is associated with many important aspects such as transmission, treatment, and pathogenesis. In these parasites, tissue tropism is achieved by mechanisms such as sequestration, extravasation, transcellular migration, and vascular permeability. In contrast, little is known about the mechanism behind organ tropism/preference and the potential implication in parasitic metazoans like Echinococcus. Since intraspe

Attenuating hypoxia driven malignant behavior in glioblastoma with a novel hypoxia-inducible factor 2 alpha inhibitor.

Hypoxia inducible factor (HIFs) signaling contributes to malignant cell behavior in glioblastoma (GBM). We investigated a novel HIF2α inhibitor, PT2385, both in vitro, with low-passage patient-derived cell lines, and in vivo, using orthotopic models of glioblastoma. We focused on analysis of HIF2α expression in situ, cell survival/proliferation, and survival in brain tumor-bearing mice treated with PT2385 alone and in combination with standard of care chemoradiotherapy. HIF2α expression increased with glioma grade, with over half of GBM specimens HIF2α positive. Staining clustered in perivascu

Si-Wu-Tang ameliorates chemotherapy-induced premature ovarian insufficiency by regulating a signaling pathway in endoplasmic reticulum stress.

BACKGROUND: Premature ovarian insufficiency (POI) is a clinically refractory reproductive system disorder. Our previous studies demonstrated that the traditional Chinese herbal formula Si-Wu-Tang (SWT) might ameliorate POI in animal models by improving ovarian angiogenesis, yet the underlying mechanisms remained unclear. PURPOSE: To explore the mechanisms of the traditional Chinese medicine (TCM) formula SWT on human umbilical vein endothelial cells (HUVECs) and animal models of POI. By studying its mechanisms, the research hopes to reveal the connotation of 'nourishing blood and activating b

Preclinical Animal Models for Temporomandibular Joint Tissue Engineering.

There is a paucity of in vivo studies that investigate the safety and efficacy of temporomandibular joint (TMJ) tissue regeneration approaches, in part due to the lack of established animal models. Review of disease models for study of TMJ is presented herein with an attempt to identify relevant preclinical animal models for TMJ tissue engineering, with emphasis on the disc and condyle. Although degenerative joint disease models have been mainly performed on mice, rats, and rabbits, preclinical regeneration approaches must employ larger animal species. There remains controversy regarding the p

The role of rapamycin in neural tube defects: insights from bioinformatics analysis and experimental validation.

Neural tube defects (NTDs) are the most severe congenital malformations of the central nervous system. Fetal developmental homeostasis disruptions heighten NTDs risk through aberrant maternal-fetal signaling and periconceptional medication exposure. Rapamycin, an inhibitor of the mTOR signaling pathway, is a widely used clinical drug whose effects on embryonic neural development remain unclear. This study aimed to identify the potential drug targets for rapamycin in embryonic neural development. 18 rapamycin-NTDs targets were obtained by the network pharmacology approach. Functional enrichment

Lactylation of GDF15 Regulates Angiogenesis in Ischemic Stroke via Activating PI3K/AKT/mTOR Pathway.

Ischemic stroke (IS) is a major health burden. The role of growth differentiation factor 15 (GDF15) in IS has been scarcely reported, and this study aimed to explore its expression and underlying mechanisms. The middle cerebral artery occlusion (MCAO) mice model and oxygen-glucose deprivation (OGD) mice brain microvascular endothelial cells (MBMVECs) models were established. Brain infarction size and neurological score were calculated following MCAO surgery. Cell counting kit-8, Transwell migration, and wound healing assays were performed to evaluate the cell viability, migration, and invasion

Excessive DAO inhibits myoblast migration, leading to impaired myotube fusion and muscle strength decline by reducing ECM.

INTRODUCTION: The molecular mechanisms underlying muscle strength decline, particularly in age-related conditions such as sarcopenia, remain poorly understood. Previous omics studies have suggested a potential association between AOC1 (encoding diamine oxidase, DAO, mainly expressed in intestine) and muscle weakness, but experimental validation and mechanistic insights are lacking. This study aims to investigate the influence of secretory DAO on muscle function and its potential receptors. METHODS: Serum DAO levels were measured in elderly participants (n = 129) using ELISA and correlated wit

Prenatal Downregulation of CB1 Cannabinoid Receptors in the Mouse Prefrontal Cortex Disrupts Cortical Lamination and Induces a Transcriptional Signature Associated with Social Interaction Deficits.

Endocannabinoid signaling exerts a neurodevelopmental regulatory role via CB1 cannabinoid receptors (CB1Rs), which control pyramidal neuron differentiation, migration, and axonal guidance. Here, we investigated the long-lasting consequences of transient prenatal CB1R downregulation within the mouse prefrontal cortex by assessing its impact on gene expression, neuronal electrophysiological properties, and animal behavioral traits. Transient loss of CB1Rs as induced by in-utero small-interference RNA electroporation at Embryonic Day 14.5, when upper-layer neurons are generated, arrested cell mig

Exposure to trichloromethane via drinking water promotes progression of colorectal cancer by activating IRE1α/XBP1 pathway of endoplasmic reticulum stress.

Trichloromethane (TCM), a commonly recognized disinfection by-product formed during the chlorination of water, has been associated with the onset of colorectal cancer (CRC) in humans. Despite this, the impact of TCM on the progression of CRC remains uncertain. In this investigation, it was observed that exposure to TCM could augment the migratory capabilities of CRC cells and facilitate the advancement of colorectal tumors. To delve deeper into the mechanism responsible for TCM-induced CRC progression, we performed RNA-Seq analysis at cellular and animal levels after TCM exposure. Both the KEG

Pogostone attenuates osteolysis in breast cancer by inhibiting the NF-kB and JNK signaling pathways of osteoclast.

AIMS: Breast cancer is the most prevalent cancer in females, and approximately 70 % of all patients have evidence of metastatic bone disease, which substantially affects the quality of life and survival rate of breast cancer patients. Osteoporosis has become a global public health problem, and the abnormal activation of osteoclasts is the key to the progression of osteoporosis and the key to both diseases lies in the osteoclasts. Effective drug treatments are lacking and there is an urgent need to explore new drugs. MATERIALS AND METHODS: We observed the effects of pogostone (PO) on osteoclas

Erythropoietin Promotes Glioblastoma via miR-451 Suppression.

Erythropoietin (EPO) is an erythropoiesis stimulating growth factor and hormone. EPO has been widely used in the treatment of chronic renal failure, cancer, and chemotherapy-related anemia for three decades. However, many clinical trials showed that EPO treatment may be associated with tumorigenesis and cancer progression. EPO is able to cross blood-brain barriers, and this may lead to an increased possibility of central nervous system tumors such as glioblastoma. Indeed, EPO promotes glioblastoma growth and invasion in animal studies. Additionally, EPO increases glioblastoma cell survival, pr

Treatment with Iodine in Pregnant Rats with Marginal Iodine Deficiency Improves Cell Migration in the Developing Brain of the Progeny.

Marginal iodine deficiency is a common health problem in pregnant women. Epidemiological and animal studies had shown that marginally maternal iodine deficiency could cause the mild changes of maternal thyroid function, eventually lead to a negative effect on neurodevelopment. But the underlying mechanisms responsible for the neurological impairment remain unclear. The aim of this study is to explore whether marginally maternal iodine deficiency could produce subtle changes in cell migration and cognitive function of offspring, and the optimal time of giving intervention to minimize the advers

Control of cell migration and inflammatory mediators production by CORM-2 in osteoarthritic synoviocytes.

BACKGROUND: Osteoarthritis (OA) is the most widespread degenerative joint disease. Inflamed synovial cells contribute to the release of inflammatory and catabolic mediators during OA leading to destruction of articular tissues. We have shown previously that CO-releasing molecules exert anti-inflammatory effects in animal models and OA chondrocytes. We have studied the ability of CORM-2 to modify the migration of human OA synoviocytes and the production of chemokines and other mediators sustaining inflammatory and catabolic processes in the OA joint. METHODOLOGY/PRINCIPAL FINDINGS: OA synovioc

Long-term assessment of enriched housing and subventricular zone derived cell transplantation after focal ischemia in rats.

The potential for using stem cells to treat stroke has garnered much interest, but stem cell therapies must be rigorously tested in animal models before transplantation studies progress to clinical trials. An enriched environment enhances transplanted subventricular zone (SVZ) cell migration and functional benefit following stroke in rats. However, the ability of SVZ cells to survive, migrate, differentiate and promote functional recovery at protracted survival times (e.g., 3 months) has not been investigated. The vasoconstrictive peptide endothelin-1 was injected adjacent to the middle cerebr
