The dopaminergic projection system, basal forebrain macrosystems, and conditioned stimuli.

This review begins with a description of some problems that recently have beset an influential circuit model of fear conditioning and goes on to look at neuroanatomy that may subserve conditioning viewed in a broader perspective, including not only fear but also appetitive conditioning. The column will then focus on basal forebrain functional-anatomical systems, or macrosystems, as they have come to be called. Yet, more specific attention is then given to the relationships of the dorsal and ventral striatopallidal systems and extended amygdala with the dopaminergic mesotelencephalic projection

Serotonin neurons in the dorsal raphe control food-craving-like behavior during pregnancy in mice.

During pregnancy, physiological changes can alter food intake behaviors. However, the underlying neural mechanisms remain unclear. Here we show that female mice exhibit food-craving-like behavior during pregnancy, mirroring many patterns described in humans. We show that 5-HTDRN neuronal firing activity is reduced during pregnancy through increased small-conductance calcium-activated potassium type 3 (SK3) ion-channel activity. Genetic knockout of SK3 ion channels from 5-HTDRN neurons abolishes the pregnancy-associated suppression in 5-HTDRN neuronal firing activity and reduces food-craving-li

Changes in food intake and abnormal behavior using a puzzle feeder in newly acquired sub-adult rhesus monkeys (Macaca mulatta): a short term study.

The majority of newly acquired nonhuman primates encounter serious problems adapting themselves to new environments or facilities. In particular, loss of appetite and abnormal behavior can occur in response to environmental stresses. These adaptation abnormalities can ultimately have an affect on the animal's growth and well-being. In this study, we evaluated the affects of a puzzle feeder on the food intake and abnormal behavior of newly acquired rhesus monkeys for a short period. The puzzle feeder was applied to 47- to 58-month-old animals that had never previously encountered one. We found

The essence of appetite: does olfactory receptor variation play a role?

Olfactory receptors are G-protein-coupled chemoreceptors expressed on millions of olfactory sensory neurons within the nasal cavity. These receptors detect environmental odorants and signal the brain regarding the location of feed, potential mates, and the presence of possible threats (e.g., predators or chemical toxins). Olfactory receptors also are present in organs outside of the nasal cavity where they bind to molecules such as nutrients and metabolites from the animal's internal environment to elicit physiological responses, including changes in gut motility, ventilation rate, and cellula

Evidence that ghrelin may be associated with the food intake of gibel carp (Carassius auratus gibelio).

Ghrelin, a non-amidated peptide hormone, is a potent anorectic neuropeptide implicated in feeding regulation in mammals and non-mammalian vertebrates. However, the involvement of ghrelin in the feeding behavior of teleosts has not been well understood. To better understand the role of ghrelin in the regulation of appetite in fish, in this study, we cloned the cDNAs encoding ghrelin and investigated their mRNA distributions in gibel carp tissues. We also assessed the effects of different nutritional status on ghrelin mRNA abundance. Ghrelin mRNAs were ubiquitously expressed in ten tissues (inte

Weight gain with antidepressants and lithium.

Undesired weight gain is a common complaint of patients receiving pharmacological treatment for major affective disorders. It has been found to jeopardize patient compliance and may pose additional health hazards. A review of the literature on weight gain associated with tricyclic antidepressants, monoamine oxidase inhibitors, and lithium was carried out with the aim of deriving practical management strategies. Tricyclic antidepressants were found to stimulate appetite, carbohydrate craving, and a dose-dependent continuous weight gain of 0.57 to 1.37 kg per month of treatment. Proposed mechani

Attraction of Culex pipiens pallens (Diptera: Culicidae) to Floret Volatiles and Synthetic Blends of Its Nectar Host Plant Abelia chinensis (Rubiales: Caprifoliaceae).

Abelia chinensis R. Br. (Dipsacales: Caprifoliaceae) is one of the preferred nectar host plants for Culex pipiens pallens Coquillett (Diptera: Culicidae). However, the volatile compounds of its flowers that might be involved in directing mosquitoes' orientation to its nectaries remain unknown. In the present study, the volatile compounds released by A. chinensis florets were collected by solid phase microextraction fiber and analyzed by gas chromatography-mass spectrometry system. Based on the major component species in the volatile profile, a synthetic phytochemical blend (Blend B, composed o

The global obesity epidemic: snacking and obesity may start with free meals during infant feeding.

UNLABELLED: Feeding is vital for survival. The brain has strong hunger and reward mechanisms that ensure optimal food intake for adequate nutrition. The drive for feeding is particularly strong in humans whose large brains require large energy support. This starts immediately after birth; the newborn child being able to taste sucrose and suck the sweet and fat from its mother's milk. At present, mothers are generally advised to breastfeed children as often as they like, which may be up to 15 times a day. At the same time, childhood obesity is rapidly developing. One reason for the rapidly incr

Effect of passive zooprophylaxis on malaria transmission in The Gambia.

The effect of zooprophylaxis on malaria transmission has not been studied on the African continent despite that the World Health Organization has recommended this intervention method since 1982. The effect of passive zooprophylaxis on malaria vector abundance, mosquito feeding preferences, and infectivity was studied in an area of moderate seasonal transmission in The Gambia. A paired cohort of 204 children <7 yr of age was selected and matched in groups for presence or absence of cattle (Bos taurus) within 20 m of their bedroom. Comparisons were made between mosquitoes collected from the bedr

(-)-Norpseudoephedrine, but not (+/-)-amphetamine, prevents the increase in fluid intake associated with ethanol presentation in rats.

The effects of daily intraperitoneal administration of (+/-)-amphetamine (AMPH: 4 mg/kg) and (-)-norpseudoephedrine (NPE: 15 or 30 mg/kg) on drinking were evaluated in rats allowed to drink water or a 5% ethanol solution during the 5 h following drug administration. AMPH, but not NPE, significantly increased water intake. Substitution of ethanol for water produced a significant increase in fluid intake in the control group, whereas drinking levels remained unchanged in both AMPH and NPE treated rats. Discontinuation of drug treatment during the regimen of ethanol presentation produced a prompt

The use of a plus-maze to measure anxiety in the mouse.

To investigate whether an elevated plus-maze consisting of two open and two closed arms could be used as a model of anxiety in the mouse, NIH Swiss mice were tested in the apparatus immediately after a holeboard test. Factor analysis of data from undrugged animals tested in the holeboard and plus-maze yielded three orthogonal factors interpreted as assessing anxiety, directed exploration and locomotion. Anxiolytic drugs (chlordiazepoxide, sodium pentobarbital and ethanol) increased the proportion of time spent on the open arms, and anxiogenic drugs (FG 7142, caffeine and picrotoxin) reduced th

The effect of selective lesioning of brain catecholamine-containing neurons on the activity of various anorectics in thr rat.

An intraventricular injection of 6-hydroxydopamine to rats pretreated with pargyline, a procedure which produces a marked decrease of brain catecholamines without significant changes in the serotonin levels, significantly antagonizes the anorectic effect of amphetamine, phentermine, mazindol and diethylpropion, while the reduction of food intake induced by other drugs such as fenfluramine, p-chloroamphetamine, SE 780 and SKF 1-39728 is not significantly affected. The data suggest an involvement of brain catecholamines in the anorectic effect of amphetamine, phentermine, mazindol and diethylpro

Social living mitigates the costs of a chronic illness in a cooperative carnivore.

Infection risk is assumed to increase with social group size, and thus be a cost of group living. We assess infection risk and costs with respect to group size using data from an epidemic of sarcoptic mange (Sarcoptes scabiei) among grey wolves (Canis lupus). We demonstrate that group size does not predict infection risk and that individual costs of infection, in terms of reduced survival, can be entirely offset by having sufficient numbers of pack-mates. Infected individuals experience increased mortality hazards with increasing proportions of infected pack-mates, but healthy individuals rema

Dorsal raphe nucleus enkephalin peptide modulates behavioral preference.

The endogenous opioid system is a powerful modulator of motivation and affect. The dorsal raphe nucleus (DRN) in the midbrain has been established as an important site of opioid action and is an integral hub in behavioral modulation. To investigate the functional significance of DRN opioid signaling in aversive and appetitive behaviors we disrupted preproenkephalin (Penk) in DRN using CRISPR-Cas9 technology in Penk-Cre mice. We found that CRISPR mediated knockdown of enkephalin peptide in the DRN (DRNPenk) enhanced inflammation-induced mechanical sensitivity and odor avoidance. Additionally, l…

Considerations for Dietary Supplements after GLP-1RA Treatment: A Narrative Review.

Glucagon-like peptide-1 receptor agonist (GLP-1RA) prescriptions have significantly increased. The durability of these weight reduction drugs depends on continued treatment, and nearly half of patients discontinue GLP-1RA within the first year. Existing weight maintenance guidelines do not address challenges that arise after GLP-1RA. Furthermore, limited to no guidance is provided on the potential adjunct role of dietary supplements that may help during the transition. This narrative review aims to evaluate considerations for dietary supplements for patients discontinuing GLP-1RA. A review of

Epigenetics-Driven Appetite System Disorders and PI3K/AKT Signaling Activation Mediate a Weight Gain Resistance Phenotype Induced by a High-Fat Diet in Male Rats.

OBJECTIVE: Some patients with anorexia nervosa (AN) exhibit an aversion to high-fat food. The hypothalamus plays a crucial role in feeding behavior. This study investigated the role of epigenetic modifications in the hypothalamus of male rats exhibiting a weight gain resistance (WR) phenotype induced by a high-fat diet (HFD). METHOD: Male Sprague-Dawley (SD) rats were divided into normal diet (ND) and HFD groups for 9 weeks. Rats in the HFD group were then divided into HFD-induced obese (HFO) or HFD-induced WR phenotype (HFWR) groups, and body weight and food intake were monitored for 24 days

In vitro gastric emptying characteristics of konjac glucomannan with different viscosity and its effects on appetite regulation.

Konjac glucomannan (KGM) is associated with the satiety-enhancing property by imparting the food matrix with high viscosity. In the present study, rheology tests on KGM sol with different viscosities were conducted to understand its flow behavior as they presented in the mouth and stomach, and the in vitro gastric emptying characteristics of KGM were examined with a human gastric simulator. Then, their effects on subjective appetite, glycemia, and appetite-related hormones (insulin, GLP-1, PYY3-36, CCK-8, ghrelin) response were investigated by conducting a randomized, single-blind, crossover t

Cocaine Blocks Effects of Hunger Hormone, Ghrelin, Via Interaction with Neuronal Sigma-1 Receptors.

Despite ancient knowledge on cocaine appetite-suppressant action, the molecular basis of such fact remains unknown. Addiction/eating disorders (e.g., binge eating, anorexia, bulimia) share a central control involving reward circuits. However, we here show that the sigma-1 receptor (σ1R) mediates cocaine anorectic effects by interacting in neurons with growth/hormone/secretagogue (ghrelin) receptors. Cocaine increases colocalization of σ1R and GHS-R1a at the cell surface. Moreover, in transfected HEK-293T and neuroblastoma SH-SY5Y cells, and in primary neuronal cultures, pretreatment with cocai
