Smartphone-based lightweight AI system for real-time multiple anterior segment disease screening: development and real-world validation.

BACKGROUND: Anterior segment diseases are a major global cause of preventable blindness, especially in regions with limited access to specialized ophthalmic care. Diagnosis typically requires slit-lamp biomicroscopy, creating a significant access bottleneck in primary care and rural settings. Existing AI solutions often lack the efficiency and generalizability necessary for widespread mobile deployment and primarily focus on individual conditions, failing to meet the demand for scalable, multi-disease screening platforms. METHODS: We developed the Intelligent Detection System (IDS), a smartph

Lewis Acid-Catalyzed Strain-Enabled (3+3) Annulation of Bicyclo[1.1.0]Butanes With Indolylalcohols: Synthesis of Indole-Fused Aza-Bicyclo[3.1.1]Heptanes.

Strain release has long inspired the discovery of unique reactivity, enabling diverse synthetically valuable transformations. Bicyclo[1.1.0]butanes (BCBs), due to their high ring strain, have emerged as versatile building blocks for constructing complex bicyclic architectures. Herein, we report a Lewis acid-catalyzed formal (3 + 3) annulation of BCBs with indolyl alcohols, providing rapid access to indole-fused aza-bicyclo[3.1.1]heptanes. The reaction proceeds under mild conditions, tolerates a broad range of functional groups, and delivers rigid, sp3-rich scaffolds efficiently. Mechanistic st

Embracing macrophage complexity for research and translation.

Macrophages are among the most versatile differentiated cell types in the body, serving as key regulators of tissue development, homeostasis, inflammation, repair, and immunity. Their functional diversity cannot be captured by fixed subsets, binary activation states, or single-marker definitions. Macrophage identity emerges instead from the integration of developmental origin, tissue niche, epigenetic history, local environmental cues, and disease-associated signals. Advances in single-cell genomics, spatial profiling, epigenomics, in vivo perturbation, and computational modeling now show that

Triplex-forming oligonucleotides as programmable recognition interfaces for biosensing and translational diagnostics.

Triplex-forming oligonucleotides (TFOs) are programmable nucleic acid ligands that recognize duplex DNA through Hoogsteen or reverse Hoogsteen interactions, providing a unique molecular recognition strategy beyond conventional Watson-Crick hybridization. Recent advances in nucleic acid engineering, chemical modification, nanotechnology, and signal amplification have transformed TFOs from classical antigene agents into versatile platforms for biosensing and bioanalysis. In this review, we highlight the chemical principles governing triplex formation and discuss recent developments in TFO-enable

Nanochemical modulation of ECM-driven pseudo-resistance: convergent microenvironmental pathways in COVID-19 and cancer.

Extracellular matrix (ECM) remodeling is increasingly recognized as a central determinant of inflammation, fibrosis, and therapeutic response across chronic diseases. This Perspective examines how post-COVID-19 pulmonary fibrosis and stromal-driven resistance in solid tumors converge on a shared phenomenon, ECM-driven pseudo-resistance, an extrinsic and reversible microenvironmental state in which pathological matrix architecture, fibro-inflammatory signaling, and immune exclusion transiently limit therapeutic efficacy without conferring stable, mutation-driven cellular resistance. We highligh

High-Q microresonators unveil quantum rare events.

Classical linear optics posits that at sufficiently low intensities, light propagation in dielectric media is governed solely by their linear susceptibilities. Here, we demonstrate a departure from this paradigm in high-quality (high-Q) microresonators, where prolonged photon confinement enables rare quantum electrodynamical (QED) events, mediated by the quantum vacuum, to embed distinctive Raman signatures of the coupled analyte into the resonator's linear transmission spectrum despite their absence from linear susceptibility. We further show that increasing the amount of adsorbed analyte sam

Pyridoxal 5'-Phosphate-Dependent Enzymatic Decarboxylative Annulation.

Pyridoxal 5'-phosphate (PLP)-dependent enzymes are among the most versatile biocatalysts, yet transformations involving the Cγ-nucleophilic vinylglycine quinonoid (VGQ) intermediate remain exceptionally rare. Understanding the untapped reactivity of VGQ could open new avenues for developing PLP-dependent biocatalysts. Here, we establish a biocatalytic platform that artificially accesses and exploits the reactivity of this high-energy intermediate. By reprogramming SphA, a PLP-dependent enzyme that natively catalyzes decarboxylative Claisen condensation, to generate VGQ in situ through facile d

Maintenance of nucleosome organization through replication and transcription counteracts aberrant coalescence of active chromatin.

Nucleosomes with their associated modifications organize and regulate the genome. It is unclear how this is integrated with the requirement of replication and transcription to access the DNA template without jeopardizing chromatin function. Here, we reveal a unified requirement for the histone chaperone FACT in mediating nucleosome disruption and reassembly during mammalian replication and transcription. Upon acute FACT depletion, replisome and RNA polymerase progression is halted genome wide, and chromatin structure in their wake collapses, with reduced nucleosome occupancy, irregular spacing

Diazines-to-Diazoles Skeletal Remodeling.

Nitrogen-containing heteroarenes, particularly diazines (six-membered dinitrogen aromatics) and diazoles (five-membered dinitrogen aromatics), constitute the bedrock of modern pharmaceutical and agrochemical architectures. Synthetic capability to edit these heteroaromatic scaffolds through precise core-atom transformations opens the door to previously untapped chemical spaces, thus accelerating discovery campaigns for developing pharmaceutical drugs and agrochemicals. Here we report a photocatalytic platform for the skeletal remodeling of diazines into diazoles, encompassing three distinct per

Where metabolism and virulence meet-sulfoxide reductases as determinants of bacterial survival.

Bacterial metabolism is increasingly recognised as a driver of virulence in pathogens, and this includes the reduction of sulfoxides, which has traditionally been linked to anaerobic energy generation and maintenance of cellular proteins. Sulfoxide reduction is catalysed by two major enzyme groups, the Msr peptide methionine sulfoxide reductases that repair damaged proteins, and several types of mononuclear molybdenum enzymes that either convert small molecular S- and N-oxides or repair damaged proteins. Where these enzymes support virulence, they appear to be regulated by exposure to oxidisin

Exploring anatomical similarity in zero-shot learning for bone abnormality detection.

Deep learning models for musculoskeletal imaging often require large, annotated datasets per anatomical region. In this study we explicitly treat abnormality detection (normal vs. abnormal) on MURA (which does not provide fracture-only labels) and ask whether anatomical similarity enables zero-shot (out-of-domain) transfer-training on one body part and evaluating on unseen parts without target data access. Using study-level aggregation and Wilson 95% confidence intervals, we quantify cross-part transfer as a function of anatomical proximity. We also include a small, labeled replication subset

Structural Polymorphism of polyG Inclusions Revealed by In Situ Cryo-Electron Tomography.

NIID (Neuronal intranuclear inclusion disease) is defined by ubiquitin- and p62-positive intranuclear inclusions, yet their native ultrastructure remains unclear. Using correlative cryo-electron tomography in primary cortical neurons and brain tissue from an NIID mouse model, we show that polyG inclusions are built from interconnected ribbon-like assemblies rather than canonical amyloid fibrils. PolyG populates multiple compartment-specific ribbon states, including a nuclear ribbon network enriched in 26S proteasomes and two cytoplasmic ribbon packing states with sharply different proteasome a

Synthetic Studies toward the Anacolosin Family of Clerodane Diterpene Natural Products.

The anacolosins are an underexplored family of clerodane diterpenes with intriguing bioactivity and unique structural complexity. These molecules present a dense architectural challenge, featuring seven stereogenic centers and a distinctive five-membered bis-acetal ring system within their tricyclic core. We describe here the successful development of a scalable synthesis that completes two of three intended phases toward the total synthesis of the anacolosins. Obstacles that were overcome include the navigation of the steric demands of a Diels-Alder cycloaddition and exploiting inherent torsi

Selenium atom transfer enables selective bridged alkene-arylamine cycloaddition.

Cycloaddition reactions are central to building cyclic molecular architectures for drug discovery and materials science. However, controlling single-atom-bridged cycloaddition remains elusive. Herein, we describe a selenium-atom transfer [3 + 2 + 1] heterocycloaddition of readily available arylamines, alkenes and diselenides, enabling rapid, modular access to diverse selenium- and nitrogen-containing heterocycles, including selenomorpholines and polycyclic 1,4-selenazinanes. This metal-free method operates under mild, simple conditions with scalability, showcasing broad substrate generality an

Recapitulating gestational progression: a high-throughput 3D bioprinted vascularized human placenta demonstrating stage-specific molecular and functional signatures of human pregnancy.

The placenta is a highly dynamic organ that supports the growth and development of the fetus during gestation. Malfunction of the placenta causes disorders of the pregnancy, including preeclampsia and pre-term birth. The study of the pathophysiology of the placenta throughout pregnancy has been hindered by limited access to the human placenta and lack of predictive cellular and animal models. Here, we describe 3D bioprinted human placenta barrier (hPB) tissue models using primary human trophoblasts and stroma cells that recapitulate the physiology of the human placenta at early and late stages

A DC-sensitive video/electrophysiology monitoring unit for long-term continuous study of seizures and seizure-associated spreading depolarization in a rat model.

There has been a long-term need for a low-cost, highly efficient, and high-fidelity epilepsy monitoring unit (EMU) suitable for home-cage monitoring of small-animal models of epilepsy. We show an accessible, scalable, highly space and energy-efficient EMU capable of fulfilling chronic, continuous synchronized multiple animal monitoring jobs. Each rig within the EMU can provide 16-channel high-fidelity, DC-sensitive biopotential recordings, head acceleration monitoring, synchronized voltammetry applications, and video recording on one freely moving rat. We present the overall EMU architecture d

Electrical pulse generator for electroporation induction in myocytes: Compared effects on skeletal and cardiac cells.

High-intensity, external electric fields (HIEF) have been used in research and therapy for abnormal generation/propagation of the cardiac electrical activity (e.g., defibrillation), and for promoting access of membrane-impermeant molecules into the cytosol through electropores. This report proposes the architecture of a pulse generator designed for both pacing and electroporation induction in isolated myocytes for the study of HIEF impact and membrane repair mechanisms. The instrument was used for quantitative characterization and comparison of the sensitivity of myocytes isolated from skeleta

Exon architecture controls mRNA m6A suppression and gene expression.

N6-methyladenosine (m6A) is the most abundant messenger RNA (mRNA) modification and plays crucial roles in diverse physiological processes. Using a massively parallel assay for m6A (MPm6A), we discover that m6A specificity is globally regulated by suppressors that prevent m6A deposition in unmethylated transcriptome regions. We identify exon junction complexes (EJCs) as m6A suppressors that protect exon junction-proximal RNA within coding sequences from methylation and regulate mRNA stability through m6A suppression. EJC suppression of m6A underlies multiple global characteristics of mRNA m6A
