Fas acts as a T cell co-stimulatory receptor via both death domain-independent and -dependent mechanisms.

T cell responses are regulated by co-stimulatory and inhibitory signalling pathways, driven by receptors and their ligands designated as immune checkpoints. These receptors belong either to the immunoglobulin superfamily or the tumour necrosis factor receptor (TNFR) superfamily. Despite their crucial role in enhancing T effector cell activity, the molecular mechanisms triggered by TNFR co-stimulatory receptors, particularly their integration into the T cell receptor (TCR) signalling network, remain incompletely characterised. The receptor Fas is classically recognised as the prototypical TNFR

Differential contribution of lipoxygenase isoforms to the sequential biosynthesis of specialized pro-resolving mediators in monocellular and cell co-incubations.

Oxylipins, produced from polyunsaturated fatty acids (PUFA) by lipoxygenases (LOX), play pivotal roles in inflammation. While 5-LOX-derived leukotrienes promote inflammation, the specialized pro-resolving mediators (SPMs) contribute to inflammation resolution. The biosynthesis of the SPM members lipoxins and resolvins (RVs) supposedly relies on two sequential PUFA oxygenation steps, involving metabolism by 5-LOX and 12-/15-lipoxygenating paralogues (e.g., 12-LOX, 15-LOX-1/-2). These oxygenations may occur within one cell where both LOX isoforms are expressed or via transcellular metabolism bet

Sublethal DNA damage switches off B cell effector programs in an RA-FLS-PBMC co-culture.

Rheumatoid arthritis (RA) features lymphocyte-driven inflammation, in which B cells, alongside T cells, play key effector roles (e.g., autoantibody production, antigen presentation, cytokine or chemokine production). Within activated B cells, during normal diversification, activation-induced cytidine deaminase (AID) introduces targeted DNA lesions in immunoglobulin loci (class-switch recombination/somatic hypermutation), creating a potential vulnerability to sublethal genotoxic stress. T cells also contribute to RA pathogenesis through cytokine production and cell-mediated responses, and are e

Identification of human transferrin receptor as an entry co-receptor for parvovirus B19 infection of human erythroid progenitor cells.

Parvovirus B19 (B19V), a member of the genus Erythroparvovirus within the Parvoviridae family, infects human erythroid progenitor cells (EPCs) of bone marrow and fetal liver and causes various hematological disorders. The minor capsid protein VP1 of B19V contains a unique N-terminal region (VP1u) that facilitates virus binding and internalization into EPCs via its receptor-binding domain (RBD). We previously identified tyrosine-protein kinase receptor UFO (AXL) as a proteinaceous receptor for B19V infection of EPCs. In this study, we employed an ascorbate peroxidase 2 (APEX2)-based proximity l

Inducible T-Cell Co-Stimulator (ICOS) and ICOS Ligand: Dealing With a Two-Faced Cancer Immunoregulatory System.

BACKGROUND: ICOS (inducible T-cell co-stimulator) and ICOS ligand (ICOSL) are part of an important, complex pathway that can lead to both immune stimulation and suppression. ICOS and ICOSL have heterogeneous expression patterns between and within tumor types. METHODS: This review provides an overview of ICOS and ICOSL, their mechanisms of action, expression in cancer and other diseases, and clinical trials exploring therapies targeting ICOS. RESULTS: Because of the bidirectional immune impact of the ICOS/ICOSL signaling pathway, both ICOS agonists and antagonists are under development and ev

Mitochondria-Targeted Nanozyme Reprograms Dendritic-Cell Immunometabolism via Microenvironment-Responsive CO Release to Treat Periodontitis.

Inflammatory diseases are characterized by overactivated immune responses and a disrupted metabolic equilibrium, particularly in dendritic cells (DCs), where mitochondrial reactive oxygen species (mtROS) burst and metabolic reprogramming drives pathological maturation. While modulating immunometabolism is a promising therapeutic avenue, achieving subcellular-targeted delivery of bioactive molecules remains a formidable challenge. Here, we report a mitochondria-targeted nanozyme designed to concurrently scavenge mtROS and reprogram DC metabolism for effective anti-inflammatory therapy in period

Development and validation of a long-term co-maturation protocol for human stem cell-derived microglia and neuronal networks.

Microglia-neuron interactions play a key role in a variety of central nervous system disorders. Technologies using human induced pluripotent stem cells (hiPSCs) have been developed to model human brain cells with the goal to understand their function. To effectively study neuro-immune crosstalk and investigate microglial contributions to neuronal network development and function, both microglia and neurons should co-mature allowing for long-term interactions throughout their differentiation. Here, we present a co-maturation protocol that robustly generates glutamatergic neuronal networks conta

Establishing a Microfluidic Co-Culture Blood-Brain Barrier Model Using Human Brain Endothelial Cells and Astrocytes.

The blood-brain barrier (BBB) is a highly selective physiological interface that regulates molecular transport between the bloodstream and the central nervous system. In vitro BBB models are valuable tools for studying endothelial barrier formation, cell-cell interactions, and responses to dynamic flow conditions. This protocol describes the establishment of a three-dimensional microfluidic co-culture BBB model using human brain endothelial cells and primary human astrocytes cultured in separate but interconnected compartments of a microfluidic chip. The protocol includes chip preparation, ext

A Gelatin-Oxidized Dextran Hydrogel for Co-Delivery of γδT Cells and Immunostimulatory Agents Potentiates Local Cancer Immunotherapy.

Current αβT cell-based immunotherapies largely relying on the use of autologous T cells not only face limitations such as complex manufacturing processes, long production cycles, and high costs, but are also susceptible to immune escape due to tumor antigen loss or downregulation. In contrast, γδT cells offer a unique "off-the-shelf" advantage owing to their HLA-independent recognition of tumor cells, enabling them to target and eliminate malignancies through multiple mechanisms without the need for patient-specific matching. To address the suppressive tumor microenvironment that often impedes

Regulation of IL-10 production in dendritic cells is controlled by the co-activation of TLR2 and Mincle by Lactiplantibacillus plantarum OLL2712.

We showed that Lactiplantibacillus plantarum OLL2712 (OLL2712) strongly induces interleukin (IL)-10 production in immune cells. Although beneficial effects of this strain have been observed in both mice and humans, the mechanisms underlying IL-10 induction remain unclear. In this study, we found that OLL2712 co-activates two pattern recognition receptors, leading to IL-10 production in the mouse-derived thermosensitive dendritic cell line, tsDC. We first revealed the involvement of the Toll-like receptor (TLR)2-Myeloid differentiation primary response gene (MYD) 88 pathway in OLL2712-induced I

An Injectable Hydrogel Co-Delivering Propranolol and Imiquimod to Synergistically Enhance Immunogenic Cell Death after Melanoma Resection.

Melanoma remains one of the most aggressive skin cancers with high recurrence rates following surgical resection. To overcome the challenges of residual tumor cells and postoperative immune resistance, an injectable hydrogel is developed that co-delivers propranolol (Pro) and imiquimod (IMQ). The rationale is to achieve a synergistic therapeutic effect by enhancing immunogenic cell death (ICD) and strengthening antitumor immunity. The hydrogel is synthesized via boronate ester crosslinking between HA-EGCG and CMCS-PBA and demonstrates excellent injectability, self-healing, biodegradability, an

Tumor-Treating Fields Alter Nanomechanical Properties of Pancreatic Ductal Adenocarcinoma Cells Co-Cultured with Extracellular Matrix.

Tumor-Treating Fields (TTFields), a novel therapeutic avenue, is approved for therapy in Glioblastoma multiforme, malignant pleural mesothelioma, and metastatic non-small cell lung cancer (NSCLC). In pancreatic ductal adenocarcinoma (PDAC), several clinical trials are underway to improve outcomes, yet a significant knowledge gap prevails involving the cell-extracellular matrix (ECM) crosstalk. Herein, we hypothesized that treatment with TTFields influence this crosstalk, which is reflected by the dynamic alteration in nanomechanical properties (NMPs) of cells and the ECM in a co-culture system

Efficient cell model for assessing inflammatory responsive genes in Mycobacterium tuberculosis and SARS-CoV-2 co-infection.

Mycobacterium tuberculosis (Mtb) and severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) may induce immunopathology with extensive lung damage in hosts. To elucidate the dynamics of co-infection Mtb and SARS-CoV-2 and its impact on inflammatory mediators' expression, we conducted a study to evaluate A549, lung epithelial cells, as a potential model for hosting both pathogens simultaneously. Cell infection initiated with Mtb H37Rv and following a 24-h incubation period, the cells were then infected with SARS-CoV-2. After a 72 h incubation period, a precision test was conducted for both

Evaluation of the effect of two adjuvants co-administered with nasal recombinant hemagglutinin influenza vaccine on immune cells in mice.

OBJECTIVES: Vaccination is the most potent and cost-effective way to combat the threat of influenza outbreaks. Strategies to enhance influenza vaccine immunogenicity include the use of high antigen dose vaccines and the inclusion of an appropriate adjuvant. The benefits of adjuvants include enhanced immunogenicity, antigen-sparing, and greater duration of protection. However, adjuvants can increase vaccine reactogenicity and may adversely impact vaccine safety; hence, both risks and benefits need to be carefullyconsidered when adding adjuvants to a vaccine. METHODS: In this study, we examined

Transition from preclinical to clinical application of CTLA4-Ig co-stimulation blockage in beta-cell replacement therapy.

Beta-cell replacement therapies, including islet and pancreas transplantation, offer promising results in term of glycemic control for patients with type 1 diabetes experiencing high glycemic variability and severe hypoglycemia. However, long-term insulin independence remains challenging due to progressive graft function decline. Immunosuppressive regimens, especially calcineurin inhibitors such as tacrolimus, are known to be diabetogenic, contributing to the paradox of impaired beta-cell function in a diabetes treatment setting. Recent studies have focused on CTLA4-Ig (e.g., belatacept) as a

Co-Cultivation With New Glucose-Sparing Chlorella Algae Boosts Tissue Culture Efficiency by Reducing Cell Waste.

Mammalian cell culture technologies are crucial for recombinant protein production, organoid generation, medical applications, and the generation of in vitro cultivated meat. However, they are limited by high costs, lack of vascular O2-provision, and the resultant inhibition of 3D tissue formation. Effective media and nutrient usage, oxygenation, and waste management are key to improvement. Microalgae utilize organic or inorganic CO2 to produce O2 from light, which complements O2-consuming and CO2-respiring mammalian cells in culture. However, common microalgal cultivation conditions differ in

Patient-derived organoid co-culture systems as next-generation models for bladder cancer stem cell research.

Three-dimensional patient-derived organoids (PDOs) have emerged as a powerful model for investigating the molecular and cellular mechanisms underlying bladder cancer, particularly in the context of cancer stem cells (CSCs) and drug screening. However, a significant limitation of conventional PDOs is the absence of tumor microenvironment (TME), which includes critical stromal, immune and microbial components that influence tumor behavior and treatment response. In this review, we provide a comprehensive overview of the recent advancements in PDO co-culture systems designed to integrate TME elem

Charge transfer dynamics and tuning of reorganization energy in graphene-encapsulated co-based alloy catalysts for fuel cells.

Graphene-encapsulated metal alloy nanocatalysts with a metal@graphene type architecture are unique catalysts with multifunctional properties. However, effectively integrating these physicochemical properties to enhance electrocatalytic performance still presents a significant challenge. Here, using density functional theory (DFT) calculations, we first reveal that tuning the composition of CoxPdy (x ≫ y) alloys optimizes structural reorganization energy and enhances charge transfer from the metal core to the graphene shell, thereby improving oxygen reduction reaction (ORR) activity. Environmental chromium and cadmium co-exposure induces GADD45A-mediated rDNA instability in human cohorts and cell models.

Exposure to heavy metals such as hexavalent chromium [Cr(VI)] and cadmium (Cd) poses a major environmental and occupational health concern due to their widespread presence and genotoxic potential. While the toxicological profiles of individual metals are well-characterized, their combined effects on genome stability remain poorly understood. Ribosomal DNA (rDNA), a repetitive and transcriptionally active genomic region essential for protein synthesis, is increasingly recognized as a sensitive target of environmental stress and genomic instability. Here, we investigated electroplating workers s
