Restoration of oxidative stress by HNF4α deletion in VWA8-null hepatocytes.

VWA8 is a mitochondrial protein expressed at high levels in mitochondria-enriched organs such as the liver, kidney, and heart. Deletion of VWA8 (VWA8-KO) in AML12 mouse hepatocytes leads to oxidative stress characterized by elevated production of reactive oxygen species (ROS) and, subsequently, higher oxidative capacity. Concurrently, there is a substantial rise in the expression of the transcription factor HNF4α in VWA8-KO hepatocytes. Thus, exploring the potential significance of HNF4α in the observed effects warrants investigation. In this study, we deleted the HNF4α gene using CRISPR/Cas9

Arterial chemoreceptor input to nucleus tractus solitarius.

The arterial chemoreceptors play an important role in the reflex regulation of blood pressure and respiration. To investigate the initial integration of chemoreceptor inputs within the central nervous system, intracellular recordings were obtained in pentobarbital-anesthetized, paralyzed, and mechanically ventilated cats, from 58 cells within the nucleus of the tractus solitarius (NTS) that were depolarized by activation of the ipsilateral carotid body chemoreceptors. Close arterial injection of less than 100 microliters CO2-saturated bicarbonate evoked depolarizations of membrane potential wi

Candida albicans erythroascorbate peroxidase regulates intracellular methylglyoxal and reactive oxygen species independently of D-erythroascorbic acid.

Candida albicans D-erythroascorbate peroxidase (EAPX1), which can catalyze the oxidation of D-erythroascorbic acid (EASC) to water, was observed to be inducible in EAPX1-deficient and EAPX1-overexpressing cells via activity staining. EAPX1-deficient cells have remarkably increased intracellular reactive oxygen species and methylglyoxal independent of the intracellular EASC content. The increased methylglyoxal caused EAPX1-deficient cells to activate catalase-peroxidase and cytochrome c peroxidase, which led to defects in cell growth, viability, mitochondrial respiration, filamentation and viru

Role of transcobalamins I, II, and III in the transfer of vitamin B12 to human bone marrow cells in vitro.

A study of the uptake of transcobalamin-bound 57Co-cyanocobalamin by suspensions of human bone marrow cells has indicated that these cells can take up vitamin B12 from all 3 transcobalamins (I, II, and III). Similar transport processes were involved in the uptake from the 3 transcobalamins; uptake was dependent on the presence of calcium ions, cellular respiration and free sulphydryl groups. These results suggest that contrary to current belief, all 3 transcobalamins play a role in the transfer of vitamin B12 to tissue cells.

Oxygen-dependent redox control enables growth-decoupled biotransformations in Cupriavidus necator.

Genetic instability and environmental heterogeneity present persistent challenges to the stable microbial production of reduced, value-added chemicals. This work shows that limiting oxygen availability redirects electrons from respiration toward targeted reductive pathways in Cupriavidus necator, steering growth-decoupled biotransformations - exemplified by the conversion of acetoin to 2,3-butanediol and of glycerol to 1,3-propanediol. In cell suspension assays under a hydrogen-containing atmosphere, anoxic conditions stabilised near-stoichiometric acetoin-to-2,3-butanediol conversion (93% mol

Human Mesenchymal Stromal Cells Attenuate Hyperoxia-Induced Cellular Impairment of Immature Oligodendrocyte and Neurons.

Preterm infants are at high risk of developing long-term brain injury such as encephalopathy of prematurity (EoP). Hyperoxia is a major contributor to EoP, affecting white and grey matter, with immature oligodendrocytes and hippocampal neurons being particularly vulnerable. While no causal therapy is available, mesenchymal stromal cells (MSCs) show therapeutic potential and are considered a promising candidate, although their effector mechanisms remain incompletely understood. Primary oligodendrocytes were isolated from mixed glial cultures of P0-P2 rats and hippocampal neurons from E16 rat em

Evaluating metabolic changes in H9c2 cardiomyoblasts exposed to LPS: Towards understanding sepsis mechanisms.

Sepsis is a major public health concern associated with high mortality rates, particularly due to sepsis-induced myocardial dysfunction (SIMD), which affects about 50% of septic patients. This study investigates how mitochondrial dysfunction contributes to SIMD by examining metabolic changes in H9c2 cardiomyoblasts exposed to varying concentrations of lipopolysaccharide (LPS), a bacterial endotoxin, to enhance our understanding of the relationship between infection severity and metabolic responses. H9c2 cells were treated with LPS at concentrations of 0.5, 1, 2.5, and 5 µg/mL for 24 or 48 hour

Ginsenosides attenuate bioenergetics and morphology of mitochondria in cultured PC12 cells under the insult of amyloid beta-peptide.

BACKGROUND: Mitochondrial dysfunction is one of the significant reasons for Alzheimer's disease (AD). Ginsenosides, natural molecules extracted from Panax ginseng, have been demonstrated to exert essential neuroprotective functions, which can ascribe to its anti-oxidative effect, enhancing central metabolism and improving mitochondrial function. However, a comprehensive analysis of cellular mitochondrial bioenergetics after ginsenoside treatment under Aβ-oxidative stress is missing. METHODS: The antioxidant activities of ginsenoside Rb1, Rd, Re, Rg1 were compared by measuring the cell surviva

Haploinsufficiency due to a novel ACO2 deletion causes mitochondrial dysfunction in fibroblasts from a patient with dominant optic nerve atrophy.

ACO2 is a mitochondrial protein, which is critically involved in the function of the tricarboxylic acid cycle (TCA), the maintenance of iron homeostasis, oxidative stress defense and the integrity of mitochondrial DNA (mtDNA). Mutations in the ACO2 gene were identified in patients suffering from a broad range of symptoms, including optic nerve atrophy, cortical atrophy, cerebellar atrophy, hypotonia, seizures and intellectual disabilities. In the present study, we identified a heterozygous 51 bp deletion (c.1699_1749del51) in ACO2 in a family with autosomal dominant inherited isolated optic at

The role of the "Thiodendron" consortium in postulating the karyomastigont chimaera of the endosymbiosis theory by Lynn Margulis.

The endosymbiosis theory of the origin of eukaryotic cell was first proposed more than a hundred years ago. In the second half of the 20th century, Lynn Margulis suggested a new interpretation of the origin of the nucleus in modern eukaryotes. The background was the study of the consortium "Thiodendron", a symbiotic bacterial community, which includes anaerobic aerotolerant motile spirochaetes and sulfidogenic bacteria (sulfidogens) of vibrioid form with a fermentation type of metabolism. Spirochaetes supply sulfidogens with metabolites (pyruvate and, probably, organic nitrogenous products of

Ursodeoxycholic Acid Improves Mitochondrial Function and Redistributes Drp1 in Fibroblasts from Patients with Either Sporadic or Familial Alzheimer's Disease.

Alzheimer's disease (AD) is the leading cause of dementia worldwide. Mitochondrial abnormalities have been identified in many cell types in AD, with deficits preceding the development of the classical pathological aggregations. Ursodeoxycholic acid (UDCA), a treatment for primary biliary cirrhosis, improves mitochondrial function in fibroblasts derived from Parkinson's disease patients as well as several animal models of AD and Parkinson's disease. In this paper, we investigated both mitochondrial function and morphology in fibroblasts from patients with both sporadic and familial AD. We show

Treating SCA1 Mice with Water-Soluble Compounds to Non-Specifically Boost Mitochondrial Function.

Mitochondrial dysfunction plays a significant role in the aging process and in neurodegenerative diseases including several hereditary spinocerebellar ataxias and other movement disorders marked by progressive degeneration of the cerebellum. The goal of this protocol is to assess mitochondrial dysfunction in Spinocerebellar ataxia type 1 (SCA1) and assess the efficacy of pharmacological targeting of metabolic respiration via the water-soluble compound succinic acid to slow disease progression. This approach is applicable to other cerebellar diseases and can be adapted to a host of water-solubl

Can periodic breathing have advantages for oxygenation?

A model of the chemoreceptor mediated control of breathing is analysed. Hypoxia is simulated as low equilibrium arterial oxygen both at altitude and at sea level. Conditions for stable and unstable equilibria are examined. Transition to instability leads to periodic breathing and the model predicts that this can cause arterial oxygen to oscillate asymmetrically about the equilibrium in some cases, causing the average to rise. Periodic forcing functions have been applied to cerebral blood flow with the same effect on arterial oxygen. Such asymmetric oscillations leading to enhanced oxygenation
