OBJECTIVE: To determine the prevalence and clonality of non-toxigenic Corynebacterium diphtheriae biovar gravis in a community with two cases of endocarditis caused by this organism.
SETTING: A Koorie (Aboriginal) community in Gippsland, eastern Victoria, in 1994.
METHODS: Nose and throat swabs were collected from 359 community contacts of the cases and cultured for C. diphtheriae. Strains isolated from the contacts were compared by pulsed-field gel electrophoresis (after digestion with Sma1, Not1 and Sfi1) with those from the invasive cases in the same community, another invasive case in Vi…

The susceptibilities of 265 strains of Corynebacterium species and other non-spore-forming gram-positive bacilli to 18 antimicrobial agents were tested. Most strains were susceptible to vancomycin, doxycycline, and fusidic acid. Corynebacterium jeikeium and Corynebacterium urealyticum were the most resistant organisms tested. Resistance to beta-lactams, clindamycin, erythromycin, azythromycin, ciprofloxacin and gentamicin was common among strains of Corynebacterium xerosis and Corynebacterium minutissimum. Ampicillin resistance among Listeria monocytogenes was more prevalent than previously re…

Corynebacterium striatum is a gram-positive curved bacilli included in "diphtheroids". It is now being reported as a significant multidrug resistant nosocomial pathogen in immunocompromised patients. We describe a case of Catheter-related bloodstream infection caused by Corynebacterium striatum in an 86-year-old patient suffering from Acute Myeloid leukaemia. Rapid MALDI-TOF MS based identification of paired blood culture isolates confirmed true CRBSI. Prompt catheter removal and vancomycin therapy led to recovery. This case underscores the significance of MDR Corynebacterium striatum in oncol…

Corynebacterium striatum, which inhabits the human skin and mucous membranes, is generally considered to have low pathogenic potential. It rarely causes infectious endocarditis in immunocompetent hosts without underlying cardiac disease. We report a case of infectious endocarditis caused by multidrug-resistant Corynebacterium striatum in a 75-year-old man with no history of cardiac disease or other predisposing conditions following endoscopic mucosal resection of the rectum. When C. striatum is isolated from blood cultures, careful evaluation of potential underlying risk factors is essential t…

γ-Hydroxybutyrate (GHB) is an important C4 platform chemical, serving as a crucial precursor for the synthesis of various bulk chemicals, including γ-butyrolactone (GBL) and 1,4-butanediol (1,4-BDO). In this study, we report the systematic metabolic engineering of Corynebacterium glutamicum for the biological production of GHB from glucose via the introduction of a glutamate-derived pathway. We showed that C. glutamicum is a promising host for producing GHB due to its higher tolerance to GHB as compared to other chassis. By screening key enzymes capable of converting glutamate into GHB and blo…

Bacterial keratitis is a serious, potentially sight-threatening complication in neglected cases of corneal trauma. Few bacteria and fungi are implicated in the pathogenesis of bacterial keratitis after trauma; however, keratitis by an opportunistic organism is rare. We report here a case of keratitis caused by Corynebacterium jeikeium (C. jeikeium) after accidental trauma with rice grains. The patient presented to us with chronic keratitis, which responded to 5% fortified vancomycin eye drops after microbiological investigation and drug sensitivity.

We describe a case of chondroblastic osteosarcoma of the vertebral column in a 67-yr-old male in whom the preoperative diagnosis was made by … l as colonies of gram-positive bacilli within the necrotic tumor. Culture confirmed the presence of Corynebacterium species. It is likely that these skin organisms were introduced at the time of FNAB. This case demonstrates the value of FNAB in the diagnosis of primary bone tumors and reports a rare complication of this procedure.

Corynebacterium diphtheriae septicaemia is rarely encountered, usually in very particular situations: children with severe congenital heart disease or after heart surgery. Rare cases have been reported in immunodepressed adults or drug addicts. We observed a case in a formerly healthy 41-year-old woman who was hospitalized for fever unresponsive to bacampicillin. In this patient, no portal of entry could be identified; there was no history of past surgery nor drug abuse. The patient was not immunodepressed and HIV serology was negative. The last anti-diphtheria vaccination had been given at th…

Dramatic reductions in the incidence of diphtheria and high levels of childhood vaccination in recent decades have led the United States to establish the goal of diphtheria elimination among persons < or = 25 years of age by the year 2000. In 1990, an unimmunized 25-month-old child died of respiratory diphtheria in Dade County, Florida, before treatment with diphtheria antitoxin could be instituted. Twenty-three asymptomatic household contacts and other close contacts of the child were identified, cultured for Corynebacterium diphtheriae, given antimicrobial prophylaxis, and vaccinated with di…

Intestinal barrier dysfunction is a core pathological feature of inflammatory bowel disease (IBD). Postbiotics have emerged as promising alternatives to probiotics due to their superior stability and safety. C. glutamicum is an industrial microorganism being Generally Recognized as Safe (GRAS); however, the efficacy and mechanism of its lysate in repairing the intestinal barrier remain largely unexplored. In this study, a dextran sulfate sodium (DSS)-induced colitis mouse model and a lipopolysaccharide (LPS)-induced Caco-2 cell inflammatory barrier model were employed to evaluate the barrier-r

BACKGROUND: Septic arthritis of native joints remains an uncommon but serious clinical condition affecting approximately two per 100,000 individuals annually. The synovial membrane, being highly vascular and lacking a protective basement membrane, is particularly susceptible to hematogenous bacterial seeding, especially in patients with pre-existing joint disease. Corynebacterium propinquum, typically regarded as a respiratory commensal, has rarely been implicated in deep-seated infections.
CASE SUMMARY: We report the first known case of C. propinquum isolated from synovial fluid in a patient…

Corynebacterium ulcerans is a zoonotic pathogen causing cutaneous diphtheria. In Belgium, the bacterium is the cause of hedgehog diphtheric disease (HDD) that appears to have emerged from an endemic state within the European hedgehog (Erinaceus europaeus) population. In spring 2025, 11 hedgehogs admitted to a wildlife rehabilitation center were euthanized due to their severe condition. Blood was collected for anti-diphtheria toxin antibody detection, and lesion swabs (if present) and oral cavity swabs were taken, followed by necropsy. Of nine hedgehogs exhibiting HDD-like lesions, five were co…

To accurately detect internal and environmental cues, bacteria have evolved signal transduction pathways such as two-component systems (TCSs) to reprogram appropriate genetic and physiological functions for adaptation and survival. The MprAB TCS is commonly found in actinobacteria and has been associated with important processes such as mycobacterial virulence, nutrient starvation, and environmental stress, particularly cell envelope stress. However, a comprehensive investigation of the function and response network of the MprAB TCS in corynebacteria remains to be carried out. In this study, w

BACKGROUND: Nontoxigenic Corynebacterium diphtheriae, often associated with wounds, can rarely cause infective endocarditis (IE). Five patients with C. diphtheriae IE were identified within 12 months at a Seattle-based hospital system. We reviewed prior C. diphtheriae-positive cultures to determine if detections had increased over time and evaluated epidemiologic trends.
METHODS: We conducted a formal electronic health record search to identify all patients aged ≥18 years with C. diphtheriae detected in a clinical specimen (ie, wound, blood, sputum) between 1 September 2020 and 1 April 2023. …

Patient 1 was a 70-year-old woman with refractory diffuse large B-cell lymphoma who received allogeneic peripheral blood stem cell transplantation from an HLA-haploidentical related donor. Upper back pain appeared on day63, and Th8-Th9 pyogenic spondylitis was diagnosed based on magnetic resonance imaging (MRI). Blood culture on day14 identified Corynebacterium striatum as the causative bacteria of blood stream infection (BSI). The pyogenic spondylitis resolved after treatment with daptomycin for 2 months. Patient 2 was a 65-year-old man with relapsed angioimmunoblastic T-cell lymphoma who rec…

The bacterium Clostridium botulinum, well-known for producing botulinum neurotoxins, which cause the severe paralytic illness known as botulism, produces C2 toxin, a binary AB-toxin with ADP-ribosyltranferase activity. C2 toxin possesses two separate protein components, an enzymatically active A-component C2I and the binding and translocation B-component C2II. After proteolytic activation of C2II to C2IIa, the heptameric structure binds C2I and is taken up via receptor-mediated endocytosis into the target cells. Due to acidification of endosomes, the C2IIa/C2I complex undergoes conformational

Circular economy holds great potential to minimize the use of finite resources, and reduce waste formation by the creation of closed-loop systems. This also pertains to the utilization of sidestreams in large-scale biotechnological processes. A flexible feedstock concept has been established for the industrially relevant Corynebacterium glutamicum, which naturally synthesizes the yellow C50 carotenoid decaprenoxanthin. In this study, we aimed to use a preprocessed aquaculture sidestream for production of carotenoids, including the fish feed ingredient astaxanthin by C. glutamicum. The addition…

Caseous lymphadenitis (CLA) is a bacterial infection caused by Corynebacterium pseudotuberculosis (C. pseudotuberculosis) that affects sheep and goats, leading to abscess formation in their lymph nodes. The present study aimed to isolate and identify C. pseudotuberculosis from CLA in smallholder sheep and goats, and determine the resistance patterns, virulence, and resistance genes of the isolates. Additionally, genotypic and phylogenetic analysis of the isolates was conducted using ERIC-PCR and DNA sequencing techniques. A cross-sectional study examined 220 animals (130 sheep and 90 goats) fr…
