Investigation of antimicrobial and antivirulence effects of lactobionic acid and carvacrol, individually and in combination, against Escherichia coli isolates.

BACKGROUND: Multidrug-resistant (MDR) Escherichia coli represents a major global health concern due to its strong biofilm-forming ability, which enhances antimicrobial tolerance and contributes to persistent infections. This study aimed to evaluate the individual and combined antibiofilm and antivirulence activities of lactobionic acid (LBA) and carvacrol (CAR) against clinical E. coli isolates. METHODS AND RESULTS: Fifty-one clinical isolates, including E. coli ATCC 25,922, were assessed for biofilm formation. Antimicrobial activity of LBA and CAR was determined through minimum inhibitory con

Polymeric blends enabling controlled delivery of Lippia graveolens bioactives for wound repair: in vitro and in vivo evaluation.

Chronic wounds are characterized by an increased susceptibility to microbial colonization, which results in prolonged inflammation and the development of microbial biofilms. The inadequate use of systemic antibiotics has led to the emergence of antibiotic-resistant microorganisms. Therefore, the use of essential oils has gained significant importance. Lippia graveolens Kunth essential oil (OR) presents antioxidant, anti-inflammatory, antimicrobial and antifungal properties, which could enhance the wound healing process. In this study, we prepared PCL microfiber mats (MFs) loaded with either OR

The Transient Receptor Potential Melastatin 7 (TRPM7) Inhibitors Suppress Seizure-Induced Neuron Death by Inhibiting Zinc Neurotoxicity.

Transient receptor potential melastatin 7 (TRPM7) is an ion channel that mediates monovalent cations out of cells, as well as the entry of divalent cations, such as zinc, magnesium, and calcium, into the cell. It has been reported that inhibitors of TRPM7 are neuroprotective in various neurological diseases. Previous studies in our lab suggested that seizure-induced neuronal death may be caused by the excessive release of vesicular zinc and the subsequent accumulation of zinc in the neurons. However, no studies have evaluated the effects of carvacrol and 2-aminoethoxydiphenyl borate (2-APB), b

Carvacrol from Moringa oleifera as a potential antidiabetic agent using integrated in-silico approach inhibiting TCF7L2.

Diabetes mellitus is a major health concern worldwide; lifestyle and rising urbanization are the key contributing factors. The genetic factors implicated in type 2 diabetes include the transcription factor 7-like 2 (TCF7L2) gene on chromosome 10q25.3, which has been greatly linked with diabetes, but the mechanisms and therapeutic effect on this gene are yet to be clearly defined. The objective of the research was to discover and screen natural phytochemicals of Moringa oleifera, especially carvacrol, as promising TCF7L2 inhibitors through combined in-silico methods. We have used a computationa

Artificial intelligence-assisted QSAR modeling of phenolic monoterpenes targeting Helicobacter pylori.

As a gram-negative bacterium implicated in various gastrointestinal problems, Helicobacter pylori is classified by the World Health Organization as a group 1 carcinogen. Considering multidrug resistance, particularly to clarithromycin and metronidazole, the efficacy of standard eradication regimens has been compromised causing the urgent need for novel therapeutic agents. Phenolic monoterpenes, volatile plant-derived metabolites such as thymol, carvacrol, vanillin and eugenol, exhibit promising antimicrobial activity against H. pylori however, with limitations due to poor solubility and rapid

Ion-Dependent ATPase Activity and Metabolic Gene Expression in TNF-α-Challenged Skeletal Muscle Cells: Mechanistic Characterisation of Carvacrol's Bioenergetic Effects.

Tumour necrosis factor-alpha (TNF-α) disrupts bioenergetic homeostasis in skeletal muscle cells through the suppression of ion-dependent ATPase activities, mitochondrial depolarisation, and impairment of antioxidant defences. Carvacrol, a phenolic monoterpenoid constituent of thyme and oregano essential oil, has been shown to exert cytoprotective effects in TNF-α-challenged L6 rat myoblasts. The mechanistic basis of these effects, specifically the relationship between membrane-associated ATPase function, mitochondrial polarisation status, and transcriptional regulation of metabolic stress-resp

β-CD-MOF-based edible antimicrobial packaging film with humidity-controlled carvacrol release for preserving fresh strawberry.

The development of safe, environmentally friendly, edible antimicrobial packaging films represents a promising alternative to conventional plastic packaging for reducing spoilage and extending the shelf life of fresh food. Here, we propose a novel strategy to construct edible β-CD-MOF/carvacrol@zein (BCCZ) composite films by intertwining β-CD-MOF loaded with the antimicrobial essential oil carvacrol, and zein. The resulting BCCZ films exhibit high humidity-triggered, long-lasting bactericidal efficacy, effective fruit preservation, and excellent biosafety. Characterization revealed that BCCZ f

Combined effect of conventional antimicrobials with essential oils and their main components against resistant Streptococcus suis strains.

Streptococcus suis is a major swine pathogen. Different strategies to reduce the antimicrobial resistance to conventional antimicrobials (AMBs) have been proposed, including the combined use with essential oils (EOs). The objective of this work was to evaluate the effectiveness of the combination of cinnamon, oregano, common thyme and red thyme EOs and their main components (cinnamaldehyde, carvacrol and thymol) with conventional AMBs (oxytetracycline, trimethoprim-sulfamethoxazole, gentamicin and penicillin) against field resistant S. suis strains. The checkerboard method was used to assess t

Carvacrol improves neurological function by inhibiting TRPM7-mediated BBB disruption and hemorrhage after TBI.

Traumatic brain injury (TBI) triggers a cascade of secondary pathophysiological events, including blood-brain barrier (BBB) disruption, intracerebral hemorrhage, neuroinflammation, neuronal death, and persistent neurological deficits. Although transient receptor potential melastatin 7 (TRPM7) is known to regulate cellular stress responses, its role in TBI-induced vascular dysfunction remains unclear. This study aimed to investigate the involvement of TRPM7 in BBB damage and hemorrhage after TBI and to evaluate the therapeutic potential of carvacrol, a TRPM7 inhibitor. TBI was induced in male m

Carvacrol improves cognitive dysfunction by decreasing amyloid-β accumulation and regulating neuroinflammation in ovariectomized renovascular hypertensive rats.

Hypertension contributes to both the development and progression of brain damage and cognitive dysfunction in the postmenopausal period in women. Carvacrol (CAR), which can easily cross the blood-brain barrier, exhibits neuroprotective properties due to its antioxidant, anti-inflammatory, and anti-apoptotic effects. In the present study, we have examined the effect of CAR treatment on learning-memory impairment in a post-menopausal hypertensive rat model that was induced by ovariectomy following two-kidney, one-clip renovascular hypertension surgery. From the third week after the establishment

5-FU and the dietary flavonoid carvacrol: a synergistic combination that induces apoptosis in MCF-7 breast cancer cells.

Along with the benefits of chemotherapy in the treatment of breast cancer, the side effects of these drugs along with drug resistance make their use complicated. One of the solutions to overcome this problem is the use of herbal products and combination therapy. In this research, we try to investigate the effects of carvacrol, a monoterpene flavonoid, in combination with the chemotherapy drug 5-FU. Combination index method was used for the drug-drug interactions analysis based on the Chou and Talalay method and the data from MTT assays. Apoptosis was assessed by the ELISA cell death method. P-
